RESUMO

The present study deals with the metabolomic status of Ulva cells undergoing phase transition (vegetative, determination and differentiation) when exposed to different abiotic conditions. The objective was to study whether metabolite changes occurring during the phase transition reveal any commonality among differential abiotic conditions. The phase transition was followed through microscopic observations and 1H NMR characterization at 0 h, 24 h, and 48 h after the incubation of the thallus under abiotic conditions, such as different salinities (20-35 psu), temperatures (20-35 °C), photoperiods (18 : 6, 12 : 12, and 6 : 18 D/N), light intensities (220, 350, and 500 µmol photons m-2 s-1), nitrate (0.05-0.2 g L-1) and phosphate (0.05-0.2 g L-1) concentrations. Microscopic analysis revealed the role of all abiotic conditions except variable salinity and phosphate concentration in phase transition. NMR analysis revealed that glucose increased in the determination phase [7.58 to 9.62 normalized intensity (AU)] and differentiation phase (5.85 to 6.41 AU) from 20 °C to 25 °C temperature. Coniferyl aldehyde increased in vegetative (5.79 to 6.83 AU) and differentiation (6.66 to 7.40 AU) phases from 20 °C to 30 °C temperature. The highest average (22.97) was found in photoperiod (average range = 0-122.91) and the highest SD (24.73) in salinity (SD range = 1.86-57.04) in region 9 (creatinine and cysteine) of the differentiation phase. A total of 30 metabolites were identified under the categories of sugars, amino acids, and aromatic compounds. The present study will aid in understanding the mechanisms underlying cell differentiation during reproduction. The result may serve as an important reference point for future studies, besides helping in controlling seedling preparation for commercial farming as well as the management of rapid green tide formation.

RESUMO

The custard apple (Annona squamosa) fruits were procured from local market, irradiated with radiation doses 0, 0.25, 0.50, 0.75, 1.00, 1.25, 1.50, 1.75 kGy and then treated with benzyl adenine (50 and 100 part per million) and stored at ambient temperature (25 ± 5 °C, Relative Humidity 90 ± 2%) for 12 days. The treated fruits were evaluated for sensory (viz; flavour, texture, internal and external colour) and chemical constituents (viz; Total Soluble Solids, titrable acidity, ascorbic acid, free soluble sugar, reducing sugar. non reducing sugar, carbohydrate) during storage. The study concluded that radiation dose of 1.5 kilo Gray along with 50 ppm benzyl adenine enhanced in shelf-life of custard apple fruits by 6 days at ambient temperature with good pulp texture, flavour, colour and nutritional quality as compared to control.

RESUMO

We demonstrate a pulse sequence using symmetry-based rotor-synchronised RN(n)(nu) sequences for homonuclear dipolar decoupling that achieves pure absorption phase high-resolution (1)H spectra in solid-state NMR. This sequence is compared with the phase-modulated Lee-Goldburg scheme. Experimental results are shown for samples of glycine and L-histidine.HCl.H(2)O for magic-angle-spinning frequencies in the range of 14-30 kHz and at two different magnetic fields.

RESUMO

We compare the performance of the windowed phase-modulated Lee-Goldburg (wPMLG) and the windowed decoupling using mind boggling optimisation (wDUMBO) sequences at various magic-angle spinning rates and nutation frequencies of the pulses. Additionally, we introduce a supercycled version of wDUMBO and compare its efficiency with that of the non-supercycled implementation of wDUMBO. The efficiency of the supercycled version of wPMLG, denoted wPMLG-S2, is compared with a new supercycled version of wPMLG that we notate as wPMLG-S3. The interaction between the supercycled homonuclear dipolar decoupling sequences and the sample rotation is analysed using symmetry-based selection rules.

The design of variants of the swept-frequency two-pulse phase modulation sequence for heteronuclear dipolar decoupling in solid-state NMR is reported, their performance evaluated, and compared with other established sequences like TPPM and SPINAL. Simulations performed to probe the role of the homonuclear (1)H-(1)H bath show that the robustness of the decoupling schemes improves with the size of the bath. In addition, these simulations reveal that the homonuclear (1)H-(1)H bath also leads to broad baselines at high MAS rates. Results from a study of the SPINAL decoupling scheme indicate that optimisation of the starting phase and phase increment improves its performance and efficiency at high MAS rates. Additionally, experiments performed on a liquid crystal display the role of the initial phase in SPINAL-64 and sequences in the SW(f)-TPPM family.

Recently, a sequence for heteronuclear dipolar decoupling in solid-state NMR, namely SWf-TPPM, was introduced by us. Under magic-angle spinning (MAS), the decoupling efficiency of the sequence was unaffected over a range of values for various experimental parameters such as the pulse length, pulse phase, and 1H resonance offset. We here demonstrate its use in multiple-quantum (MQ) and high-resolution (HR) MAS experiments. This sequence further improves the MQMAS spectra compared to the earlier reported decoupling sequences with improved immunity to any missets of the pulse length, pulse phase and decoupler offset. In contrast, for HRMAS, the simple CW scheme is as efficient as any of the decoupling schemes that were studied.

A theoretical treatment of heteronuclear dipolar decoupling in solid-state nuclear magnetic resonance is presented here based on bimodal Floquet theory. The conditions necessary for good heteronuclear decoupling are derived. An analysis of a few of the decoupling schemes implemented until date is presented with regard to satisfying such decoupling conditions and efficiency of decoupling. Resonance conditions for efficient heteronuclear dipolar decoupling are derived with and without the homonuclear (1)H-(1)H dipolar couplings and their influence on heteronuclear dipolar decoupling is pointed out. The analysis points to the superior efficiency of the newly introduced swept two-pulse phase-modulation (SW(f)-TPPM) sequence. It is shown that the experimental robustness of SW(f)-TPPM as compared to the original TPPM sequence results from an adiabatic sweeping of the modulation frequencies. Based on this finding alternative strategies are compared here. The theoretical findings are corroborated by both numerical simulations and representative experiments.

Recently we introduced a radiofrequency pulse scheme for heteronuclear dipolar decoupling in solid-state nuclear magnetic resonance under magic-angle spinning [R.S. Thakur, N.D. Kurur, P.K. Madhu, Swept-frequency two-pulse phase modulation for heteronuclear dipolar decoupling in solid-state NMR, Chem. Phys. Lett. 426 (2006) 459-463]. Variants of this sequence, swept-frequency TPPM, employing frequency modulation of different types have been further tested to improve the efficiency of heteronuclear dipolar decoupling. Among these, certain sequences that were found to perform well at lower spinning speeds are demonstrated here on a liquid-crystal sample of MBBA for application in static samples. The new sequences are compared with the standard TPPM and SPINAL schemes and are shown to perform better than them. These modulated schemes perform well at low decoupler radiofrequency power levels and are easy to implement on standard spectrometers.
