The leaves of Artocarpus tonkinensis are used in Vietnamese traditional medicine for treatment of arthritis, and the compound maesopsin 4-O-ß-D-glucoside (TAT-2), isolated from them, inhibits the proliferation of activated T cells. Our goal was to test the anti-proliferative activity of TAT-2 on the T-cell leukemia, Jurkat, and on the acute myeloid leukemia, OCI-AML. TAT-2 inhibited the growth of OCI-AML (and additional acute myeloid leukemia cells) but not Jurkat cells. Growth inhibition was shown to be due to inhibition of proliferation rather than increase in cell death. Analysis of cytokine release showed that TAT-2 stimulated the release of TGF-ß, yet TGF-ß neutralization did not reverse the maesopsin-dependent effect. Gene expression profiling determined that maesopsin modulated 19 identifiable genes. Transcription factor CP2 was the gene most significantly modulated. Real-time PCR validated that up-regulation of sulphiredoxin 1 homolog (SRXN1), hemeoxygenase 1 (HMOX1), and breast carcinoma amplified sequence 3 (BCAS3) were consistently modulated.

From Fissistigma pallens (Fin. & Gagn.) Merr. (Annonaceae), a Vietnamese folk medicinal plant, a novel eudesmane glycoside named fissispallin (1) has been isolated, besides afzelin. Their structures were elucidated by spectroscopic methods (1H, 13C and 2D NMR).

Chemical investigation of Stephania rotunda Lour. growing in Viet Nam led to the isolation and structural elucidation of three new alkaloids, 5-hydroxy-6,7-dimethoxy-3,4-dihydroisoquinolin-1(2H)-one (1), thaicanine 4-O-beta-D-glucoside (6), as well as (-)-thaicanine N-oxide (4-hydroxycorynoxidine) (8), along with 23 known alkaloids. These structures were determined on the basis of MS and NMR spectroscopic data.

A new flavone named miliufavol [8-(2-hydroxybenzyl)-5-hydroxy-2-(4-hydroxy-3-methoxyphenyl)-3,7-dimethoxy-4H-chromen-4-one or 8-C-(o)-hydroxybenzylpachypodol] from Miliusa balansae (Annonaceae) was isolated and structurally elucidated by spectroscopic means besides four known flavones: ombuine, chrysosplenol B, pachypodol and chrysosplenol C. These flavonoids exhibited interesting cytotoxic activity against three human cell lines (KB, Hep-G2, RD) with IC50 values < 5 microg/ml.

Activity-guided fractionation of the n-butanol extract from the leaves of Artocarpus tonkinensis led to the isolation of the auronol glycosides maesopsin 4-O-glucoside (1), as well as the new alphitonin-4-O-glucoside (2). These structures were identified on the basis of MS and NMR spectroscopic data. The lymphocyte stimulation test showed both compounds having immunosuppressive activity.

2,4-Dimethoxy-2-methyl-6H-pyran-3-one (1), a hitherto unknown natural product, and the calcium salt of rehmapicroside (2) have been isolated from rhizomes of the Vietnamese variety of Rehmannia glutinosa Libosch together with a series of known compounds: norcarotenoids (3-5), 2-formyl-5-hydroxy-methylfurane (6), the iridoid rehmaglutin D (7), iridoid glycosides (8-12) and phenylethyl alcohol glycosides (13-17). Their structures were determined by mass and NMR spectroscopy.

Phytochemical studies on the leaves of Fissistigma bracteolatum yielded besides the two known compounds 2-hydroxy-3,4,6-trimethoxychalcone (1) and 5,7,8-trimethoxyflav-3-ene (2), five new chalconoids 2-hydroxy-3,4,6-trimethoxychalcene (3), 2-hydroxy-3,4,6-trimethoxydihydrochalcone (4), 2'-hydroxy-3',4',6'-trimethoxydihydrochalcone (5), 2'-hydroxy-3',4',6'-trimethoxy-beta'-methoxychalcane (6) and 2'-hydroxy-3',4',6'-trimethoxy-beta'-ethoxychalcane (7). The structures of these compounds were determined by mass and NMR spectroscopic methods.

From roots of Homalomena aromatica three new sesquiterpene alcohols, 1 beta, 4 beta, 7 alpha-trihydroxyeudesmane, homalomenol A, and homalomenol B were isolated together with oplopanone, oplodiol and bullatantriol. The structures of the new constituents were elucidated by spectroscopic investigations and chemical transformations.

A new triterpene and its glycosides were isolated from the bark of Schefflera octophylla together with asiatic acid and asiaticoside. Based on spectroscopic data, especially 2DNMR, and chemical transformations the structures of the new compounds were determined as 3 alpha-hydroxy-urs-12-ene-23,28-dioic acid and 3 alpha-hydroxy-urs-12-ene-23,28-dioic acid 28-O-[alpha-L-rhamnopyranosyl (1----4)-O-beta-D-glucopyranosyl (1----6)]-beta-D-glucopyranoside. For the first time asiaticoside was isolated from a plant other than Centella asiatica.

In addition to 3-epi-betulinic acid, three triterpene glycosides were isolated from leaves of Schefflera octophylla. The structures of the glycosides have been determined as 28-O-[alpha-L-rhamnopyranosyl(1----4)-O-beta-D-glucopyranosyl(1----6)-be ta-D- glucopyranosides of 3 alpha-hydroxy-lup-20(29)-ene-23,28-dioic acid, 3 alpha,11 alpha- dihydroxy-lup20(29)-ene-23,28-dioic acid and 3-epi-betulinic acid by spectroscopic data and chemical transformations. The last two compounds were found for the first time in the plant kingdom.

Based on spectroscopic data and chemical transformations, the structures of the new constituents were determined as 3-epi-betulinic acid 3-O-beta-D-glucopyranoside 28-O-[alpha-L-rhamnopyranosyl(1----4)-O-beta-D-glucopyranosyl(1----6)]-b eta- D-glucopyranoside and alpha-L-rhamnopyranosyl(1----4)-O-beta-D-glucopyranosyl(1----6)-beta-D- glucopyranose.
