RESUMO

Light intensity is a major environmental factor affecting the growth and survival of trees in a forest. The effect of light reduction on photosynthesis and photorespiration of an evergreen broad-leaved tree, Phoebe bournei (Hemsley) Yang was examined with three levels of full light, 50.5% light, and 21.8% light. The results showed that shading led to significant increase in plant height and crown diameter. Light-saturated leaf photosynthetic rate (Amax), maximal carboxylation activity (Vcmax), maximum electron transfer rate (Jmax), stomatal conductance (gs), mesophyll conductance (gm) and chloroplast CO2 concentration (Cc) significantly increased in response to shade. Photorespiratory CO2 release rate (PR) was higher in plants grown under shade conditions than under full light. The relative limitations of gm (lm) was higher than the relative limitations of gs (ls) and the relative limitations of biochemical factors (lb) in leaves of P. bournei grown under full light, whereas lm was lower than ls and lb under shade. Our results suggest that increase of photosynthesis in P. bournei leaves grown under shade is associated with enhanced CO2 diffusion and biochemistry. And we propose that enhancement of the photorespiratory is essential for shade leaves to improve photosynthesis.

DNA and RNA contain diverse chemical modifications that exert important influences in a variety of cellular processes. In addition to enzyme-mediated modifications of DNA and RNA, previous in vitro studies showed that pre-modified nucleoside triphosphates (NTPs) can be incorporated into DNA and RNA during replication and transcription. Herein, we established a chemical labeling method in combination with liquid chromatography-electrospray ionization-mass spectrometry (LC-ESI-MS) analysis for the determination of endogenous NTPs in the mammalian cells and tissues. We synthesized 8-(diazomethyl)quinoline (8-DMQ) that could efficiently react with the phosphate group under mild condition to label NTPs. The developed method allowed sensitive detection of NTPs, with the detection limits improved by 56-137 folds. The results showed that 12 types of endogenous modified NTPs were distinctly determined in the mammalian cells and tissues. In addition, the majority of these modified NTPs exhibited significantly decreased contents in human hepatocellular carcinoma (HCC) tissues compared to tumor-adjacent normal tissues. Taken together, our study revealed the widespread existence of various modified NTPs in eukaryotes.

Analytical methods based on multiplug filtration cleanup coupled with pulse glow discharge-ion mobility spectrometry and liquid chromatography tandem mass spectrometry were developed for the analysis of tricaine mesylate residue in fish and fish-raising water samples. A silica fiber holder and an appropriate new interface were designed to make the direct introduction of the fiber into the pulse glow discharge-ion mobility spectrometry introduction mechanism. The multiplug filtration cleanup method with adsorption mixtures was optimized for the determination of tricaine mesylate in fish samples. Good linear relationships were obtained by the two methods. For fish samples, limits of detection were 6 and 0.6 µg/kg by ion mobility spectrometry and liquid chromatography with tandem mass spectrometry, respectively. The matrix effect of the established liquid chromatography tandem mass spectrometry method was negligible for fish samples but that of the ion mobility spectrometry method was not. The two methods were compared. The ion mobility spectrometry system could be used a rapid screening tool on site with the advantage of rapidity, simplicity, and portability, and the liquid chromatography tandem mass spectrometry system could be used for validation in laboratory conditions with the advantage of lower limit of detection, stability, and precision.
