RESUMO

The paper investigates the specific features of chronorhythmologic changes of the ion-regulating function of the kidneys under the hypofunction of the pineal gland. For the study of hypofunction of the pineal gland, animals were maintained under constant light stimulation during 7 days. It has been established that the hypofunction epiphysis point out disturbances of the phasic structure of the ion-regulating function in relation to chronograms of the intact group of rats. In terms of physiological pinealectomy observed in- crease in the excretion of sodium ions (6,11±0,81 mmol/h) and the concentration of this cation in the blood plasma remained elevated around the clock. The obtained results indicate significant changes of integral characteristics of the rhythms of electrolyte metabolism and the important role of the pineal gland in ensuring physiological homeostasis.

The dynamics of the balance of indices of pro- and p53- Bcl-2 anti-apoptotic processes in the hippocampus of rats with experimental diabetes mellitus (DM) complicated by incomplete global cerebral ischemia-reperfusion was investigated. It is shown that p53 proapoptotic processes in animals without diabetes after 20 minutes of ischemia/l hour reperfusion in all fields of the hippocampus are activated in the background of increasing Bcl-2 antiapoptotic processes in the fields CAl, CA2, CA4 and depression of it - in the CA3 field. In the early postischemic period in rats with DM activity of the p53-proapoptotic processes in fields CAl, CA3, CA4 significantly exceeds that in non-diabetic rats (area of p53- IRM increases on 110, 60 and 27 %), and was significantly lower than that detected in CA2 field. On the 12th day of post-ischemic period, activation of apoptosis in field CAl occurs in the background of inert antiapoptotic processes, in animals without diabetes, as well as in diabetic rats, but the indicators characterizing of apoptotic activity in rats with diabetes were higher (specific contents of p53 protein and area ofp53 -IRM increases on 38 and 43 %). During this period, in the CA2 region of the non-diabetic animals, some depression of the antiapoptotic processes with a slight predominance of proapoptotic processes was detected. In the field of CA3 region of rats without diabetes, the retention of activity of proapoptotic processes and the deepening in the dynamics of depression of antiapoptotic processes were showed. In rats with DM, the oppression of both mechanisms with a significant depression of antiapoptotic processes was observed. On the 12th day of experiment in the field CA4, the most balanced relationship were detected between the studied of the processes due to their parallel and unidirectional changes both in the rats without diabetes as well as with DM. The results point on the modifying effect ofDM on susceptibility ofhippocampal fields to ischemic-reperfusion injury.

RESUMO

The influence of bilateral 20-minute carotid ischemia with reperfusion of various duration upon the activity of apoptotic processes in the neurons and glia cells of the cerebral cortex in the temporal lobe of rats has been studied. Bilateral carotid ischemia with one-hour reperjusion has been indicated to increase the location density of p53+-neurocytes in this region of the cerebral cortex, increases percentage and density of p53+-nerve cells and reduces the percentage and density of p53+-glia cells on the 12th day of the post-ischemic period. Ischemic-reperfusion lesion of the brain increases the concentration of p53 protein in glia cells of the cerebral cortex in the temporal region in the early ischemic-reperfusion period and reduces it in nerve and glia cells in the late one.

Janus kinases Jak1 and Tyk2 play an important role in urokinase-type plasminogen activator (uPA)-dependent signaling. We have recently demonstrated that both kinases are associated with the uPA receptor (uPAR) and mediate uPA-induced activation of signal transducers and activators of transcription (Stat1, Stat2, and Stat4) in human vascular smooth muscle cells (VSMC). Janus kinases are not only required for Stat activation but may also interfere with other intracellular signaling pathways. Here we report that in VSMC, Tyk2 interacts with a downstream signaling cascade involving phosphatidylinositol 3-kinase (PI3-K). We demonstrate that uPA induces PI3-K activation, which is abolished in VSMC expressing the dominant negative form of Tyk2. The regulatory subunit p85 of PI3-K co-immunoprecipitates with Tyk2 but not with Jak1, Jak2, or Jak3, and uPA stimulation increases the PI3-K activity in Tyk2 immunoprecipitates. Tyk2 directly binds to either of the two Src homology 2(SH2)p85 domains in a uPA-dependent fashion. We provide evidence that the Tyk2-mediated PI3-K activation in response to uPA is required for VSMC migration. Thus, two unrelated structurally distinct specific inhibitors of PI3-K, wortmannin and LY294002, prevent VSMC migration induced by uPA. No migratory effect of uPA was observed in VSMC expressing the dominant negative form of Tyk2. Our results underscore the versatile function of Tyk2 in uPA-related intracellular signaling and indicate that PI3-K plays a selective role in the regulation of VSMC migration.

RESUMO

The effect of GABA receptors agonists on the stress-induced beta-endorphin levels in the preoptic area and mediobasal hypothalamus of the intact and prenatally stressed male albino rats was studied. It has been found out that stimulation of GABAa-receptor complex by means of the muscimol leads to increasing of beta-endorphin levels in the preoptic area and mediobasal hypothalamus of the control animals. GABAb receptor activation by means of the baclofen decreases opioids level in the mediobasal hypothalamus. Prenatal stress eliminates stimulant effect of the muscimol on beta-endorphin levels in the investigated brain structures and leads to the opioid level decreasing after baclofen influence in preoptic area.

RESUMO

Nonviral transfection is one of the modern methods for the incorporation of foreign genes into cells. This process involves uptake of foreign genetic material by the cell and further trafficking through the cytoplasm to the nucleus. Elucidation of cytoplasmic pathways of transfection complexes can be useful to improve already existing gene delivery systems or to establish new systems. To monitor transfection complexes in the cell during transfection, we elaborated a method for the visualization of transfection complexes by introducing digoxigenin-labelled nucleotides into foreign DNA followed by detection of digoxigenin label with the use of antibodies directed against digoxigenin. This procedure allowed the visualization of DNA in transfection complexes and to monitor these complexes in cells during transfection.

Threat of accident development at the chemical industry objects, as well as tasks of medical support in chemical weapon destruction necessitate improvement of the toxicant biological indication methods, creation of protective-decontaminating prescriptions to protect open areas of skin, and also development of etiotropic and pathogenetic therapy agents for poisonings The Scientific-Research-Test Center (Medicobiologic Protection) studies of every of the above trends have allowed to create and introduce into table medical support of the RF Armed Forces some new indication-means samples, antichemical packets, antidotes and means of pathogenetic therapy for convulsive and bronchospastic syndromes, to motivate use of pharmacological agents to stop toxic pulmonary edema and cardio-vascular failure.

RESUMO

cDNA with a high degree of homology to the rac1 gene of the Ras-like protein from the HL-60 cell line was isolated from a bovine retina cDNA library. The homologies of the nucleotide and deduced amino acid sequences were 90 and 98%, respectively. The product of cDNA expression in E. coli cells purified by chromatography displayed GTP-binding and GTPase activities. The data suggest that the Rac1 protein is one of the low-molecular GTP-binding proteins in photoreceptor membranes of vertebrates.

RESUMO

Toxicity of water treated by two disinfectants, AOKh-K and gaseous chlorine, was studied by biotesting on Daphnia magna, Tetrahymena pyriformis (TP) and renal epithelium of porcine embryo (REPE). The study showed that AOKh-K preparation is nontoxic and chlorine toxic for TP and REPE in concentrations 2 and 3 mg/l.

RESUMO

The article studies the informative value of test objects for the evaluation of potable water which contains toxic substances. A number of test objects is proposed by the authors to estimate the chemical harmlessness of water: tetrachimens, Daphnia and peroxide ferment system. The data obtained with the help of test-objects possess a sufficient selective sensibility as far as maximum allowable concentration of various toxic substances is concerned. The authors stress that these methods of biological testing may complement or precede the traditional methods of water evaluation.
