RESUMO

Single hole transport and spin detection is achievable in standard p-type silicon transistors owing to the strong orbital quantization of disorder based quantum dots. Through the use of the well acting as a pseudo-gate, we discover the formation of a double-quantum dot system exhibiting Pauli spin-blockade and investigate the magnetic field dependence of the leakage current. This enables attributes that are key to hole spin state control to be determined, where we calculate a tunnel couplingtcof 57µeV and a short spin-orbit lengthlSOof 250 nm. The demonstrated strong spin-orbit interaction at the interface when using disorder based quantum dots supports electric-field mediated control. These results provide further motivation that a readily scalable platform such as industry standard silicon technology can be used to investigate interactions which are useful for quantum information processing.

Manipulation of carrier densities at the single electron level is inevitable in modern silicon based transistors to ensure reliable circuit operation with sufficiently low threshold-voltage variations. However, previous methods required statistical analysis to identify devices which exhibit random telegraph signals (RTSs), caused by trapping and de-trapping of a single electron. Here, we show that we can deliberately introduce an RTS in a silicon nanowire transistor, with its probability distribution perfectly controlled by a triple gate. A quantum dot (QD) was electrically defined in a silicon nanowire transistor with a triple gate, and an RTS was observed when two barrier gates were negatively biased to form potential barriers, while the entire nanowire channel was weakly inverted by the top gate. We could successfully derive the energy levels in the QD from the quantum mechanical probability distributions and the average lifetimes of RTSs. This study reveals that we can manipulate individual electrons electrically, even at room temperature, and paves the way to use a charged state for quantum technologies in the future.

The Random Telegraph Noise (RTN) in an advanced Metal-Oxide-Semiconductor Field-Effect Transistor (MOSFET) is considered to be triggered by just one electron or one hole, and its importance is recognised upon the aggressive scaling. However, the detailed nature of the charge trap remains to be investigated due to the difficulty to find out the exact device, which shows the RTN feature over statistical variations. Here, we show the RTN can be observed from virtually all devices at low temperatures, and provide a methodology to enable a systematic way to identify the bias conditions to observe the RTN. We found that the RTN was observed at the verge of the Coulomb blockade in the stability diagram of a parasitic Single-Hole-Transistor (SHT), and we have successfully identified the locations of the charge traps by measuring the bias dependence of the RTN.

Green tea ('Sencha'), made from the leaves of Camellia sinensis, is the most well-researched antioxidant beverage. The major source of its antioxidant activity is polyphenols, consisting mainly of catechins (flavan-3-ols). However, little is known about the physiological effects of green tea aroma, which lacks catechins. In the present study, we performed inhalation experiments with green tea aroma to evaluate its antioxidant activity in mice. As a result, the urinary 8-hydroxydeoxyguanosine levels were significantly decreased in comparison with those of the non-treated group, and the serum antioxidant capacity was significantly increased by the inhalation administration of green tea aroma. Furthermore, the increase in the urinary 8-hydroxydeoxyguanosine levels due to whole-body X-ray irradiation was significantly suppressed by the inhalation of green tea aroma. This is the first study to show the antioxidant activity of green tea aroma in vivo.

Quasi-phase-matched (QPM) LiNbO3 devices having a multiperiod-periodically-poled structure, which we call multiperiod-QPM LiNbO3 devices, are capable of shifting the idler waveband during waveband conversion via cascaded difference-frequency generation (cascaded DFG). However, these multiperiod-QPM devices have a problem that they produce extra ripples between QPM peaks in the phase-matching curve. These ripples cause crosstalk between wavebands arising from sum-frequency generation (SFG) between the signal and idler wavebands and subsequent DFG between the SFG wavelength and the signal waveband. To decrease the size of the ripples and thus that of the crosstalk, an apodized multiperiod-QPM device is developed. In demonstrating waveband conversion for low crosstalk with this device, we measure the dependence of the idler power, the crosstalk power, and their ratio on the signal power. This measurement shows that it agrees well with theoretical prediction and that the obtained feature of crosstalk reduction is kept even for decreased signal power.

Yeast RNA is a good source of nucleotide seasoning, and more than half of yeast RNA consists of ribosomal RNA (rRNA). Previously, we reported the development of a Saccharomyces cerevisiae strain displaying a 1.4- to 2.3-times higher RNA content than the wild-type strain through the isolation of dominant suppressors (designated SupA to SupG strains) from a Δrrn10 disruptant showing decreased rRNA transcription. In the present study, the cloning of one of the genes responsible for the suppression was attempted using a genomic library from the SupD strain. NOP15, a gene involved in ribosome biogenesis, was found to be responsible for suppressing the growth defect of the Δrrn10 disruptant. The isolated NOP15 allele (designated NOP15(T-279C)) possessed a single T to C substitution at nucleotide position-279 of NOP15. The transcription level of NOP15(T-279C) in the originally isolated SupD strain was 2-fold higher than that in the Δrrn10 disruptant. Furthermore, a dose-dependent relationship between the transcription level of NOP15 and total amount of RNA in the Δrrn10 disruptant was observed: the enhanced transcription due to the NOP15(T-279C) allele is involved in the suppression mechanisms in the SupD strain. Introduction of the NOP15(T-279C) allele into the wild-type strain increased the total RNA content by 1.4-fold. These results indicate that the transcription level of NOP15 is an important determinant of the productivity of RNA and that its increased transcription provides an effective approach to obtain higher RNA yields in yeast.

We report phase-transparent waveband conversion with polarization insensitivity based on second harmonic (SH) wave pumped difference frequency generation (DFG) using multiple-quasi-phase-matched LiNbO(3) (QPM-LN) waveguides. Flexible waveband conversion is demonstrated over the entire C-band using a tunable DFB-LD array (TLA) as a pump source for a multiple-QPM-LN waveguide. The penalty free waveband conversion of 43 Gb/s return-to-zero differential quadrature phase-shift-keying (RZ-DQPSK) waveband signals is successfully achieved.

We developed an apodized multiple quasi-phase-matched (QPM) LiNbO(3) device to reduce the cross talk that occurs during waveband conversion. Since we found that the cross talk arises from ripples between multiple QPM peaks in the phase-matching curve, we reduced the amplitude of the ripples by employing the apodized structure. Using this device, we succeeded in demonstrating waveband conversion with low cross talk.

Accumulated evidences indicate that reactive oxygen species (ROS) are involved in the pathophysiology of aging process. Antioxidants are believed to play an important role in the defense system to counteract ROS in the body. While excess hydrophilic antioxidants can be excreted easily in urine, lipophilic antioxidants can penetrate into blood lipoproteins and cell membranes, and may maintain long and high bioavailability. These lipophilic antioxidants are thus expected to contribute greatly to the prevention of age-related diseases. Oils extracted from plant seeds are known to contain various lipophilic antioxidants such as vitamin E (alpha-tocopherol) and carotenoids. They are known to not only decrease serum low-density-lipoprotein (LDL) level, but also prevent oxidation of LDL. In addition to vitamin E (alpha-tocopherol) and carotenoids, other lipophilic antioxidants such as gamma-oryzanol and sesaminol (from sesamolin) are in rice bran and sesame, respectively. They are sometimes called "vitamin-like food factors" or "biofactors." Although there are several methods for measuring the total antioxidant activities for various plant extracts, most of these methods are designed for hydrophilic antioxidants, and not for lipophilic antioxidants present in various plant seed oils. In this report, we present an assay method for the total potency of antioxidants that are soluble in oil (PAO-SO) utilizing bathocuproine (BC) as a chromogen. BC-based antioxidant activity assay shows good linearity (r(2) = 0.9986), good reproducibility (CV < 10%), and good recovery (86-91%) when dl-alpha-tocopherol, for example, is added to sesame oil. Total antioxidant activity of rape-seed oil, olive oil, and sesame oil could also be successfully measured.

The measurement of absorption spectra using angle-dependent terahertz (THz) time-domain spectroscopy for amino acid single crystals of l-cysteine and l-histidine is reported for the first time. Linearly polarized THz radiation enables us to observe angle-dependent far-infrared absorption spectra of amino acid single crystals and determine the direction of the oscillating dipole of the molecules in the 20-100 cm(-1) range. By comparing the THz spectra of a single crystal and powder, we found that there was a clear hydrogen-bond peak in the crystal spectrum as a result of the larger hydrogen-bond network. The low-temperature THz spectra of amino acid microcrystals showed more intermolecular vibrational modes than those measured at room temperature. An ab initio frequency calculation of a single amino acid molecule was used to predict the intramolecular vibrational modes. The validity of the calculation models was confirmed by comparing the results with experimentally obtained data in the Raman spectral region.

The quantitative analysis of amino acids by terahertz (THz) time-domain absorption spectroscopy is demonstrated. The optical densities of the amino acids were found to be linearly proportional to the concentration. The molar absorption coefficients of L-glutamic acid (L-Glu), L-glutamic acid sodium salt (Na-L-Glu), L-glutamic acid hydrochloric salt (HCl-L-Glu), L-cysteine (L-Cys), and L-histidine (L-His) were calculated by averaging the THz spectra of the amino acids at several different concentrations in approximately the 0.2-1.0 mol L(-1) range. The concentrations of L-Glu, L-Cys, and L-His mixed samples were successfully calculated with errors of less than 11% and 20% when their concentrations were higher than 0.45 and 0.22 mol L(-1), respectively, by using the obtained molar absorption coefficient.

We propose a THz notch filter that uses the absorption of the intermolecular hydrogen bonds in a molecular crystal such as sucrose. Terahertz (THz) time-domain spectroscopy was used to investigate the absorption characteristics of a rotationally oriented sucrose single crystal in the 0.3 - 3.0 THz frequency range. The crystal was set so that the (100) face (cleavage) was normal to the THz propagation direction. The two lowest frequency intermolecular hydrogen bonding bands clearly exist at room temperature and at 1.45 and 1.64 THz when the b-axis of the crystal is parallel and perpendicular to the THz polarization, respectively. In contrast, they disappear when the b-axis is reversed. This absorption feature means that it would be possible to utilize a sucrose crystal as a notch filter with a 1.45-THz band center for the 0.1-1.7 THz range and with a 1.64-THz band center for the 0.1-1.9 THz range. When the crystal is rotated, the transmitted intensities of the frequency components near the absorption bands alternate as a sine function against angle. The spectroscopic properties of the sucrose filter were confirmed by continuous THz wave imaging.

We previously reported that intracellular free cholesterol at physiological concentrations regulates the activity of neutral cholesterol esterase (N-CEase) in macrophages. The objective of the present study is to investigate whether the regulation of N-CEase by cholesterol is generally observed in other types of cells such as adipocytes with high activity of hormone-sensitive lipase (HSL), the same gene product as N-CEase. 3T3-L1 adipocytes were cultured with and without cholesterol (1-30 microg/mL) or 25-hydroxycholesterol (0.1-10 microg/mL), and changes in the N-CEase activity, expression of HSL mRNA, and protein were examined. Incubation (24 h) of cells with cholesterol did not change N-CEase activity, but incubation with 25-hydroxycholesterol decreased the activity in a concentration-dependent manner by 24 (24 h) and 54% (36 h). Quantitative reverse transcription-PCR indicated that 25-hydroxycholesterol (10 microg/mL) did not influence expression of HSL mRNA. However, Western blot analysis showed that this sterol reduced HSL protein by 72 (24 h) and by 93% (36 h), respectively. It was concluded that sterol-mediated regulation of HSL/N-CEase occurs not only in macrophages but also in adipocytes, and regulation appears to occur not at a transcriptional level but by a post-transcriptional process. Sterol-mediated proteolysis may be involved in the loss of HSL protein.

Malignant stroke-prone spontaneously hypertensive rats (M-SHRSP) develop hypertension and stroke at earlier ages than do nonmalignant SHRSP. Our previous findings suggested that reactive oxygen species were involved in the development of stroke in this strain. Nitric oxide (NO) which is more released at ischemia, might play a crucial role in stroke development by producing peroxynitrite, a neurotoxic substance. This study investigated whether the development of cerebrovascular lesion in M-SHRSP could be assessed by the fluctuation of serum NO(x) concentration, and whether peroxynitrite is associated with brain damage. Serum NO(x) levels were examined using an automated NO detector. Stroke-onset was temporally assessed according to a known method: changes in body weight, water intake, and neurologic symptoms. Cerebral lesions were confirmed by magnetic resonance imaging (MRI), and Evans blue extravasation at autopsy. MRI taken just after estimated stroke onset disclosed brain lesions. The baseline serum NO(x) level remained at 15-18 micromol/l, but the level gradually increased prior to stroke, and significantly at stroke onset. A marked rise in serum NO(x) occurred subsequently at poststroke. Immunohistochemical staining of nitrotyrosine, a peroxynitrite marker, was detected around vessels, neuronal cells and parenchyma in cerebral lesions. Stroke occurred in 50% of male M-SHRSP at 80 days of age. In conclusion, this study provides the first evidence for fluctuation of serum NO(x) at the onset of spontaneous stroke accompanying the appearance of peroxynitrite in brain lesions. Monitoring serum NO(x) would serve to assess the development of brain lesions at least in spontaneous stroke model.
