The genus Vigna is an agronomically important taxon, with many of its species inhabiting a wide range of environments and offering numerous useful genes for the improvement of the cultivated types. The present study aimed to detect the genomic regions associated with yield-attributing traits by genome-wide association mapping. A diverse panel of 98 wild and cultivated Vigna accessions (acc.) belonging to 13 different species was evaluated for yield and related traits during the kharif season of 2017 and 2018. The panel was also genotyped using 92 cross-genera and cross-species simple sequence repeat markers to study the population genetic structure and useful market-trait associations. The PCA and trait correlation established relationships amongst the traits during both seasons while 100-seed weight (HSW) had a positive correlation with pod length (PL), and days to first flowering (DFF) with days to maturity (DM). The population genetic structure analysis grouped different acc. into three genetically distinct sub-populations with SP-1 comprising 34 acc., SP-2 (24 acc.), and SP-3 (33 acc.) and one admixture group (7 acc.). Mixed linear model analysis revealed an association of 13 markers, namely, VR018, VR039, VR022, CEDG033, GMES0337, MBSSR008, CEDG220, VM27, CP1225, CP08695, CEDG100, CEDG008, and CEDG096A with nine traits. Seven of the aforementioned markers, namely, VR018 for plant height (PH) and terminal leaflet length (TLL), VR022 for HSW and pod length (PL), CEDG033 for DFF and DM, MBSSR008 for DFF and DM, CP1225 for CC at 30 days (CC30), DFF and DM, CEDG100 for PH and terminal leaflet length (TLL), and CEDG096A for CC30 and chlorophyll content at 45 days were associated with multiple traits. The marker CEDG100, associated with HSW, PH, and TLL, is co-localized in gene-encoding histone-lysine N-methyltransferase ATX5. Similarly, VR22, associated with PL and HSW, is co-located in gene-encoding SHOOT GRAVITROPISM 5 in mungbean. These associations may be highly useful for marker-assisted genetic improvement of mungbean and other related Vigna species.

This study is one of the first systematic attempts to examine the possibility of a common treatment facility (CTF) to treat infectious healthcare waste (HCW) in Nepal. First, the survey was conducted in 14 healthcare facilities (HFs) ranging from health posts to large hospitals selected from 120 total HFs in Nepalgunj sub-metropolitan city (SMC), a rapidly urbanising city of Nepal to investigate the current practices of HCW management (HCWM) and to estimate the waste generation and characteristics in the different HFs. The result shows that the average unit waste generation rate for health posts, clinics, urban health centres, and hospitals was estimated at 1.397 kg day-1, 1.608 kg day-1, 0.178 kg day-1 and 1.818 kg bed-1 day-1, respectively. Of the total 1242 kg day-1 HCW generated in Nepalgunj SMC, 73% is infected in the current situation, but if fully sorted at source, only 32% of the waste will be infected. Based on these HCW generation data and fraction of infectious waste, including waste management practices, three different scenarios are proposed for the capacity assessment and designing implementation modality of the CTF to treat infectious waste from all HFs of Nepalgunj SMC as a case study where an integrated solid waste management facility including material recovery facility and sanitary landfill site for municipal solid waste management is already in operation. The different implementation analyses are discussed, and the best implementation arrangement has been recommended for the sustainability of the project. This approach can be replicated in other cities alone or regions with many neighbouring cities of Nepal and explores a workable solution for HCWM in the rapidly urbanising cities of developing countries to help them improve their condition.

Enrichment of carbon dioxide (CO2) in environment is a major factor for enhancement of global warming on Earth surface. Microalgal consortia play an important role in inhibiting the alarming fluxes of CO2 through sustainable mechanism of bioconversion of CO2 into biomass. In the present investigation, eight heterogeneous consortia of cyanobacteria and green algae such as MC1, MC2, MC3, MC4, MC5, MC6, MC7, and MC8 for the sustainable utilization of effective CO2 sequestration and biomass production were studied. Two factorial central composite designs (% CO2 and pH) were used for optimization of cellular morphology, growth, and development of consortia. The photosynthetic quantum yield of consortium MC8 was found to be maximum (0.61) in comparison with other consortia. The morphological and physiological behavior of the above consortium was analyzed under C, 5, 10, and 15% concentrations of CO2 resource capture in 250 mL BG-11+ medium. We have identified that 10% CO2 concentrated medium maximally promoted the cellular growth in terms of cell dimension, dried biomass, carbohydrate, and lipid contents in this consortium. As such, the elemental composition of carbon and carbon capturing capability was high at 10% CO2 concentration. However, further CO2 enrichment (15%) led to decline in growth and morphology of cell size as compared to control. The results indicate that the optimum CO2 enrichment in consortia exhibits potent commercial utilization for rapid biomass production and plays a distinguished role in global carbon sequestration and mitigation agent.

Nepal moved from unitary system with a three-level federal system of government. As federalism accelerates, the national health system can also speed up its own decentralization process, reduce disparities in access, and improve health outcomes. The turn towards federalism creates several potential opportunities for the national healthcare system. This is because decision making has been devolved to the federal, provincial and local governments, and so they can make decisions that are more representative of their localised health needs. The major challenge during the transition phase is to ensure that there are uninterrupted supplies of medical commodities and services. This requires scaling up the ability of local bodies to manage drug procurement and general logistics and adequate human resource in local healthcare centres. This article documents the efforts made so far in context of health sector federalization and synthesizes the progress and challenges to date and potential ways forward. This paper is written at a time while it is critical to review the federalism initiatives and develop way forward. As Nepal progress towards the federalized health system, we propose that the challenges inherent with the transition are critically analysed and mitigated while unfolding the potential of federal health system.

Hydroponic experiments were conducted to investigate impact of laser ablated copper oxide nanoparticles (CuO-NPs) on rice seedlings. The present work demonstrates that exposure of lower concentrations (5, 10, 20, and 50 µM) of CuO-NPs enhance growth (in terms of fresh and dry weight and length), of rice seedlings. However, at higher concentrations (100, 200, and 500 µM) of CuO-NPs, growth (in terms of length, fresh weight and dry weight) decreased significantly (P < 0.05). Further, photosynthetic pigments (total chlorophyll and carotenoids) and protein contents were also found to be in accordance with the results of growth. This had occurred due to enhanced level of CuO-NPs accumulation at higher doses which also enhanced the level of oxidative stress markers such as hydrogen peroxide (H2O2) and malondialdehyde (MDA). Chlorophyll a fluorescence parameters (Fv/Fm and qP and except NPQ) and amount of some minerals (Ca, Mg, Na, and K) increased at lower concentrations of CuO-NPs. In contrast, the levels of Fv/Fm and qP were significantly (P < 0.05) reduced at higher concentration of CuO-NPs, which might be due to enhanced accumulation of Cu and oxidative stresses markers. Our results showed that lower dosages of pulsed laser ablated CuO-NPs (5, 10, 20, and 50 µM) might be beneficial for growth and development of rice seedlings.

A set of experiments was conducted to provide significant insights of micro-algal consortia regarding chromium adsorption. Four monocultures; Scenedesmus dimorphus, Chlorella sp., Oscillatoria sp., and Lyngbya sp., and their synthetic consortia were evaluated initially for chromium bio-adsorption at four different regimes of hexavalent chromium i.e. 0.5, 1.0, 3.0 and 5.0 ppm. Based on findings, only 1.0 and 5.0 ppm were considered for future experiments. Consequently, three different types of monoculture and consortia cells namely; live cells, heat-killed cells, and pre-treated cells were prepared to enhance their adsorption potential. Maximal adsorption of 112% was obtained at the dose of 1.0 ppm with 0.1% SDS pre-treated consortia cells over live consortia cells. In support, atomic absorption spectroscopy, laser induced breakdown spectroscopy, pulse amplitude modulated chlorophyll fluorescence, and scanning electron microscopy were performed to assess the structural and functional changes within consortia and their utilization in mitigation of elevated chromium levels.
