Dog-mediated rabies kills tens of thousands of people each year in India, representing one third of the estimated global rabies burden. Whilst the World Health Organization (WHO), World Organization for Animal Health (OIE) and the Food and Agriculture Organization of the United Nations (FAO) have set a target for global dog-mediated human rabies elimination by 2030, examples of large-scale dog vaccination programs demonstrating elimination remain limited in Africa and Asia. We describe the development of a data-driven rabies elimination program from 2013 to 2019 in Goa State, India, culminating in human rabies elimination and a 92% reduction in monthly canine rabies cases. Smartphone technology enabled systematic spatial direction of remote teams to vaccinate over 95,000 dogs at 70% vaccination coverage, and rabies education teams to reach 150,000 children annually. An estimated 2249 disability-adjusted life years (DALYs) were averted over the program period at 526 USD per DALY, making the intervention 'very cost-effective' by WHO definitions. This One Health program demonstrates that human rabies elimination is achievable at the state level in India.

Interleukin (IL)-10 has been implicated in persistence of pathogens in a number of chronic infections. Infected CD4+ cells upon reactivation with HIV antigens were also shown to produce IL-10, which might contribute to their persistence. Hence, it is crucial to determine mechanisms regulating IL-10 production after activation with HIV antigens for devising effective blocking strategies. In this study, ERK-, T-bet- and FoxP3-dependent pathways were evaluated for their possible roles in IL-10 production by infected CD4+ cells after reactivation with HIV Env. Intracellular and secreted IL-10 levels were determined by flow cytometry and Bioplex assay after treating PBMCs with PD98059, tipifarnib and cyclosporin A for blocking of ERK-, T-bet-and FoxP3-dependent pathways, respectively. Baseline levels of T-bet, pERK were higher in P24+ CD4+ cells as compared to uninfected CD4+ cells, which increased further after activation with Env. Inhibition of T-bet resulted in 2.3-fold reduction of IL-10 expression whereas ERK and FoxP3 inhibition failed to cause suppression of IL-10 expression. Conversely, IL-10 secreted by PBMCs was inhibited maximally after ERK inhibition suggesting its role in regulation of cytokine secretory pathway. IFN-γ was found to be suppressed after treatment with inhibitors of all these pathways. Thus, the study highlighted need for IL-10 blockade along with the use of antigens for therapeutic vaccinations or latency reversal and identified the T-bet-dependent pathway as an important pathway regulating IL-10 production by infected CD4+ cells. However, simultaneous blockade of IFN-γ precludes use of inhibitor of this pathway as an IL-10 blocking strategy.

The PM(2.5) and PM(10) samples were collected during Diwali celebration from study area and characterized for ionic concentration of four anions (NO(3) (-), NO(2) (-), Cl(-), SO(4) (2-)) and five cations (K(+), Mg(2+), NH(4) (+), Ca(2+), Na(+)). The results showed that the ionic concentrations were three times compared to those on pre and post Diwali days. Predominant ions for PM(2.5) were K(+) 33.7 µg/m(3), Mg(+) 31.6 µg/m(3), SO(4) (2-) 22.1 µg/m(3), NH(4) (+) 17.5 µg/m(3) and NO(3) (-) 18 µg/m(3) and for PM(10) the ionic concentrations were Mg(+) 29.6 µg/m(3), K(+) 26 µg/m(3), SO(4) (2-) 19.9 µg/m(3), NH(4) (+) 16.8 µg/m(3) and NO(3) (-) 16 µg/m(3). While concentration of SO(2) and NO(2) were 17.23, 70.33 µg/m(3) respectively.

Rural India derives its energy needs for cooking and heating through the use of fuel wood and for lighting and agricultural operations through kerosene and diesel. Use of fuel wood has aggravated the problem of de-forestation, while availability of kerosene and diesel cannot be guaranteed due to corrupt practices in the public distribution systems. In contrast, urban India derives its energy needs through LPG cylinders, petrol, and electricity. However, their cost and uncertainty rendered them beyond the reach of lower income population. This scenario is more or less true with many developing countries. To meet these objectives, biogas generation from biodegradable waste using anaerobic digestion (AD) appears to be a sustainable avenue as it could be used for (a) water and space heating of farmhouses, animal shelters, (b) generating steam for food processing plants, and (c) electricity generation, in addition to reducing the pollution/hazard potential of these wastes. Many of the underdeveloped and developing countries are in the temperate zone and thus mesophilic AD could provide a desired pathway to achieve a long delayed need of energy for comfortable living, farming, and industrial operations. Efforts made in this direction are reviewed in the present article.

RESUMO

After introducing thermophilic anaerobic digestion (AD), characteristics of thermophilic methanogens are provided. Accordingly, (a) site of occurrence, (b) morphological characteristics (shape and motility), (c) biochemical characteristics (Gram character and % G+C profile), (d) nutritional characteristics (NaCl requirement and substrate specificity), and (e) growth characteristics (pH and temperature) of thermophilic methanogens are described. Some studies of the thermophilic AD are cited with their operational management problems. Subsequently, strategies to maximize net energy production are given, including mode of heating the bioreactors, role of agitation to promote AD performance and mode/intensity of mixing. Finally, advantages as well as drawbacks of AD under thermophilic conditions are given, concluding with its applications.
