[Immunologic correction using thymus gland preparation (T-activin) in the programmed treatment of patients with non-lymphoid leukemia]. / Immunokorrektsiia preparatom timusa (T-aktivinom) v programmnom lechenii detei, bol'nykh s ostrym nelimfoidnym leikozom.

Data have been presented on the observation of 80 children with acute myeloid leukemia, among them 26 patients who, in addition to chemotherapy, were given tactivin treatment. The authors have proved the necessity of long-term tactivin administration in acute myeloid leukemia (no less than 2-3 years). that permits not only reducing the incidence of intercurrent diseases, but also increasing the duration of the remission period.

[Changes in T-lymphocyte subpopulations in uveitis]. / Izmeneniia subpopuliatsii T-limfotsitov pri uveitakh.

Examinations of 30 patients with uveites, carried out by the method developed by Moretta et al., have revealed changes in T-lymphocyte subpopulations, mainly of the immunodeficiency type, with the reduction of the total T-lymphocyte count in 53.3% of patients and a significant decrease of the T mu/T gamma coefficient in 60% of patients. Stage II immunodeficiency status has been diagnosed in the majority of patients with generalized and anterior uveites. Stage II T-helper deficit has been most frequently detected in uveites coursing for 1-3 years with an exudative hemorrhagic component. The detection of the types and variant of immunoregulatory lymphocyte disorders helps choose the treatment strategy, including immunocorrecting drugs.

[T-lymphocyte subpopulations in autoimmune and subacute thyroiditis treated with T-activin]. / Subpopuliatsii T-limfotsitov pri autoimmunnom i podostrom tireo- iditakh v protsesse lecheniia T-aktivinom.

The results of examination of 15 patients with autoimmune and subacute thyroiditis are analyzed. T-cellular immunity parameters have been examined, i.e. total T-lymphocytes, T-suppressors, T-helpers, killers. The study has revealed a reduction of the total T-lymphocyte count and a shifted ratio of the T-lymphocyte subpopulations, these changes being more manifest in autoimmune thyroiditis. T-activin++ therapy resulted in a manifest tendency to normalization of the counts of T-lymphocytes and their subpopulations.

Monoclonal antibodies OKT 3, OKT 4 and OKT 8 were used in 20 patients with Hodgkin's disease in order to characterize lymphocyte subpopulations in the peripheral blood, lymph nodes and spleen. As compared to the normal donors' peripheral blood and other parameters, the content of the OKT 3+, OKT 4+ cells in the involved lymph nodes and in the splenocyte suspension turned out to be high.

Adherent cells were released from human peripheral blood mononuclears and the remaining mononuclears were separated by the density gradient centrifugation on percoll. Five fractions obtained were tested for cytotoxicity against K-562 and HeLa tumour target cells. Cell morphology and reactivity to some monoclonal antibodies were also studied. Fractions 2 and 3 were enriched by large granular lymphocytes (LGL) and BMA-070 positive cells and showed increased cytotoxicity only to K-562 target cells, while fraction 1 contained 9.5% of monocytes, but no LGL, and showed increased cytotoxicity against both K-562 and HeLa target cells.

The patients with Hodgkins disease possessed an increased number of the T gamma-cells, a decreased number of T mu cells and higher endogenic suppressor activity as compared with healthy donors. Subpopulation of ferritin-carrying lymphocytes was found only in patients, whereas in donors these cells were not revealed at all. It was shown that ferritin receptors expressed only on T gamma-cells. For determination of the functional role of T-cell subpopulation before and after Con-A stimulation were studied. The suppression activity correlated with the T gamma-cell levels.

A simplified micromodification of the leukocyte adherence inhibition (LAI) test using both leukocytes and lymphocytes from peripheral blood of cancer patients is discussed. Data on oncoimmunologic specificity and cellular mechanisms underlying the proposed procedure as well as the results of a comparative study of LAI factor-producing T-cell and immunoregulatory (Tmu and T gamma) lymphocyte subpopulations in breast cancer patients are presented. The LAI-test micromodification may be adjunct in breast cancer diagnosis. It may be employed for monitoring T-cell specific immunity in cancer patients and further study of LAI factor-producing T-cells as a component of the structure of T-cell subpopulations.

[Isolation of mononuclear cells from the bone marrow of patients with hemoblastoses using one-step ficoll-verographin density gradient separation]. / Vydelenie mononuklearnykh kletok iz kostnogo mozga bol'nykh gemoblastozami putem razdelenia v odnostupenchatom gradiente plotnosti fikoll-verografin.

The one-step ficoll-verographin density-gradient centrifugation was first used for isolating mononuclear cells from human bone marrow. Children suffering from various hemoblastoses were donors. The morphological control of the cells isolated from the interface layer and May-Grünwald stained showed a high efficiency of the procedure proposed. Due to the nosological form of malignancy mononuclears composed from 100% (in the relapse of acute lymphocytic leukemia) to 81% (in hemoblastosis with "intact" bone marrow: lymphogranulomatosis and lymphosarcoma without leukemization) among the isolated cells. Thus, the one-step ficoll-verographin density-gradient centrifugation may be used for extensive clinical research, permitting more time-consuming procedures to be avoided.

[Dynamics of immunoregulator T-lymphocytes and its importance in the chemo-immunotherapy of lymphoproliferative diseases in children]. / Dinakmika immunoreguliatornykh T-limfotsitov i ee znachenie pri khimioimmunoterapii limfoproliferativnykh zabolevanii u detei.

The levels of T-lymphocytes in untreated lymphoid malignancies in children and those after different schemes of chemoimmunotherapy were compared. The ratio of T-helper and T-suppressor cells changed in the course of treatment in correlation with the chemotherapeutic effect: the fraction of T mu cells practically did not change whereas the T gamma cell fraction decreased considerably in patients who revealed a good response to therapy. BCG treatment stimulated both populations but the response of T gamma cells was stronger. Transplantation of thymus was followed by an increase on T mu cells.
