RESUMO

Cartilaginous fishes (chimaeras and elasmobranchs -sharks, skates and rays) hold a key phylogenetic position to explore the origin and diversifications of jawed vertebrates. Here, we report and integrate reference genomic, transcriptomic and morphological data in the small-spotted catshark Scyliorhinus canicula to shed light on the evolution of sensory organs. We first characterise general aspects of the catshark genome, confirming the high conservation of genome organisation across cartilaginous fishes, and investigate population genomic signatures. Taking advantage of a dense sampling of transcriptomic data, we also identify gene signatures for all major organs, including chondrichthyan specializations, and evaluate expression diversifications between paralogs within major gene families involved in sensory functions. Finally, we combine these data with 3D synchrotron imaging and in situ gene expression analyses to explore chondrichthyan-specific traits and more general evolutionary trends of sensory systems. This approach brings to light, among others, novel markers of the ampullae of Lorenzini electro-sensory cells, a duplication hotspot for crystallin genes conserved in jawed vertebrates, and a new metazoan clade of the Transient-receptor potential (TRP) family. These resources and results, obtained in an experimentally tractable chondrichthyan model, open new avenues to integrate multiomics analyses for the study of elasmobranchs and jawed vertebrates.

Myriophyllum spicatum, more commonly known as Eurasian watermilfoil (EWM), is one of the most invasive aquatic plants in North America, causing negative ecological and economic impacts in ecosystems where it proliferates. Many control strategies have been developed and implemented to mitigate EWM growth and spread, although the results are mixed and there is no consensus on lake-specific strategies. Here, we describe the development of a predictive model using a support vector technique, that predicts the success of biological pest control using Euhrychiopsis lecontei (the milfoil weevil), a milfoil specialist, to reduce EWM in lakes. Such a model is informed by lake characteristics (limnological and landscape) and augmentation strategies. To develop our predictive model, we performed a metadata analysis from 133 published peer-reviewed literature and professional reports of milfoil weevil augmentation field experiments that contained information on lake characteristics. The predictive model's algorithm uses a support vector machine (SMV) to learn patterns among lake characteristics, along with the recorded augmentation strategy and the reported success of each study, where success is a measure of EWM change over a season and is recorded in a variety of ways (e.g., EWM biomass change, EWM percent change, EWM visual change, etc.,). Overall, the model results suggests that shallower lakes, more frequent weevil augmentations, and larger weevil overwintering habitat are the most important predictors for EWM reduction success by weevil augmentation. Although watermilfoil weevil augmentation is a promising mitigation strategy, it may not work for all lakes. However, in terms of suggesting weevil augmentation, our model is a valuable tool for lake stakeholders and resource managers, who can use it to determine whether milfoil weevil augmentation, which can be very costly due to the difficulties in finding and raising milfoil weevils, will be a useful and sustainable approach to control EWM in their lake community.

Black carbon (BC) emissions from aircraft engines lead to an increase in the atmospheric burden of fine particulate matter (PM2.5). Exposure to PM2.5 from sources, including aviation, is associated with an increased risk of premature mortality, and BC suspended in the atmosphere has a warming impact on the climate. BC particles emitted from aircraft also serve as nuclei for contrail ice particles, which are a major component of aviation's climate impact. To facilitate the evaluation of these impacts, we have developed a method to estimate BC mass and number emissions at the engine exit plane, referred to as the Smoke Correlation for Particle Emissions-CAEP11 (SCOPE11). We use a data set consisting of SN-BC mass concentration pairs, collected using certification-compliant measurement systems, to develop a new relationship between smoke number (SN) and BC mass concentration. In addition, we use a complementary data set to estimate measurement system loss correction factors and particle geometric mean diameters to estimate BC number emissions at the engine exit plane. Using this method, we estimate global BC emissions from aircraft landing and takeoff (LTO) operations for 2015 to be 0.74 Gg/year (95% CI = 0.64-0.84) and 2.85 × 1025 particles/year (95% CI = 1.86-4.49 × 1025).

INTRODUCTION: Posturographic measurements are often performed in hospital in the context of assessing fall risks in elderly subjects. These hospital visits may generate different kinds of stimuli that could influence test outcomes. STUDY AIM: The aim was to investigate whether posturographic measurements performed both at home and in hospital in a randomized order provide similar data. METHODS: Thirty-five healthy elderly subjects (average age 72.5 ± 3.7 years) were assigned to two postural evaluations performed in a random order: one at home and one at the hospital. Before the first evaluation, subject's levels of anxiety, depression and stress were assessed. Then, the area of body sway, velocity and medial-lateral and antero-posterior amplitudes were recorded twice, first with the subject's eyes opened and then with eyes closed. RESULTS AND DISCUSSION: The posturographic data obtained in hospital and at home were quite similar. However, when the group that was first evaluated in hospital was compared with the group first evaluated at home, the medio-lateral amplitude was observed to significantly decrease in the second evaluation compared with the data obtained in the first trial (P<0.05), and this decrease was significantly higher in the first group (P<0.05). For the eyes-opened condition, we found significant correlations between anxiety and the area of body sway, stress and the area of body sway, and anxiety and the medial-lateral amplitude. CONCLUSIONS: Psychological factors may influence some posturographic data, and carrying out posturographic evaluations at home for elderly subjects could be a reasonable strategy.

Identifying the social-demographic characteristics of elderly people who fall would enable us to define the typical profile of the elderly who are at risk of falling. OBJECTIVE: We aimed to isolate studies in which the social-demographic risk factors for falls among the elderly have been evaluated and to carry out a meta-analysis by combining the results of all of these selected studies. METHOD: We did a systematic literature review using the key words "accidental fall / numerical data" and "risk factors." Inclusion criteria entailed the selection of articles with the following characteristics: population of subjects aged 60 years or over, falls that took place in everyday life, and social-demographic risk factors for falls. RESULTS: 3,747 indexed articles published between 1981 and 2007 were identified, and 177 studies with available data were included, of which 129 had data on social-demographic risk factors for falls. Difficulties in activities of daily living (ADL) or in instrumental activities of daily living (IADL) double the risk of falling: The OR and 95% Cl were 2.26 (2.09, 2.45) for disturbance ADL and 2.10 (1.68, 2.64) for IADL. The OR and 95% Cl for Caucasians were 1.68 (0.98 - 2.88) and 0.64 (0.51 - 0.80) for Hispanics. In the subgroup of patients older than eighty, being married protected people from falling with an OR and 95% Cl =0.68 (0.53 - 0.87). CONCLUSION: Defining factors that create a risk of falling and protect elderly people from falls using social-demographic characteristics lets us focus on an "at risk" population for which a specific program could be developed.

Serially homologous structures are believed to originate from the redeployment of a genetic cascade in different locations of the body. Serial homologs may diverge at the genetic and morphological level and acquire developmental independency (individualization). Teeth are repeated units that form dentitions found on different bones of the oral-pharyngeal cavity in gnathostomes and provide a good model to study such processes. Previous comparisons of dlx gene expression patterns between mouse oral teeth and zebrafish pharyngeal teeth showed a high level of divergence. Furthermore, these genes are differentially expressed in different teeth of the zebrafish, and in the mouse they are responsible for tooth identity (incisors vs. molars). We examined the potential divergence of dlx gene expression between oral and pharyngeal teeth by examining the expression pattern in the development of the first generation teeth of the medaka and comparing it with data from the zebrafish and the mouse. Out of the seven medaka dlx genes, five are expressed during odontogenesis compared with six in both the zebrafish and the mouse. The only difference observed between oral and pharyngeal teeth in the medaka is an earlier expression of dlx5a in the oral dental epithelium. The subset of dlx genes expressed in the medaka, zebrafish, and mouse is slightly different but their detailed expression patterns are highly divergent. Our results demonstrate a low constraint on dlx gene expression shuffling in the odontogenic cascade within osteichtyans but the non-individualization of oral and pharyngeal dentitions in the medaka.

Phosphate (Pi) is an essential element for plant development and metabolism. Due to its low availability and mobility in soils, it is often a limiting nutrient for their growth. This phenomenon is reinforced by the formation of insoluble complexes in the environment with many cations, affecting the solubility of both phosphate and associated ions. This interaction is investigated here for iron, a strong phosphate chelator. Depleting the medium in phosphate clearly resulted in an increase of iron content in Arabidopsis. These modifications triggered molecular responses linked with iron status (transport, homeostasis and accumulation). Interestingly, physiological modifications affecting iron storage were also observed. The accumulation of phosphate/iron complexes in the vacuoles of plants grown in Pi-rich medium disappeared in Pi-depleted medium in favor of accumulation of iron inside the chloroplasts, likely associated with ferritin.

We have used dlx genes to test the hypothesis of a separate developmental program for dermal and cartilage bones within the neuro- and splanchnocranium by comparing expression patterns of all eight dlx genes during cranial bone formation in zebrafish from 1 day postfertilization (dPF) to 15 dPF. dlx genes are expressed in the visceral skeleton but not during the formation of dermal or cartilage bones of the braincase. The spatiotemporal expression pattern of all the members of the dlx gene family, support the view that dlx genes impart cellular identity to the different arches, required to make arch-specific dermal bones. Expression patterns seemingly associated with cartilage (perichondral) bones of the arches, in contrast, are probably related to ongoing differentiation of the underlying cartilage rather than with differentiation of perichondral bones themselves. Whether dlx genes originally functioned in the visceral skeleton only, and whether their involvement in the formation of neurocranial bones (as in mammals) is secondary, awaits clarification.

In order to investigate similarities and differences in genetic control of development among teeth within and between species, we determined the expression pattern of all eight Dlx genes of the zebrafish during development of the pharyngeal dentition and compared these data with that reported for mouse molar tooth development. We found that (i) dlx1a and dlx6a are not expressed in teeth, in contrast to their murine orthologs, Dlx1 and Dlx6; (ii) the expression of the six other zebrafish Dlx genes overlaps in time and space, particularly during early morphogenesis; (iii) teeth in different locations and generations within the zebrafish dentition differ in the number of genes expressed; (iv) expression similarities and differences between zebrafish Dlx genes do not clearly follow phylogenetic and linkage relationships; and (v) similarities and differences exist in the expression of zebrafish and mouse Dlx orthologs. Taken together, these results indicate that the Dlx gene family, despite having been involved in vertebrate tooth development for over 400 million years, has undergone extensive diversification of expression of individual genes both within and between dentitions. The latter type of difference may reflect the highly specialized dentition of the mouse relative to that of the zebrafish, and/or genome duplication in the zebrafish lineage facilitating a redistribution of Dlx gene function during odontogenesis.

A model system based on the Arabidopsis thaliana (L.) Heynh. Ws ecotype and its naturally colonizing Pseudomonas thivervalensis rhizobacteria was defined. Pseudomonas strains colonizing A. thaliana were found to modify the root architecture either in vivo or in vitro. A gnotobiotic system using bacteria labelled with green fluorescent protein revealed that P. thivervalensis exhibited a colonization profile similar to that of other rhizobacterial species. Mutants of A. thaliana affected in root hair development and possible hormone perception were used to analyze the plant genetic determinants of bacterial colonization. A screen for mutants insensitive to P. thivervalensis colonization yielded two mutants found to be auxin resistant. This further supports a proposed role for bacterial auxin in inducing morphological modifications of roots. This work paves the way for studying the interaction between plants and non-pathogenic rhizobacteria in a gnotobiotic system, derived from a natural association, where interactions between both partners can be genetically dissected.

A novel Arabidopsis DHDPS gene named DHDPS2 was found through identification of a mutant by promoter trapping. The mutation promotes a reduction of growth resulting from combination of a defect in lysine biosynthesis and accumulation of a toxic level of threonine or derived products. The mutant also modifies the amino acid composition issuing from the pyruvate and aspartate pathways, affecting mainly the root compartment. These data are in accordance with the expression of DHDPS2 in the root apex as visualized by expression of the GUS reporter gene. This suggests that a large proportion of the amino acids derived from pyruvate and aspartate are synthesized in this organ.

Cytochrome oxidase from both pea leaves and bovine heart shows lower activity under a mixture of 79% N2O/21% O2 than under ambient air. This inhibition is not detectable below 5 microM cytochrome c but appears with increasing concentrations of cytochrome c. These results suggest that the N2O-induced inhibition of cytochrome c oxidase is modulated by cytochrome c concentration. This seems to concern only the lowest affinity site of the oxidase. Apparently, N2O and cytochrome c do not share the same site of fixation on the oxidase.

The effects of the herbicide PROPYZAMIDE are studied in laboratory and field conditions. The modifications involved are characterized by measurement of 14C-glucose mineralization and radioactivity incorporation into the soil fractions. In laboratory conditions, temperature and moisture are kept stable and the experiment is performed during less than 24 hours. In these conditions, Kerb 50 (commercial formulation of propyzamide) and the emulsifier (material used in propyzamide formulation) exert little effect on 14CO2 evolution. In field conditions, propyzamide andKerb 50 are applied once at two different doses: at field rate (1,5 kg/ha) and twentyfold this rate. Essays are duplicated. The herbicide (propyzamide in Celanol and Kerb 50) and the emulsifiers alone (Celanol and the material used in propyzamide formulation) are applied on the soil surface (application date: 3.02.81). Two weeks later and then every month during four months, samples are taken to the depth of about 5 cm (Propyzamide migrates very slowly in the first centimeters of the soil). The characterization experiment is performed on 10 g soil samples by 14C-glucose incubation at 28 degrees C during two hours. 14CO2 evolved is measured after incubation and acidification with HCl. Then radioactivity distribution in the soil is counted after chemical fractionation of soil. This distribution is about 10-16.5% as 14CO2, 22-37% in the acid-soluble fraction, 10-25% in the alkali-soluble fraction and 15-45% in the human fraction (measured as 14CO2 evolved after combustion). This distribution is little modified by the herbicides or the emulsifiers but its evolution is significantly related to environmental conditions (temperature). Nevertheless a few modifications are observed. They can be due to the herbicide propyzamide itself but the emulsifiers and the degradation products of propyzamide can also influence the measurement (After forty days in the soil, 70-95% of the starting active ingredient have disappeared). They can also be a result of the initial effects of the products (modification of the microflora and of the environment).
