RESUMO

The aim of our investigations was to describe the morphological changes in the epithelium of the ampulla region of the ewe uterine tube ampulla on days 2, 7, 17, 25 and 34 post partum. Shortly after the parturition the epithelium was low. Ciliated cells were dark with few kinocilia and secretory cells were light in colour and contained no secretory granules. On days 17 and 25 post partum the ciliated cells were high. They had well-developed granular endoplasmic reticulum, ribosomes, and developing basal corpuscles and kinocilia. Secretory cells contained a well-developed Golgi complex, granular endoplasmic reticulum and numerous secretory granules of varied size, shape, and density. On day 34 post partum, the ciliated cells had numerous kinocilia on their apical surface. Secretory cells released their product into the oviduct lumen through pores or high protrusions of cells detached, decomposed, and became the part of the secretion.

RESUMO

The effect of hypodynamia during 9, 30, 60 and 90 days on the striated muscle m. gastrocnemius in Japanese quails was studied. We have found different developed dystrophic changes of mitochondria, myofibrils and sarcotubular system. Changes of the studied tissue caused by hypodynamia in the first two groups (9 and 30 days hypodynamy) are the reaction to stress and developing situation. Gradually Japanese quails have come up to the influence of the hypodynamia (60 days hypodynamia). In the last experimental group deposit changes were observed (90 days hypodynamia). In comparison with control group we have found perturbations considered as non-specific, short-term and reversible.

Sertoli cells produce special microenvironment for developing germ cells; therefore it is assumed that they play primary role in the onset and control of spermatogenesis. In this connection we extended our previous study on the ultrastructure of Sertoli cells in different domestic and wild animals with special regard to nucleolus. Sertoli cells of domestic and wild ruminants possess the typical vesicular nucleolus except for fallow deer, in this species no vesicular nucleolus occurs in Sertoli cells even during the rut. In roe.buck, another wild ruminant with seasonal spermatogenesis, cyclic changes were found in the nucleolus of Sertoli cells. If no spermatogenesis is present, the Sertoli cells have a reticular nucleolus. Membranous vesicles appear in the nucleolus of Sertoli cells of roe-buck at the onset of spermatogenesis 1-2 months before rut. In domestic ruminants with continuous spermatogenesis the vesicular nucleolus in Sertoli cells is present permanently. During postnatal development of bull and ram the vesicular nucleolus appears in Sertoli cells just before the onset of spermatogenesis. In experimental cryptorchidism of bulls a vesicular nucleolus is found in the Sertoli cells. Our observations and experiments support a hypothesis that Sertoli cells have primary role at the onset and the maintenance of spermatogenesis.

With the aid of a transmission electron microscope we studied the ultrastructure of frozen and thawed ram sperm. The cryoprotective agents do not completely prevent the occurrence of structural changes in spermatozoa. The sperm membrane system is affected. The greatest and most frequent damage is to the acrosome, the cell membrane and mitochondria are less injured. We observed regional differences in the damage to the cell membrane. It is most damaged above the acrosome, least in the postacrosomal area and in the principal piece of the tail. We found considerable variability in the changes among individual spermatozoa from the same ejaculate. The changes are not in the nature of mechanical damage as a result of the occurrence of ice microcrystals. We therefore think that the occurrence of structural changes is dependent on damage at molecular level and consists of a change in the physical-chemical properties of membranes.

The neurons of the pars caudalis nuclei tuberomammillaris (pc-NTM) were studed light-microscopically and electron-microscopically in sheep and rams of Merino breed. In our study we observed: In the regarded neural nucleus, there is the majority of the great neurons (up to 60 microns in diameter) rich in the NISSL's bodies. When stained with the cresyl violet, the NISSL's substance is apparently stored mainly in peripheral area of the cell body and in the distant parts of numerous protoplasmic processes, what evokes an impression of the "jagged" surface of these cells. After staining with paraldehyde fuchsin, we found purple coloured lumps of irregular shape stored outside the cell bodies, in the neuropil. The less extended cells, usually with lower content of NISSL bodies, are in pc-NTM less frequent. In the electron-microscopic study we identified 3 types of neurons: Cells rich in rough endoplasmic reticulum; "light" cells, "dark" cells. The cells of the 1st type were the most frequent ones. Cisterns of rough endoplasmic reticulum in the 1st type of cells are often dilated. The protoplasmic processes of these cells are frequently stepped over by flat tubuli of endoplasmic reticulum. The 2nd type of cells is characterized by the light cytoplasmic matrix, low quantity of endoplasmic reticulum and frequent occurrence of lipofuscin bodies. The 3rd type of cells are characterized by the high density of cytoplasmic matrix, well developed GOLGI complex, and very broad cisterns of endoplasmic reticulum, forming a labyrinth, and it is bound to a broad perinuclear space.
