Objectives: The objective of this study was to assess the postures that were commonly used in automobile chassis repair operations, and to evaluate shoulder girdle muscle fatigue for different combinations of the weight of hand-tools. Material and Methods: Two right muscles, including upper trapezius (UT) and middle deltoid (MD), were selected. Surface electromyography (SEMG) and a perceived level of discomfort (PLD) were used to assess the degree of shoulder girdle fatigue. Fifteen healthy young male subjects from the Northwestern Polytechnical University participated in the test. The test consisted of assuming 4 different postures and maintaining each of them for 60 s. The 4 postures varied in terms of dumbbell weights, standing for the hand-tools weight: W1 was 0.48 kg and W2 was 0.75 kg; the 4 shoulder postures were shoulder flexions of 150°, 120°, 90°, and 60°, combined with an included elbow angle of 180°, 150°, 120° and 90°, respectively. The experimental sequences were randomly selected. The signals of SEMG and the values of PLD in the shoulder girdle were recorded in 60 s. All subjects completed the whole test. The repeated measure analysis of variance (ANOVA) was performed to ascertain differences between dumbbell weight (0.48 kg and 0.75 kg) and shoulder postures (150°/180°, 120°/150°, 90°/120° and 60°/90°). The Friedman test was utilized to determine the significant differences for UT(PLD) and MD(PLD) on shoulder postures. Spearman's correlation was used to analyze the relationship between the subjective and objective measurements. Results: Significant correlational relationships existed between the UT percentage of the maximal voluntary electrical activation (%MVE) and UT(PLD) (r = 0.459, p < 0.01), between MD(%MVE) and MD(PLD) (r = 0.821, p < 0.01). The results showed that SEMG and PLD of the 4 postures under analysis differed significantly (p < 0.05). Conclusions: It was indicated that posture T4 (shoulder forward flexion 60° and included elbow angle 90°) resulted in the lowest fatigue, both in terms of the objective measure and the subjective perception, which meant that this posture was more ergonomic. BACKGROUND: Sit-stand workstations are available for office work purposes but there is a dearth of quantitative evidence to state benefits for lower limb outcomes while using them. And there are no guidelines on what constitutes appropriate sit/stand time duration. The primary aim of this study has been to compare muscle activity and perceived discomfort in the lower extremity during various combinations of sit/stand time duration associated with a sit-stand workstation separately and to evaluate the effects of the sit-stand workstation on the lower extremity during the text entry task. MATERIAL AND METHODS: During the 5 days, all participants completed a 2-h text entry task each day for various sit/stand time duration combinations as follows: 5/25 min, 10/20 min, 15/15 min, 20/10 min, 25/5 min. Lower extremity muscular exposure of 12 male and 13 female participants was collected at 8 sites by surface electromyography and body discomfort was calculated by a questionnaire under those 5 conditions. RESULTS: Results have demonstrated that lower extremity muscle activity has been significantly varied among the 5 sit/stand time duration groups. Perceived level of discomfort (PLD) has not differed significantly for 9 out of 10 body parts. CONCLUSIONS: The muscle activity of the thigh region was influenced by sit/stand time duration significantly. Ergonomic exposures of lower extremity when using a sit-stand workstation were increased, particularly during the long time standing posture. Results indicate that body mass index (BMI) and gender were not significant factors in this study. Combination of sit/stand time duration 25/5 min appears to show positive effects on relief of muscle exposure of back of thigh in the shifts of sitting and standing work position. In this study, a straight diagonal-pitched blade stirrer was designed, built and characterized in a 5-L fermenter. Compared with the six straight blade Rushton turbine, the power consumption of the new stirrer is lower at a given speed under conditions of no ventilation. The oxygen transference is poorer at the same agitation speed in the cultivation conditions and scales investigated, which confirms that the shear stress of the new stirrer is lower and the gas dispersion is weaker. The new stirrer was installed in a 5-L bioreactor and evaluated in submerged fermentation of the edible fungus Pleurotus ostreatus. The results showed that the maximum dry weight of mycelium is increased by 47 % and reached 7.47 g/L, and the maximum laccase activity is increased by 15 % up to 2,277 U/L. Glucose consumption was also found to be relatively faster. The power consumption is 2.8 % lower than that of the Rushton turbine.

AIMS: This study aimed to evaluate the different abilities of various Phellinus igniarius strains inhibiting quorum sensing (QS), to search for novel QS inhibitors from them and to analyze their inhibitory activity, with a view to their possible use in controlling infections. METHODS: The bioactive metabolites produced by P. igniarius cultures were tested for their abilities to inhibit QS-regulated behavior. All P. igniarius strains were cultured in potato-dextrose medium by large-scale submerged fermentation. The culture supernatant was condensed into 0.2-fold volumes by freeze drying. The condensed supernatant was sterilized by filtration through a 0.22-µm membrane filter and added to Chromobacterium violaceum CV026 cultures, which were used to monitor QS inhibition. Inhibitory activity was measured by quantifying violacein production using a microplate reader. RESULTS: The bioactive metabolites produced by 10 P. igniarius strains could inhibit violacein production, a QS-regulated behavior in C. violaceum. Furthermore, these strains could be roughly categorized into three groups on the basis of their inhibitory activities. CONCLUSIONS: P. igniarius strains can produce QS-inhibitory compounds and have different abilities to inhibit QS.

RESUMO

Tremella fuciformis produces several bioactive secondary metabolites including exopolysaccharides. Cultivation of the fungus was carried out in a three-stage process consisting of a 1.5-day cultivation with orbital shaking at 200 rpm, a 1.5-day cultivation with reciprocal shaking at 200 strokes, and a 1.5-day cultivation with orbital shaking at 200 rpm. Exopolysaccharide production and specific production rate reached 5.80 g L(-1) and 0.15 d(-1), respectively, which is an increase of 260% and 200% compared with the corresponding values for fermentations with orbital shaking only, and of 243% and 150% compared with the corresponding values for fermentations with reciprocal shaking only. The three-stage culture method is time-saving and easy to operate.

Extracts of Lingzhi or Reishi medicinal mushroom, Ganoderma lucidum, inhibited quorum sensing in Chromobacterium violaceum CV026. G. lucidum fruiting bodies were milled and extracted with ethyl acetate. The crude extract was dissolved in an appropriate concentration of methanol, sterilized by filtration through a 0.22-µm membrane filter, and added to Ch. Violaceum CV026 cultures, which were used as an indicator to monitor quorum sensing inhibition. Inhibitory activity was measured by quantifying violacein production using a microplate reader. Methanol-soluble compounds extracted from G. lucidum significantly inhibited quorum sensing-controlled behavior in Ch. Violaceum in a concentration-dependent manner. The results suggest that compounds in G. lucidum might be useful to control and handle detrimental infections caused by human, animal, and plant pathogens. Further studies are in progress in our lab to isolate the specific compounds from G. lucidum extract, evaluate them as quorum sensing inhibitors, and analyze their mechanism of action.
