RESUMO

Plant disease management has not significantly changed in the past 50 years, even as great strides have been made in the understanding of fungal biology and the etiology of plant disease. Issues of climate change, supply chain failures, war, political instability, and exotic invasives have created even more serious implications for world food and fiber security, and the stability of managed ecosystems, underscoring the urgency for reducing plant disease-related losses. Fungicides serve as the primary example of successful, widespread technology transfer, playing a central role in crop protection, reducing losses to both yield and postharvest spoilage. The crop protection industry has continued to improve upon previous fungicide chemistries, replacing active ingredients lost to resistance and newly understood environmental and human health risks, under an increasingly stricter regulatory environment. Despite decades of advances, plant disease management continues to be a constant challenge that will require an integrated approach, and fungicides will continue to be an essential part of this effort.

RESUMO

Several studies have demonstrated the advantages of using interactive voice response (IVR) technology to collect self-report data from research participants and recipients of psychological/medical services. IVR allows participants to phone a computer and respond to recorded questions by pressing the appropriate touch-tone keys on their telephone. Because this technology offers substantial benefits in terms of cost and efficiency, it is surprising that it has not been more widely utilized by researchers and practitioners. Along with the automation of the administration and scoring of tests or surveys, IVR provides for questioning to be adapted to the participants' responses. One possible explanation for the failure to exploit this technology is the absence of easy-to-use software that does not require programming knowledge. This article describes IVR Test & Survey, a program to facilitate the administration, scoring, and analysis of information collected with the use of IVR technology.

Seventeen leguminous species common to the Pacific Northwest were evaluated as potential hosts of the pea cyst nematode, Heterodera goettingiana, in both greenhouse and field experiments. In all experiments, juveniles of H. goettingiana penetrated roots of these 17 species with the exception of greenhouse-grown chickpea. Nematodes molted and developed into swollen third-stage or fourth-stage juveniles in many of the plants, but cyst development occurred only in the field on green pea, edible dry pea, and faba bean. More H. goettingiana cysts developed on fava bean than on green pea or edible dry pea. In H. goettingiana-infested soils, cropping sequences that include fava bean and pea should be avoided. However, certain legumes, such as winter vetch, may have the potential of serving as trap crops for H. goettingiana in this region.

We tested the hypothesis that isolates of Hirsutella rhossiliensis from host nematodes in the family Hoplolaimidae (Rotylenchus robustus and Hoplolaimus galeatus) would be more virulent to R. robustus than would isolates from host nematodes not in the Hoplolaimidae (Heterodera schachtii and Criconemella xenoplax). Nematodes were touched to 10-20 spores of different isolates and incubated at 20 C in 4.5 mM KC1; the percentage of nematodes colonized (filled with hyphae) was determined after 2, 5, 10, 20, and 30 days. The hypothesis was rejected because isolates from H. schachtii and C. xenoplax were equivalent or better at parasitizing R. robustus than were isolates from R. robustus and H. galeatus. In addition, the R. robustus and H. galeatus isolates were as pathogenic to C. curvata as they were to R. robustus, but produced fewer spores per colonized nematode (H. schachtii) than did the other isolates.

Numbers of cyst and root-knot nematodes and percentage parasitism by the nematophagous fungus Hirsutella rhossiliensis were quantified in microplots over 2 years. The microplots contained either sugarbeets in loam infested with Heterodera schachtii or tomatoes in sand infested with Meloidogyne javanica. The fungus was added to half of the microplots for each crop. Although H. rhossiliensis established in both microplot soils, the percentage of nematodes parasitized did not increase with nematode density and nematode numbers were not affected by the fungus. The results indicate that long-term interactions between populations of the fungus and cyst or root-knot nematodes will not result in biological control.

Thirty-two weed species common in South Carolina and one cultivar of tobacco were evaluated as hosts of Meloidogyne arenaria race 2 and M. incognita race 3 in the greenhouse. Egg mass production and galling differed (P < 0.05) among weed species. Chenopodium album, Euphorbia maculata, and Vicia villosa were good hosts of M. arenaria. Amaranthus palmeri, Rumex crispus, Amaranthus hybridus, Ambrosia artemisiifolia, lpomoea hederacea var. integriuscula, Setaria lutescens, Sida spinosa, Portulaca oleracea, and Rumex acetosella were moderate hosts. Taraxacum officinale, Ipomoea hederacea, Cyperus esculentus, Cynodon dactyIon, Echinochloa crus-galli, Eleusine indica, Sorghum halepense, Setaria viridis, Digitaria sanguinalis, and Datura stramonium were poor hosts for M. arenaria. Amaranthus palmeri, Amaranthus hybridus, Chenopodium album, Euphorbia maculata, Setaria lutescens, Vicia villosa, Sida spinosa, Rumex crispus, and Portulaca oleracea were moderate hosts and Ipomoea hederacea var. integriuscula, Xanthium strumarium, Cyperus esculentus, Cynodon dactylon, Paspalum notatum, Eleusine indica, Setaria viridis, and Rumex acetosella were poor hosts for M. incognita. None of the above were good hosts for M. incognita. Tobacco 'PD4' supported large numbers of both nematode species.

The application of preexisting DNA hybridization techniques was investigated for potential in determining populations of specific gene sequences in environmental samples. Cross-hybridizations among two degradative plasmids, TOL and NAH, and two cloning vehicles, pLAFR1 and RSF1010, were determined. The detection limits for the TOL plasmid against a nonhomologous plasmid-bearing bacterial background was ascertained. The colony hybridization technique allowed detection of one colony containing TOL plasmid among 10(6) Escherichia coli colonies of nonhomologous DNA. Comparisons between population estimates derived from growth on selective substrates and from hybridizations were examined. Findings indicated that standard sole carbon source enumeration procedures for degradative populations lead to overestimations due to nonspecific growth of other bacteria on the microcontaminant carbon sources present in the media. Population estimates based on the selective growth of a microcosm population on two aromatic substrates (toluene and naphthalene) and estimates derived from DNA-DNA colony hybridizations, using the TOL or NAH plasmid as a probe, corresponded with estimates of substrate mineralization rates and past exposure to environmental contaminants. The applications of such techniques are hoped to eventually allow enumeration of any specific gene sequences in the environment, including both anabolic and catabolic genes. In addition, this procedure should prove useful in monitoring recombinant DNA clones released into environmental situations.
