RESUMO

Understanding dose-responses is crucial for determining the utility of biomarkers in ecotoxicity assessment. Nitrofurazone is a broad-spectrum antibiotic that is widely used in the aquaculture industry in China despite its detrimental effects on ecosystems. Potential dose-response models were examined for the effect of nitrofurazone on two antioxidant enzymes, superoxide dismutase (SOD) and glutathione peroxidase (GPx), in the ciliated protozoan Euplotes vannus. This was achieved by measuring enzyme activity and gene expression profiling of SOD and GPx in ciliate cells exposed to nitrofurazone at doses ranging from 0 to 180mgl-1 for 6h, 12h, 18h and 24h. Dose-response dynamics were characterized by mathematical models. Results showed that: 1) dose-response patterns differed significantly among the tested endpoints, nitrofurazone concentrations and durations of exposure; 2) GPx activity was the best candidate biomarker because of its linear dose-response relationship; 3) SOD activity and mRNA relative expression levels of GPx and SOD are also candidate biomarkers but their dose-responses were non-linear and therefore more difficult to interpret; 4) partitioning the dose-response dynamic model by piecewise function can help to clarify the relationships between biological endpoints. This study demonstrates the utility of dynamic model analysis and the potential of antioxidant enzymes, in particular GPx activity, as a candidate biomarkers for environmental monitoring and risk assessment of nitrofurazone in the aquaculture industry.

Although salinity fluctuation is a prominent characteristic of many coastal ecosystems, its effects on biological adaptation have not yet been fully recognized. To test the salinity fluctuations on biological adaptation, population growth dynamics and Na+ /K+ -ATPase activity were investigated in the euryhaline bacterium Idiomarina sp. DYB, which was acclimated at different salinity exposure levels, exposure times, and shifts in direction of salinity. Results showed: (1) bacterial population growth dynamics and Na+ /K+ -ATPase activity changed significantly in response to salinity fluctuation; (2) patterns of variation in bacterial growth dynamics were related to exposure times, levels of salinity, and shifts in direction of salinity change; (3) significant tradeoffs were detected between growth rate (r) and carrying capacity (K) on the one hand, and Na+ /K+ -ATPase activity on the other; and (4) beneficial acclimation was confirmed in Idiomarina sp. DYB. In brief, this study demonstrated that salinity fluctuation can change the population growth dynamics, Na+ /K+ -ATPase activity, and tradeoffs between r, K, and Na+ /K+ -ATPase activity, thus facilitating bacterial adaption in a changing environment. These findings provide constructive information for determining biological response patterns to environmental change.

This paper presents investigations of segregation distortion of six rice F2 populations generated from reciprocal F1 hybrids grown at three locations varied at altitudes from 400 to 2200 m. The F1s were derived from reciprocal crosses between cv. XMG, which is a japonica landrace traditionally grown at 2650 m altitude, and cv. N34, which is a japonica restorer possessing a fertility restoring (Rf) gene and cytoplasm of male sterility (CMS) donated by an indica cultivar. Among nine morphological traits of the F2 populations, only one was in normal distribution, eight were distorted in all or at least one population. Out of 16 polymorphic PCR markers, 10 markers distributed on 7 chromosomes were significantly distorted. Among these markers, RMAN7 and RM257 were distorted in both of the reciprocal populations, which suggested that nuclear genes had strong effects on segregation distortion. The other makers were distorted only in the populations with cytoplasm donated by XMG or N34. The results indicated that segregation of DNA markers was affected by cytoplasm background. Segregation distribution was also affected by altitude, since segregation distortions of most of the markers were detected not in all the three populations generated from F1 grown at the three altitudes, but only in one population from F1 grown at one altitude. Marker M45461, which is located within Rf-1 locus, was severely distorted towards N34 in all the populations with cytoplasm donated by N34, but not in the populations with cytoplasm provided by XMG. The results indicated that interaction between CMS and Rf gene had strong effects on distortion. Results of this study indicated that japonica cytoplasm did not cause distortion favouring a special parent, but indica cytoplasm made distortion favouring a maternal parent. The results suggested that indica cytoplasm was not well compatible with japonica nuclear background, while japonica cytoplasm did not have such trouble with indica nuclei. This study also found that the six F2 populations were divergent into two groups due to difference of cytoplasm background.
