RESUMO

We examine how the occupation of farming structures the stress experiences of individuals through the timing and placement of actions. Further, we show how occupations have effects that spillover into family and friendship relationships. We find that farming affects both exposure and vulnerability to stressors. Specifically, farm men are more exposed to financial and job-related stressors, while less prone to marital conflict, than non-farmers. Given the importance of cohesion in farm family operations, farm men are more vulnerable to such conflict when it occurs. However, farm men are unaffected, if not consoled, by knowledge of undesirable events in the lives of their friends. We explore this finding and conclude that farm men use downward social comparisons to cope with the high levels of uncertainty characteristic of farming in the aftermath of the 1980s farm crisis.

Basic fibroblast growth factor (FGF-2) is a potent mitogen which is required for normal development, particularly the development of the skeletal system, where the inhibition of FGF binding to its receptor results in various skeletal malformations. The present study employed a newly engineered line of FGF-2 transgenic mice to determine the effects of overexpressing FGF-2 on limb bone ontogeny. We collected radiographic and weight data longitudinally and obtained the length, proximal, distal, and minimum diaphyseal widths of the humerus, femur, and tibia. Because growth is nonlinear with respect to time, we used the Gompertz mathematical model to obtain parameters describing rate and timing for each individual for each measurement. Differences in the parameters due to genotype and sex were subsequently tested with ANOVA. Transgenic animals exhibited consistently shorter limb bones which were generally wider at the epiphyses than those of controls. Parameters of early growth, including initial size and proportional rate of growth, appeared to be most directly responsible for significant differences in final size; however, exponential decay of growth was also a marginally significant factor. There were no differences between the genotypes in body weight, indicating that the shape anomalies observed in transgenic mice were a direct result of the action of FGF-2 rather than a general runting phenomenon.

RESUMO

The authors describe mammographic and pathologic evidence of the growth of a fibroadenoma in an 82-year-old obese woman, 44 years after menopause, who had never been treated with hormone therapy. The patient's obesity is hypothesized as the causal factor for increased estrogen levels leading to growth of the fibroadenoma.

RESUMO

Estimating microbial standing crops and microbial production in natural habitats has been difficult for microbial ecologists. The present paper describes a simple spectrophotometric assay based on the hydrolysis of fluorescein diacetate which estimates well the standing crops of microbial cells on coniferous needles and twigs. A technique is also presented for correlating optical density readings with actual dry weights of microbial cells epiphytic on needles, and thus for standardizing the assay. The assay shows promise of broad applicability to other microbial habitats.

Degradation of intrinsic hepatic [(14)C]haem was analysed as (14)CO formation in living rats and in hepatic microsomal fractions prepared from these animals 16h after pulse-labelling with 5-amino[5-(14)C]laevulinic acid, a precursor that labels bridge carbons of haem in non-erythroid tissues. NADPH-catalysed peroxidation of microsomal lipids in vitro (measured as malondialdehyde) was accompanied by loss of cytochrome P-450 and microsome-associated [(14)C]haem (largely cytochrome P-450 haem), but little (14)CO formation. No additional (14)CO was formed when carbon tetrachloride and 2-allyl-2-isopropylacetamide were added to stimulate lipid peroxidation and increase loss of cytochrome P-450 [(14)C]haem. Because the latter effect persisted despite inhibition of lipid peroxidation with MnCl(2) or phenyl-t-butylnitrone(a spin-trapping agent for free radicals), it was concluded that carbon tetrachloride, as reported for 2-allyl-2-isopropylacetamide, may promote loss of cytochrome P-450 haem through a non-CO-forming mechanism independent of lipid peroxidation. By comparison with breakdown of intrinsic haem, catabolism of [(14)C]methaemalbumin by microsomal haem oxygenase in vitro produced equimolar quantities of (14)CO and bilirubin, although these catabolites reflected only 18% of the degraded [(14)C]haem. This value was increased to 100% by addition of MnCl(2), which suggests that lipid peroxidation may be involved in degradation of exogenous haem to products other than CO. Phenyl-t-butylnitrone completely blocked haem oxygenase activity, which suggests that hydroxy free radicals may represent a species of active oxygen used by this enzyme system. After administration of carbon tetrachloride or 2-allyl-2-isopropylacetamide to labelled rats, hepatic [(14)C]haem was decreased and haem oxygenase activity was unchanged; however, (14)CO excretion was either unchanged (carbon tetrachloride) or decreased (2-allyl-2-isopropylacetamide). These changes were unaffected by cycloheximide pretreatment. From the lack of parallel losses of cytochrome P-450 [(14)C]haem and (14)CO excretion, one may infer that an important fraction of hepatic [(14)C]haem in normal rats is degraded by endogenous pathways not involving CO. We conclude that carbon tetrachloride and 2-allyl-2-isopropylacetamide accelerate catabolism of cytochrome P-450 haem through mechanisms that do not yield CO as an end product, and that are insensitive to cycloheximide and independent of haem oxygenase activity.

RESUMO

A patient with multiple gastroduodenal ulcers and gastric hypersecretion due to histamine excess associated with basophilic leukemia is described. Treatment with the histamine H-2-receptor blocker, cimetidine, was able to abolish the gastric hypersecretion and gastrointestinal blood loss. A literature review produced four other cases of multiple gastroduodenal ulceration with this disease, suggesting that it is more common than previously recognized. The availability of a potent H-2-receptor antagonist offers effective and specifically tailored therapy to counteract the gastrointestinal effects of hyperhistaminemia in basophilic leukemia and other "histamine excess" diseases.

RESUMO

The weight-average molecular weight of the Mo-Fe protein isolated from Azotobacter vinelandii has been determined by sedimentation-equilibrium techniques. In buffer, the value is 245000+/-5000; in 8M-urea, the value is 61000+/-1000. The protein was separated into two components by chromatography on CM-cellulose in 7M-urea, pH 4.5. These components have similar molecular weights but were shown to differ in charge, amino acid content and arginine-containing peptides. It is proposed that the tetramer has the subunit composition (nalpha2nbeta2).

The extent of biodegradation of o-benzyl-p-chlorophenol, marketed as a germicide under the name Santophen((R)) 1 (Monsanto Co.), in river water, sewage, and activated sludge was determined. Biodegradation was assessed by use of a colorimetric procedure for phenolic materials, carbon analysis, and CO(2) evolution. In unacclimated river water, 0.1 mg of Santophen 1 per liter was degraded within 6 days. In sewage, 0.5 and 1.0 mg/liter levels of Santophen 1 were degraded in 1 day. Acclimated activated sludge achieved 80% biodegradation of 1.0 mg/liter Santophen 1 in 8 h and 100% in 24 h. When effluent from a semicontinuous activated sludge unit, acclimated to 20 mg of Santophen 1 per liter was used as the inoculum for the CO(2) evolution procedure, 60% of the total theoretical CO(2) was evolved from Santophen 1. Based on the results of these studies, indicating Santophen 1 to be readily biodegraded in at least four biological systems, the continued use of present levels of Santophen 1 should present no significant environmental problems.
