RESUMO

The present study aimed at investigating how long-chain non-coding RNA (lncRNA) tumor suppressor candidate 7 (TUSC7) regulates the malignant biological behavior of osteosarcoma cells. Tumor tissues and adjacent tissues of 30 patients with osteosarcoma were collected, and the expression levels of lncRNA TUSC7 and miR-375 were detected by RT-qPCR. lncRNA TUSC7 mimic and miR-375 mimic transfection models were established in MG63 osteosarcoma cells, and Transwell assays were used to detect the migration ability of MG63 cells. An MTT assay was used to assess the proliferation ability of MG63 cells. lncRNA TUSC7 in osteosarcoma tissue was significantly lower than that of adjacent tissues, while miR-375 levels were significantly higher than that of adjacent tissues; the two levels have a negative correlation. lncRNA TUSC7 mimic inhibited MG63 proliferation and migration abilities. miR-375 mimic promoted MG63 proliferation and migration abilities. The lncRNA TUSC7 mimic and miR-375 mimic co-transfection system could partially rescue the inhibition of lncRNA TUSC7 mimic on MG63 cells. In conclusion, lncRNA TUSC7 inhibited the proliferation and migration of MG63 osteosarcoma cells by regulating miR-375.

OBJECTIVE: To study the clinical effects of electrical stimulation therapy on lumbar disc herniation-induced sciatica and its influence on peripheral reactive oxygen species (ROS) level. METHODS: 100 patients with lumbar disc herniation-induced sciatica were selected, and were randomly divided into the control and research group. The control group was treated with traction and other basic therapies, while the research group was treated with electrical stimulation. The pain degrees, peripheral ROS levels and clinical effects prior to treatment and at 4 weeks after treatment were examined. RESULTS: The total cure-remarkable-effectiveness rate of patients in research group was higher than that in control group (p<0.05). Before treatment, the pain rating index (PRI), present pain intensity (PPI) and visual analogue scale (VAS) score had no statistically significant differences between the two groups. After treatment, PRI, PPI and VAS scores in the two groups were lower than those prior to treatment; these indexes in research group were lower than those in control group, and the differences were statistically significant (p<0.05). After treatment, the peripheral ROS levels in the two groups were lower than those before treatment; it was lower in research group than that in control group (p<0.05). CONCLUSION: Electrical stimulation has a significant effect in the treatment of lumbar disc herniation-induced sciatica, which can effectively reduce the pain, alleviate the clinical symptoms and signs of patients, regulate the peripheral ROS level, and prevent the oxidative damage of myocardial tissues.

This study aimed to explore whether the genetic variant of COL11A1 is functionally associated with the development of lumbar disc herniation (LDH) in Chinese population. SNP rs1676486 of COL11A1 was genotyped in 647 patients and 532 healthy controls. The differences of genotype and allele distributions between LDH patients and healthy controls were evaluated using the χ² test. One-way ANOVA test was used to compare the relationship between genotypes and clinical features including tissue expression of COL11A1 and the degree of disc degeneration. Patients were found to have a significantly higher frequency of TT than the controls (10.2% versus 7.3%, P = 0.004). Besides, the frequency of allele T was found to be remarkably higher in the patients than the controls (34.8% versus 28.1%, P < 0.001) with an odds ratio of 1.36 (95% confidential interval=1.14-1.63). Patients with genotype TT were found to have remarkably more severe disc degeneration (P = 0.02). Besides, the expression of COL11A1 in the lumbar disc was significantly lower in the patients with genotype TT than in those with genotype CT or CC (P < 0.001). Moreover, the expression level was inversely correlated with the severity of disc degeneration (P < 0.001). We confirmed that the rs1676486 of COL11A may be functionally associated with LDH in the Chinese population. Extracellular matrix related proteins may play an important role in the pathogenesis of LDH. Our findings shed light on a better understanding of the pathogenesis of LDH, which could be a promising target for a novel treatment modality of LDH.
