RESUMO

Correlations between genetic polymorphisms of three matrix metalloproteinase (MMP) genes and susceptibility to chronic obstructive pulmonary disease (COPD) were investigated. Relevant case-control studies were selected using rigorous inclusion and exclusion criteria. The comprehensive Meta-analysis 2.0 software was used to conduct the statistical analysis. An odds ratio with 95% confidence intervals was applied to assess the correlation between genetic polymorphisms of MMPs and susceptibility to COPD. Twelve high-quality studies were selected for inclusion in this meta-analysis. These studies included a combined total of 1533 COPD patients and 1530 healthy controls. The result of the meta-analysis showed that MMP-9 rs3918242 C > T was significantly correlated with increased susceptibility to COPD. However, MMP-1 rs1799750 1G > 2G and MMP-3 rs3025058 5A > 6A were not associated with COPD risk (all P > 0.05). Based on our meta-analysis, MMP-9 rs3918242 C > T is correlated with susceptibility to COPD, but MMP-1 rs1799750 1G > 2G and MMP-3 rs3025058 5A > 6A are not. These results should be further confirmed using a larger sample size.

By using CFU-GM/CFU-L colony assay, NBT/MTT reductant test and DNA fragmentation analysis, we studied the effects of Sophora flavescens (SF) on CFU-GM proliferative ratio in human normal bone marrow/umbilical cord blood and on proliferation, differentiation and apoptosis in human acute myelogenous leukemia HL-60 cells. The results showed that 5, 10, 15, 20 micrograms.microliter-1 of SF significantly inhibited proliferation and induced apoptosis in the HL-60 cells in a dose-dependent fashion. Fifteen micrograms.microliter-1 of SF also induced differentiation in HL-60 cells. Furthermore, cytotoxic activity of SF(5-15 micrograms.microliter-1) was not apparent on human normal hematopoietic progenitors(CFU-GM). The results indicate that an appropriate concentration of SF has a selective antileukemic effect. Thus, these are important impetuses for further research of SF as an anticancer agent.

The widespread and depth-in investigations on the pulmonary change in DM have been documented abroad. Some revealed that the abnormalities of pulmonary function usually exist in DM. There has been no similar report found in China so far. In this study, ventilation, small airway function (V25 and V50), diffusion capacity and artery blood gas studies have been done in 60 DM and 62 healthy subjects respectively. The results have shown that there is a significant difference between NIDDM group and control group (P less than 0.05) in V50, V25, DLCO and the reduced value of PaO2, and there exists remarkably significant difference between the IDDM group and control group (P less than 0.01) in the decreased values of TLC, FEV1.0%, V50, V25, DLCO, PaCO2 and PaO2.
