Organosolv lignin extracted from vine pruning residues was added to hydroxypropyl methylcellulose (HPMC)-based films using three strategies: i) lignin incorporated into the film (lignin-based film), ii) lignin nanoparticles (LNPs) incorporated into the film (LNPs-based film), and iii) lignin coated on HPMC films' surface (lignin-coated film). The films obtained were evaluated in terms of morphology, water barrier and mechanical properties, and antioxidant capacity. Results showed that LNPs incorporation did not affect the films´ water vapour permeability (WVP). Nonetheless, the lignin-based and lignin-coated films improved the water barrier properties of HPMC-based films, achieving a 31.5 and 36 % reduction of WVP, respectively. The morphological evaluation, performed by scanning electron microscopy, revealed films' morphology changes with the lignin incorporation, which was more evident in the lignin-based films. Fourier transform infrared spectroscopy (FTIR) showed minor changes in the film's structure using the different lignin incorporation methods. The mechanical properties were improved, including a significant increase in the tensile strength in the lignin-based and lignin-coated films. All films showed high radical scavenging activity (RSA) after 24 h, with a gradual increase in the lignin-coated films over time. The lignin-coated films showed to be the most promising incorporation strategy to improve the HPMC-based film's properties.

Cardiovascular diseases, resulting from the deposition of clots in blood vessels, are the leading cause of death worldwide. Fibrinolytic enzymatic activity can catalyze blood clot degradation. Findings show that 36 fungal isolates recovered from Caatinga soils have the potential to produce fibrinolytic protease under submerged conditions. About 58 % of the isolates displayed fibrinolytic activity above 100 U/mL, with Mucor subtilissimus UCP 1262 being the most active. The protease was biochemically and biophysically characterized, showing that the enzyme had a high affinity for SAApNA substrate and was significantly inhibited by fluoride methyl phenyl sulfonyl-C7H7FO2S, suggesting that it is a chymotrypsin-like serine protease. The highest enzyme activity was detected at pH 5.0 and 28 °C. This fibrinolytic protease's far-UV circular dichroism (CD) showed that its secondary structure was primarily α-helical. The purified fibrinolytic enzyme may represent a novel therapeutic agent for treating thrombosis. At temperatures above 65 °C, the enzyme lost all its secondary structure. Its melting temperature was 58.1 °C, the denaturation enthalpy 85.1 kcal/mol, and the denaturation entropy 0.26 kcal/K∙mol.

This study investigates the thermochemical decomposition and gasification performance of biochars produced from blends of waste lignocellulosic biomass and waste insulation electrical cables at varying temperatures. Characterization tests revealed changes, particularly in ash content (27.5 %-34 %) and elemental composition, with nitrogen content increasing notably in biochar samples compared to the original feedstock. Van Krevelen diagrams demonstrated a reduction in O/C and H/C ratios with increasing production temperature, resembling fossil fuels more closely. The thermogravimetric and the derived thermogravimetric profiles illustrated distinct degradation stages influenced by heating rates and production temperature. Macro-TGA tests provided insights into biomass residue behavior under gasification conditions, indicating higher reaction rates at elevated temperatures. Syngas analysis highlighted the impact of temperature and equivalence ratio on syngas composition, with higher temperatures favoring hydrogen-rich gas production. The observed trends in cold gas efficiency (42.61 %-50.40 %) and carbon conversion efficiency (45.83 %-50.40 %) underscore the significance of temperature control in maximizing gasification performance. Biochars produced at higher temperatures demonstrated superior gasification performance, suggesting potential for optimizing biochar production processes to enhance energy recovery and waste valorization.

Non-alcoholic fatty liver disease (NAFLD) presents a pervasive global pandemic, affecting approximately 25 % of the world's population. This grave health issue not only demands urgent attention but also stands as a significant economic concern on a global scale. The genesis of NAFLD can be primarily attributed to unhealthy dietary habits and a sedentary lifestyle, albeit certain genetic factors have also been recorded to contribute to its occurrence. NAFLD is characterized by fat accumulation in more than 5 % of hepatocytes according to histological analysis, or >5.6 % of lipid volume fraction in total liver weight in patients. The pathophysiology of NAFLD/non-alcoholic steatohepatitis (NASH) is multifactorial and the mechanisms underlying the progression to advanced forms remain unclear, thereby representing a challenge to disease therapy. Despite the substantial efforts from the scientific community and the large number of pre-clinical and clinical trials performed so far, only one drug was approved by the Food and Drug Administration (FDA) to treat NAFLD/NASH specifically. This review provides an overview of available information concerning emerging molecular targets and drug candidates tested in clinical studies for the treatment of NAFLD/NASH. Improving our understanding of NAFLD pathophysiology and pharmacotherapy is crucial not only to explore new molecular targets, but also to potentiate drug discovery programs to develop new therapeutic strategies. This knowledge endeavours scientific efforts to reduce the time for achieving a specific and effective drug for NAFLD or NASH management and improve patients' quality of life.

BACKGROUND: Childhood obesity presents a significant public health concern globally, with implications for cardiovascular health and metabolic syndrome. In Portugal, approximately 31.6% of children are affected, highlighting the urgency for intervention strategies. This study aimed to assess the prevalence of overweight and obesity in Portuguese school-aged children, with a focus on sex and age differences. METHODS: Anthropometric measurements were conducted on 1564 children aged 6-10 years, including weight, height, and skinfold thickness. Body Mass Index (BMI) and the percentage of body fat were calculated using established methods. RESULTS: The results revealed significant differences in BMI (≤0.001) and body fat percentage (≤0.001) among different BMI categories, with a notable prevalence of overweight and obesity, particularly among boys. A total of 37% of the studied population is overweight or obese, among which 40.1% and 33.9% are boys and girls, respectively. CONCLUSIONS: This study highlights statistically significant differences in BMI and body fat percentage for both sexes in different BMI categories. A large proportion of the population is overweight or obese, with a greater prevalence in boys. In short, childhood obesity has a negative impact on body composition and is associated with significant differences in anthropometric parameters, emphasizing the importance of preventative and intervention strategies to address this health problem.

This paper details the interoperability challenges and methodologies employed, highlighting the UNICOM Test Lab's role in validating and testing tools for IDMP standards implementation, aiming for semantic interoperability and a safe pharmaceutical supply chain within the EU.

Boletus edulis (BE) is a mushroom well known for its taste, nutritional value, and medicinal properties. The objective of this work was to study the biological effects of BE extracts on human colon carcinoma cells (Caco-2), evaluating parameters related to oxidative stress and inflammation. In this study, a hydroethanolic extract of BE was obtained by ohmic heating green technology. The obtained BE extracts are mainly composed of sugars (mainly trehalose), phenolic compounds (taxifolin, rutin, and ellagic acid), and minerals (K, P, Mg, Na, Ca, Zn, Se, etc.). The results showed that BE extracts were able to reduce cancer cell proliferation by the induction of cell cycle arrest at the G0/G1 stage, as well as cell death by autophagy and apoptosis, the alteration of mitochondrial membrane potential, and caspase-3 activation. The extracts modified the redox balance of the cell by increasing the ROS levels associated with a decrease in the thioredoxin reductase activity. Similarly, BE extracts attenuated Caco-2 inflammation by reducing both iNOS and COX-2 mRNA expression and COX-2 protein expression. In addition, BE extracts protected the intestine from the oxidative stress induced by H2O2. Therefore, this study provides information on the potential use of BE bioactive compounds as anticancer therapeutic agents and as functional ingredients to prevent oxidative stress in the intestinal barrier.

Utilizing techniques for reducing multivariate data is essential for comprehensively understanding the variations and relationships within both biomechanical and physiological datasets in the context of youth football training. Therefore, the objective of this study was to identify the primary factors influencing training sessions within a standard microcycle among young sub-elite football players. A total of 60 male Portuguese youth sub-elite footballers (15.19 ± 1.75 years) were continuous monitored across six weeks during the 2019-2020 in-season, comprising the training days from match day minus (MD-) 3, MD-2, and MD-1. The weekly training load was collected by an 18 Hz global positioning system (GPS), 1 Hz heart rate (HR) monitors, the perceived exertion (RPE) and the total quality recovery (TQR). A principal component approach (PCA) coupled with a Monte Carlo parallel analysis was applied to the training datasets. The training datasets were condensed into three to five principal components, explaining between 37.0% and 83.5% of the explained variance (proportion and cumulative) according to the training day (p < 0.001). Notably, the eigenvalue for this study ranged from 1.20% to 5.21% within the overall training data. The PCA analysis of the standard microcycle in youth sub-elite football identified that, across MD-3, MD-2, and MD-1, the first was dominated by the covered distances and sprinting variables, while the second component focused on HR measures and training impulse (TRIMP). For the weekly microcycle, the first component continued to emphasize distance and intensity variables, with the ACC and DEC being particularly influential, whereas the second and subsequent components included HR measures and perceived exertion. On the three training days analyzed, the first component primarily consisted of variables related to the distance covered, running speed, high metabolic load, sprinting, dynamic stress load, accelerations, and decelerations. The high intensity demands have a high relative weight throughout the standard microcycle, which means that the training load needs to be carefully monitored and managed.

The aim of this study was to test a machine learning (ML) model to predict high-intensity actions and body impacts during youth football training. Sixty under-15, -17, and -19 sub-elite Portuguese football players were monitored over a 6-week period. External training load data were collected from the target variables of accelerations (ACCs), decelerations (DECs), and dynamic stress load (DSL) using an 18 Hz global positioning system (GPS). Additionally, we monitored the perceived exertion and biological characteristics using total quality recovery (TQR), rating of perceived exertion (RPE), session RPE (sRPE), chronological age, maturation offset (MO), and age at peak height velocity (APHV). The ML model was computed by a feature selection process with a linear regression forecast and bootstrap method. The predictive analysis revealed that the players' MO demonstrated varying degrees of effectiveness in predicting their DEC and ACC across different ranges of IQR. After predictive analysis, the following performance values were observed: DEC (x¯predicted = 41, ß = 3.24, intercept = 37.0), lower IQR (IQRpredicted = 36.6, ß = 3.24, intercept = 37.0), and upper IQR (IQRpredicted = 46 decelerations, ß = 3.24, intercept = 37.0). The player's MO also demonstrated the ability to predict their upper IQR (IQRpredicted = 51, ß = 3.8, intercept = 40.62), lower IQR (IQRpredicted = 40, ß = 3.8, intercept = 40.62), and ACC (x¯predicted = 46 accelerations, ß = 3.8, intercept = 40.62). The ML model showed poor performance in predicting the players' ACC and DEC using MO (MSE = 2.47-4.76; RMSE = 1.57-2.18: R2 = -0.78-0.02). Maturational concerns are prevalent in football performance and should be regularly checked, as the current ML model treated MO as the sole variable for ACC, DEC, and DSL. Applying ML models to assess automated tracking data can be an effective strategy, particularly in the context of forecasting peak ACC, DEC, and bodily effects in sub-elite youth football training.

The aim of this study was to analyze the match-to-match variation in high-intensity demands from one Portuguese professional football team according to playing positions. Twenty-three male outfield professional football players were observed during eighteen matches of the Portuguese Second League. Time-motion data were collected using Global Positioning System (GPS) technology. Match running performance was analyzed based on the following three playing positions: defenders (DF), midfielders (MF), and forwards (FW). Repeated measures ANOVA was utilized to compare match running performance within each position role, and seasonal running variation. Practical differences were assessed using the smallest worthwhile change (SWC), coefficient of variation (CV), and twice the coefficient of variation (2CV). Significant differences were found among playing positions in total distance covered (F = 15.45, p < 0.001, η2 = 0.33), average speed (F = 12.79, p < 0.001, η2 = 0.29), high-speed running (F = 16.93, p < 0.001, η2 = 0.36), sprinting (F = 13.49, p < 0.001, η2 = 0.31), accelerations (F = 4.69, p = 0.001, η2 = 0.132), and decelerations (F = 12.21, p < 0.001, η2 = 0.284). The match-to-match running performance encompassed TD (6.59%), AvS (8.67%), HSRr (37.83%), SPR (34.82%), ACC (26.92%), and DEC (27.85%). CV values for total distance covered ranged from 4.87-6.82%, with forwards and midfielders exhibiting the greatest and smallest variation, respectively. Midfielders demonstrated the highest match-to-match variation for all other analyzed variables (8.12-69.17%). All playing positions showed significant variation in high-demanding variables (26.94-37.83%). This study presents the initial analysis of match-to-match variation in high-intensity demands within a Portuguese professional football team. Thus, the position's specificity and context can provide a helpful strategy for evaluating match-to-match running performance, and for recommending individualized training exercises based on the peak and high-intensity demands for each player's role within the game.

In this work, a multivariate analysis was carried out, using a Plackett-Burman (PB) design involving seventeen growth parameters, on carotenoids production of Pavlova gyrans (p < 0.10). Each assay was analysed regarding its content (mg g-1) of fucoxanthin (Fx), diatoxanthin, diadinoxanthin, ß-carotene (ßCar), α-carotene, and the sum of all carotenoids analysed individually (TCar). According to the statistical analysis, modified medium formulations were developed for the particular cases of Fx, ßCar, and TCar. The study showed that Fx content was positively affected by nitrogen supplementation and lower light intensities. Higher concentrations of nitrogen and iron increased the final content of ßCar as well. Similarly, salinity, light intensity, nitrogen, iron, and cobalt were identified as key factors in TCar production. The PB-based formulations showed significant improvements (p < 0.05) for TCar (11.794 mg g-1) and Fx (6.153 mg g-1) when compared to the control conditions (Walne's medium-2.010 mg g-1). Furthermore, effective control of key variables (e.g., light intensity) throughout P. gyrans growth proved successful (p < 0.05), increasing the productivity of Fx (0.759 mg L-1 d-1) and TCar (1.615 mg L-1 d-1).

OBJECTIVE: To study the possible role for HMGA2 overexpression in differentiated myometrial cells and its potential to induce a stem cell-like or dedifferentiating phenotype and drive fibroid development. DESIGN: Myometrial cells were immortalized and transduced with an HMGA2 lentivirus to produce HMGA2hi cells. In vitro stem cell assays were conducted, and ribonucleic acid from HMGA2hi and control cells as well as fibroid-free myometrial and HMGA2 fibroid (HMGA2F) tissues were submitted for ribonucleic acid sequencing. SETTING: University research laboratory. PATIENT(S): Women who underwent hysterectomy for symptomatic uterine fibroids or other gynecological conditions. INTERVENTION(S): Not applicable. MAIN OUTCOME MEASURE(S): In vitro stem cell-like properties from myometrial cell lines. Ribonucleic acid sequencing and collagen production of HMGA2-overexpressing primary leiomyoma tissue and cell lines. RESULT(S): HMGA2hi cells had enhanced self-renewal capacity, decreased proliferation, and a greater ability to differentiate into other mesenchymal cell types. HMGA2hi cells exhibited a stem cell-like signature and shared transcriptomic similarities with HMGA2F. Moreover, dysregulated extracellular matrix pathways were observed in both HMGA2hi cells and HMGA2F. CONCLUSION(S): Our findings show that HMGA2 overexpression may drive myometrial cells to dedifferentiate into a more plastic phenotype and provide evidence for an alternative mechanism for fibroid etiology, suggesting that fibroids arise not only from a mutated stem cell but also from a mutated differentiated myometrial cell.

Microalgae are a group of microorganisms, mostly photoautotrophs with high CO2 fixation capacity, that have gained increased attention in the last decades due to their ability to produce a wide range of valuable metabolites, such as carotenoids and polyunsaturated fatty acids, for application in food/feed, pharmaceutical, and cosmeceutical industries. Their increasing relevance has highlighted the importance of identifying and culturing new bioactive-rich microalgae species, as well as of a thorough understanding of the growth conditions to optimize the biomass production and master the biochemical composition according to the desired application. Thus, this review intends to describe the main cell processes behind the production of carotenoids and polyunsaturated fatty acids, in order to understand the possible main triggers responsible for the accumulation of those biocompounds. Their economic value and the biological relevance for human consumption are also summarized. In addition, an extensive review of the impact of culture conditions on microalgae growth performance and their biochemical composition is presented, focusing mainly on the studies involving Pavlovophyceae species. A complementary description of the biochemical composition of these microalgae is also presented, highlighting their potential applications as a promising bioresource of compounds for large-scale production and human and animal consumption.

Uterine fibroids are prevalent benign tumors in women that exhibit considerable heterogeneity in clinical presentation and molecular characteristics, necessitating a deeper understanding of their etiology and pathogenesis. HMGA2 overexpression has been associated with fibroid development, yet its precise role remains elusive. Mutations in fibroids are mutually exclusive and largely clonal, suggesting that tumors originate from a single mutant cell. We explored a possible role for HMGA2 overexpression in differentiated myometrial cells, hypothesizing its potential to induce a stem cell-like or dedifferentiating phenotype and drive fibroid development. Myometrial cells were immortalized and transduced with an HMGA2 lentivirus to produce HMGA2hi cells. In vitro stem cell assays were conducted and RNA from HMGA2hi and control cells and fibroid-free myometrial and HMGA2 fibroid (HMGA2F) tissues were submitted for RNA-sequencing. HMGA2hi cells have enhanced self-renewal capacity, decreased proliferation, and have a greater ability to differentiate into other mesenchymal cell types. HMGA2hi cells exhibit a stem cell-like signature and share transcriptomic similarities with HMGA2F. Moreover, dysregulated extracellular matrix pathways are observed in both HMGA2hi cells and HMGA2F. Our findings suggest that HMGA2 overexpression drives myometrial cells to dedifferentiate into a more plastic phenotype and underscore a pivotal role for HMGA2 in fibroid pathogenesis.

The conjugation of proteins with polymers offers immense biotechnological potential by creating novel macromolecules. This article presents experimental findings on the structural properties of maltose-binding protein (MBP) conjugated with linear biodegradable polyphosphoester polymers with different molecular weights. We studied isotopic effects on both proteins and polymers. Circular dichroism and fluorescence spectroscopy and small-angle neutron scattering reveal that the conjugation process destabilizes the protein, affecting the secondary more than the tertiary structure, even at room temperature, and that the presence of two domains in the MBP may contribute to its observed instability. Notably, unfolding temperatures differ between native MBP and the conjugates. In particular, this study sheds light on the complex interplay of factors such as the deuteration influencing protein stability and conformational changes in the conjugation processes. The perdeuteration influences the hydrogen bond network and hydrophobic interactions in the case of the MBP protein. The perdeuteration of the protein influences the hydrogen bond network and hydrophobic interactions. This is evident in the decreased thermal stability of deuterated MBP protein, in the conjugate, especially with high-molecular-mass polymers.

Food fraud refers to deceptive practices conducted for economic gain, and incidents of such fraud are often reported in the media and scientific literature. However, little is known about how European consumers perceive food fraud. To address this gap, a study explored Portuguese consumers' knowledge and perceptions of food fraud using qualitative methods such as free word association and semi-structured interviews. For this research, 340 participants were recruited, providing 911 valid words, classified into categories, major categories, and dimensions. Differences between consumers' previous exposure to food fraud and sociodemographic characteristics were explored. Additionally, other thirty-six participants were selected and interviewed, following a semi-structured guide. Interviews were transcribed, coded, and analyzed using a thematic analysis procedure. The results suggest that Portuguese consumers view food fraud as a morally reprehensible deception and are aware of its causes and impacts. However, not all consumers know the different forms of food fraud or the types of products vulnerable to fraud. Among the most repeated words were "deception", "expiration date", and "falsification". Despite this food fraud awareness, most consumers believed they were not exposed to food fraud and stated that they do not conduct daily practices to reduce exposure to it. Following the chi-square and Mann-Whitney tests, significant differences (p ≤ 0.05) were identified between participants exposed and not exposed to food fraud. The study also found that consumers with higher education and self-reported exposure to food fraud had a better understanding of the issue. This study provides insights for quantitative research on consumer perceptions and beliefs about food fraud to explore further vulnerable food categories and types of food fraud in real-world scenarios.

The apicomplexan parasite Cryptosporidium is a leading cause of childhood diarrhea in developing countries. Current treatment options are inadequate and multiple preclinical compounds are being actively pursued as potential drugs for cryptosporidiosis. Unlike most apicomplexans, Cryptosporidium spp. sequentially replicate asexually and then sexually within a single host to complete their lifecycles. Anti-cryptosporidial compounds are generally identified or tested through in vitro phenotypic assays that only assess the asexual stages. Therefore, compounds that specifically target the sexual stages remain unexplored. In this study, we leveraged the ReFRAME drug repurposing library against a newly devised multi-readout imaging assay to identify small-molecule compounds that modulate macrogamont differentiation and maturation. RNA-seq studies confirmed selective modulation of macrogamont differentiation for 10 identified compounds (9 inhibitors and 1 accelerator). The collective transcriptomic profiles of these compounds indicates that translational repression accompanies Cryptosporidium sexual differentiation, which we validated experimentally. Additionally, cross comparison of the RNA-seq data with promoter sequence analysis for stage-specific genes converged on a key role for an Apetala 2 (AP2) transcription factor (cgd2_3490) in differentiation into macrogamonts. Finally, drug annotation for the ReFRAME hits indicates that an elevated supply of energy equivalence in the host cell is critical for macrogamont formation.

This study assessed the chemical profiles and bioactivities of the infusions, decoctions and hydroethanolic extracts of tarragon, basil and French lavender. The extracts were chemically characterised (HPLC-DAD-ESI/MS) and their bioactivities were evaluated in vitro. All extracts revealed antimicrobial, antifungal and antioxidant properties. French lavender extracts showed higher total phenolic content, regardless of the extraction method used, and antioxidant and antitumour capacities, but no anti-inflammatory action. All basil and two of the tarragon extracts revealed anti-inflammatory power. Thus, tarragon, basil and French lavender extracts may be considered for inclusion in foods, as preservatives or functional ingredients. Nonetheless, further studies must be conducted to evaluate the pharmacokinetic parameters of the bioactive compounds.

A significant fraction of the food produced worldwide is currently lost or wasted throughout the supply chain, squandering natural and economic resources. Food waste valorization will be an important necessity in the coming years. This work investigates the ability of food waste to serve as a viable nutritional substrate for the heterotrophic growth of Chlorella vulgaris. The impact of different pretreatments on the elemental composition and microbial contamination of seven retail food waste mixtures was evaluated. Among the pretreatment methods applied to the food waste formulations, autoclaving was able to eliminate all microbial contamination and increase the availability of reducing sugars by 30%. Ohmic heating was also able to eliminate most of the contaminations in the food wastes in shorter time periods than autoclave. However, it has reduced the availability of reducing sugars, making it less preferable for microalgae heterotrophic cultivation. The direct utilization of food waste containing essential nutrients from fruits, vegetables, dairy and bakery products, and meat on the heterotrophic growth of microalgae allowed a biomass concentration of 2.2 × 108 cells·mL-1, being the culture able to consume more than 42% of the reducing sugars present in the substrate, thus demonstrating the economic and environmental potential of these wastes.
