RESUMO

Functional (synthetic) activity of acridine orange-stained blood nuclear cells was studied by microspectral fluorescent analysis in experimental myocardial infarction. A specific time course of changes in the synthetic activity of blood nuclears, described by alpha parameter, was detected in uneventful myocardial infarction and infarction complicated by suppurative inflammation of the suture. This time course reflected the reparative changes in the myocardium. Hence, alpha parameter can be regarded as an indicator of reparative processes in the myocardium, and the above method recommended for fluorescent cytodiagnosis of reparation processes after acute myocardial infarction and for predicting its course.

We report here on a retinoid model of the phenomenon discovered in 1980, in which ceroid-lipofuscin cytosomes (CLC) increased their intrinsic fluorescence when exposed to fluorescence-exciting (lambda(ex)=365nm) ultraviolet (UV) light. We modeled this effect in vitro, irradiating a methanolic solution of retinyl palmitate (RP) either synthetic or extracted from the aged rat liver retinoid. Following our model, the mechanism of this phenomenon can be explained by the photodecomposition of RP during fluorescence. Palmitic acid (PA) that quenched fluorescence was separated by photohydrolysis and some products of RP photodegradation shifted absorption spectra towards the fluorescence-exciting UV band compared with the absorption spectrum of the initial RP. The total intensity of intrinsic fluorescence at 490nm of RP photodegradation products increased several times, because relative absorption at 365 nm wavelength was higher. We report here also the established chemical formulae of two retinoids, derivatives of RP photohydrolysis tentatively called R(368) and R(346). R(368) was determined as anhydroretinol, a naturally occurring intracellular messenger in the transduction pathway of retinoids regulating growth inhibition in lymphoid line cells. Retinoid R(346) was determined to be a methanolic intermediate of RP photodegradation (4,5-dihydro-5-methoxy-Anhydroretinol).

Electron microscopic investigation of lipofuscin granules (LG) accumulation during one month in the myocardium of young rabbits in case of experimental acute infarction was performed. Accumulation of LG in cardiocytes of the ischemia-damaged myocardium area at the prenecrotic and necrotic stages was found. Analogous LG accumulation in cardiocytes in the perinecrotic area at the stage of infarction development was also observed. The fact of LG formation in fibroblast-like cells was also established. Cardiocytes and fibroblast-like cells contain a set of ultrastructurally varying LG. LG formation in functionally different cells of the ischemia-damaged myocardium is a further evidence of the active metabolic role of this organelle suggesting a common principle in LG genesis.

The increase of the own luminescence lipofuscin granules (LG) in the course of single and repeated ultraviolet (UV) excitations (365 nm) in hybridoma (retrovirus transformed) cells cultured with or without 5 x 10(-4) M centrophenoxine (CP) was studied by microspectrofluorometric method. It was shown that CP influences only the rate of photochemical changes of chromophores in LG. Kinetic patterns of the own luminescence intensity of LG at the wavelength of 540 nm during excitation by UV permit one to suggest the occurrence of the cyclic chromophore changes.

RESUMO

Lipofuscin granules (LG) are found in the cultured hybridoma cells producing monoclonal antibodies to phage. LG have been studied using light and electron microscopy. Luminescent spectra of LG clusters in hybridoma cells are presented. The increase of own luminescence intensity of LG in the course of excitation by ultraviolet (365/nm) is shown. The advantages of hybridoma cells culture for investigation of LG on the cell level are discussed.

RESUMO

The ultrastructure of lipofuscin granules (LG) in hybridoma cells is described. Based on the electron microscopic evidence and literature data the possibility of generation of LG from cell endoplasmic reticulum (ER) is discussed. It is proposed that the production of LG is connected with disturbances in glycosylation of proteins and lipids on the ER membranes.

RESUMO

The paper describes the procedure of lipofuscin isolation from bovine myocardium with the use of 15-45% (weight/volume) linear gradient of sucrose density and differential centrifugation. Two coloured populations of lipofuscin granules with different density have been distinguished. Electron microscopic studies of the isolated lipofuscin have revealed the homogeneity of fractions and good maintenance of the granules. The electron microscopic, absorptive and luminescent spectral properties of lipofuscin preparations correspond to its properties in situ. The isolation of lipofuscin preparations suggests the possibility of studying the chemical composition and intramolecular structure of the granules.

RESUMO

The luminescent characteristics of lipofuscin granules in the human brain, heart, adrenal glands, and liver have been investigated. Lipofuscin luminescent spectra in all the tissues studied were shown to be of the same type and to contain two emission bands with maxima in the yellow (550--560 nm) and red (680--690 nm) regions of the spectrum. The relative variability in the intensity of these bands determines the changes in colour from yellow to red-brown in lipofuscin luminescence observed visually. The two main emission bands in luminescent spectra indicate the presence of two main luminescent chemical compounds in lipofuscin.

RESUMO

The process of repairing the myocardium ischemized areas was studied in young rabbits. On the 14th day after operation lipofuscin granules were found to accumulate in large quantities in the myocardial infarction area. Their luminescent spectral characteristics were identical to those of lipofuscin granules of old people. Accumulation of lipofuscin in ischemized areas of the young animal heart indicates an active metabolic role played by lipofuscin in post-infarction normalization of myocardium tissue functioning.
