RESUMO

We present a reverse microemulsion synthesis procedure for incorporating methylene blue (MB), a known FDA-approved type-II red-absorbing photosensitizer and 1O2 generator, into the matrix of hydrophobic-core/hydrophilic-shell SiO2 nanoparticles. Different synthesis conditions were explored with the aim of controlling the entrapped-dye aggregation at high dye loadings in the hydrophobic protective core; minimizing dye aggregation ensured highly efficient photoactive nanoentities for 1O2 production. Monitoring the synthesis in real time using UV-vis absorption allowed tracking of the dye aggregation process. In particular, silica nanoparticles (MB@SiO2 NPs) of ∼50 nm diameter size with a high local entrapped-MB concentration (∼10-2 M, 1000 MB molecules per NP) and a moderate proportion of dye aggregation were obtained. The as-prepared MB@SiO2 NPs showed a high singlet oxygen photogeneration efficiency (ΦΔ = 0.30 ± 0.05), and they can be also considered as red fluorescent probes (ΦF ∼ 0.02, λmax ∼ 650 nm). The distinctive photophysical and photochemical characteristics of the synthesized NPs reveal that the reverse microemulsion synthesis procedure offers an interesting strategy for the development of complex theranostic nano-objects for photodynamic therapy.

Anisotropic gold nanoparticles offer potential applications due to their functionalities and shape-dependent properties. Reshaping noble metal nanoparticles is an interesting field with optical, surface-enhanced Raman spectroscopy, catalytic applications and potential application as a photothermic therapy. This work comprises a structural study on gold nano bipyramids (Au NBPs) and nanodumbbells, and the evolution of Au NBPs capped with cetyltrimethylammonium bromide and dodecanethiol through an in situ and ex situ heating process in high vacuum. Also, we study the reshaping of Au NBPs by the addition of Pt to study the surface modification and the strain generated on a single particle by geometric phase analysis.

We developed a highly sensitive silicon platform, suitable to assess the molecular organization of protein samples. Prototype platforms were obtained using different electrochemical protocols for the electrodeposition of Ag-nanoparticles onto the hydrogenated silicon surface. A platform with high Surface Enhanced Raman Scattering efficiency was selected based on the surface coverage and the number density of particles size distribution. The performance of the platform was determined by studying the interaction of Myristoylated Alanine-Rich C Kinase Substrate (MARCKS) protein with the substrate according to its molecular organization. The chemical and structural characteristics of MARCKS molecules were examined under two configurations: i) a disordered distribution given by a MARCKS solution drop deposited onto the platform and, ii) a compact monolayer transferred to the platform by the Langmuir-Blodgett method. Raman spectra show vibrational bands of Phenylalanine and Lysine residues specific for the protein effector domain, and evidence the presence of alpha helix structure in both configurations. Moreover, we distinguished the supramolecular order between the compact monolayer and random molecular distribution. The platforms containing Ag-nanoparticles are suitable for studies of protein structure and interactions, advancing a methodological strategy for our long term goal, which is to explore the interaction of proteins with model membranes.
