RESUMO

The addition of InBr3 to the oxidative Sonogashira cross-coupling reaction of 2-ethynylaniline with (E)-trimethyl(3,3,3-trifluoroprop-1-enyl)silane led to a dramatic increase in the reactivity to afford the corresponding 1,3-enynes bearing a trifluoromethyl group on their terminal sp(2) carbon. The subsequent cyclization of these 1,3-enynes under palladium catalysis provides access to the corresponding indoles bearing a 3,3,3-trifluoroprop-1-enyl group at their 2-position.

RESUMO

The presence of N-methyl-L-aspartate (NMLA) was demonstrated in bivalves, Corbicula sandai and Tapes japonica. To our knowledge, this is the first report on the occurrence of NMLA in animal tissues. NMLA in bivalve tissues was identified according to the following findings; (a) its derivatives with (+)- and (-)- 1-(9-fluorenyl)ethyl chloroformate (FLEC) behaved identically with those of authentic NMLA, respectively, on high-performance liquid chromatography (HPLC), (b) its derivatives with (+)- and (-)- FLEC behaved identically with (-)- and (+)-FLEC derivatives of authentic N-methyl-D-aspartate (NMDA), respectively, on HPLC and (c) its behavior on thin-layer chromatography was the same as those of authentic NMLA. We also describe the distribution of NMDA, and D- and L-aspartate, to which N-methylaspartate enantiomers are structurally related. NMDA was more widely dirtributed than NMLA in bivalves. These bivalves containing NMLA showed lower D-aspartate contents and D/(D+L) ratios of aspartate, than the bivalves containing NMDA.

The role of glucocorticoids in the increase by cold-exposure of the effect of alanine on the malate-aspartate shuttle was studied in perfused rat liver. The capacity of the shuttle was evaluated by measurement of changes in both the rate of glucose production from sorbitol and the ratio of lactate to pyruvate during ethanol oxidation (Biomed. Res. 6, Suppl., 1986). The effect of alanine on the shuttle capacity was decreased by adrenalectomy. When 1.5 mg/kg dexamethasone sulfate was administrated to adrenalectomized rats kept at 24 or 4 degrees C, once daily for 5 days, the effect of alanine on the shuttle increased its capacity to the level of sham-operated rats that had been exposed to 4 degrees C for 5 days. The effects of dexamethasone were blocked by the coadministration of tetracycline with the agent. Cold exposure and steroid replacement had little effect on the alanine-induced elevation of the levels of aspartate, glutamate, and alpha-ketoglutarate in liver cells. The increase of the effect of alanine could not be explained only by changes in the activity of NAD+ malate dehydrogenase and aspartate aminotransferase. The results suggest that cold exposure and replacement treatment with glucocorticoids modulate equally the effect of alanine on the capacity of the malate-aspartate shuttle.
