RESUMO

This study aimed to elucidate the role and mechanisms of Complement C5a receptor 1 (C5AR1) in driving the malignant progression of anaplastic thyroid carcinoma (ATC). C5AR1 expression was assessed in ATC tissues and cell lines. Functional assays evaluated the effects of C5AR1 knockdown on the malignant features of ATC cells. The interaction between C5AR1 and miR-335-5p was confirmed using a luciferase reporter assay and Fluorescence in situ hybridization, and the impact of C5AR1 knockdown on the Toll-like receptor (TLR) 1/2 signaling pathway was examined. In vivo studies evaluated the effects of C5AR1 modulation on tumor growth and metastasis. C5AR1 levels were elevated in ATC tumor samples and associated with poor survival in ATC patients. C5AR1 knockdown impeded ATC cell proliferation, migration, and invasion in vitro. MiR-335-5p was identified as an upstream regulator of C5AR1, which negatively modulates C5AR1 expression. C5AR1 knockdown diminished TLR1, TLR2, and myeloid differentiation primary response 88 (MyD88) levels, while C5AR1 overexpression activated this pathway. Blocking TLR1/2 signaling abrogated the oncogenic effects of C5AR1 overexpression. C5AR1 silencing inhibited tumor growth and lung metastasis of ATC cells in nude mice. C5AR1 contributes to ATC tumorigenesis and metastasis by activating the TLR1/2 pathway, and is negatively regulated by miR-335-5p. Targeting the miR-335-5p/C5AR1/TLR1/2 axis represents a potential therapeutic strategy for ATC.

Objective: On the basis of electronic medical records, the data mining technology was adopted to explore the law of chronic kidney disease (CKD) and the intervention mode of mental health of patients. Methods: Based on the electronic medical records, the corresponding data extraction, database establishment, and data cleaning of CKD were performed. After that, the related data analysis, frequency analysis, cluster analysis, and nonparametric analysis were used to explore the laws of CKD diagnosis and treatment and nursing intervention mode of mental illness. The most common causes of CKD were chronic glomerulonephritis (43.76%), aristolochic acid nephritis (16.34%), diabetic nephritis (12.87%), and hypertensive nephritis (11.58%). The major treatment method for end-stage patients was alternative therapies, accounting for 46%. Compared with the depression score before intervention, that of the patients after the mindfulness therapy (50.99 ± 9.77 vs. 47.01 ± 9.33, P=0.024 < 0.5) and target behaviour nursing intervention (52.21 ± 8.12 vs. 48.01 ± 9.33, P=0.032 < 0.05) was obviously decreased. Conclusion: The data mining technology based on electronic records showed a good application prospect in the analysis of the diagnosis and treatment of CKD; and target behaviour nursing and mindfulness intervention were effective psychological intervention models.

Ecosystems and their components provide a lot of benefits for the welfare of human beings. Coupled with increasing socioeconomic development, most of the rapidly developing and transitional countries and regions have been experiencing dramatic land use changes. This has resulted in a large amount of forestland, grassland, and wetland being occupied as residential and industrial land or reclaimed for arable land, which in turn results in a sharp deterioration of ecosystem services around the world. Shandong Province, an economically powerful province of China, was chosen as a case study in order to capture the impact of socioeconomic development on ecosystem services. By way of the study, land uses and their changes were categorized between 1980 and 2006, and the ecosystem services capital and changes of 111 counties of Shandong Province in different phases were evaluated, as well as the total ecosystem services capital, followed by the zoning of ecosystem services function region of Shandong Province. We found that the counties in mountainous areas and wetlands, where generally the prefectural-level cities are located with a rapid socioeconomic development, experienced a successive deterioration of ecosystem services especially during the 2000s. Finally, three conservation strategies for managing and improving ecosystem services were proposed and discussed with the aim of achieving coordinate and sustainable development of the socioeconomy, environment, and ecosystems not only in Shandong Province but also in other provinces of China, as well as in other developing and transitional countries and regions.
