Abstract This study analyzed the antifungal activity of phytoconstituents from linalool on Candida spp. strains, in vitro, isolated from patients with clinical diagnoses of oral candidiasis associated with the use of a dental prosthesis. Biological samples were collected from 12 patients using complete dentures or removable partial dentures and who presented mucous with diffuse erythematous or stippled features, indicating a clinical diagnosis of candidiasis. To identify fungal colonies of the genus Candida, samples were plated onto CHROMagar Candida®. The antifungal activity of linalool, a monoterpene unsaturated constituent of basil oil, was performed using the broth microdilution technique. Then, the minimum inhibitory concentration (MIC), the two subsequent stronger concentrations and the positive controls were subcultured on Sabouraud Dextrose Agar plates to determine the minimum fungicidal concentration (MFC). The experiments were performed in triplicate and nystatin was used as a positive control in all tests. Diagnoses of oral candidiasis were verified in eight patients (66.6%) and the most prevalent fungal species was Candida albicans (37.5%), followed by Candida krusei (25.0%); and Candida tropicalis (4.2%). The best antifungal activity of linalool was observed on Candida tropicalis (MIC = 500 mg/mL), followed by Candida albicans (MIC = 1.000 mg/mL), and Candida krusei (MIC = 2.000 mg/mL).Under the study conditions and based on the results obtained, it can be concluded that the Candida strains tested were susceptible to linalool.


Resumo Este estudo analisou a atividade antifúngica do fitoconstituinte linalol em cepas de Candida ssp, in vitro, isolados de pacientes com o diagnóstico clínico de candidíase oral associado ao uso de prótese dentária. As amostras biológicas foram coletadas de 12 pacientes portadores de próteses totais ou próteses parciais removíveis e que apresentavam características de mucosa eritematosa difusa ou pontilhadas, indicando um diagnóstico clínico de candidíase. Para identificar colônias de fungos do gênero Candida, as amostras foram semeadas em CHROMagar Candida®. A atividade antifúngica do linalol, um componente insaturado monoterpene de óleo de manjericão, foi realizada através da técnica de microdiluição em caldo. Em seguida, a concentração inibitória mínima (MIC), as duas concentrações consecutivas mais fortes e os controles positivos foram subcultivados em placas de Agar Sabouraud Dextrose para determinar a concentração fungicida mínima (MFC). Os experimentos foram realizados em triplicata e a nistatina foi usada como controle positivo em todos os testes. O diagnóstico de candidíase oral foi comprovado em oito pacientes (66,6%) e as espécies de fungos mais prevalentes foram Candida albicans (37,5%), seguido por Candida krusei (25,0%); e Candida tropicalis (4,2%). A melhor atividade antifúngica do linalol foi observada em Candida tropicalis (MIC = 500 mg/ml), seguido por Candida albicans (CIM = 1,000 mg/mL), e Candida krusei (CIM = 2,000 mg/mL). Sob as condições do estudo e com base nos resultados obtidos, pode-se concluir que as estirpes de Candida testadas foram susceptíveis a linalol.

OBJECTIVE: To determine the sensitivity and specificity of a new clinical test for the diagnosis of injuries to the posterolateral corner of the knee by using magnetic resonance imaging (MRI) as the reference standard. DESIGN: Diagnostic accuracy study. SETTING: A tertiary care teaching hospital. PARTICIPANTS: Twelve subjects with chronic instability of the knee and posterolateral corner injury diagnosed by intraoperative findings and 9 subjects without posterolateral corner injury (used as controls). INTERVENTIONS: Injured and uninjured knees were assessed blindly by 2 examiners using a new clinical diagnostic test (the frog-leg test) and the classic varus stress test. MAIN OUTCOME MEASURES: Injuries to the posterolateral corner of the knee identified by the 2 clinical tests. Findings were compared and examined for reproducibility, and kappa statistic was used to assess interobserver agreement. Test results were compared with those of MRI and intraoperative findings to determine diagnostic accuracy. RESULTS: The frog-leg test showed high interobserver agreement (kappa, 0.86), with a high rate (83%-100%) of agreement with the intraoperative diagnosis. The frog-leg test had high sensitivity (91.7%) and specificity (94.5%) for detecting posterolateral corner injuries. The sensitivity of the varus stress test increased from 83.3% to 90.0% when combined with the frog-leg test. CONCLUSIONS: Examiners were able to identify posterolateral corner injuries and differentiate injured from uninjured knees using the frog-leg test, which could potentially be used as an ancillary tool to the varus stress test in diagnosing injuries to the posterolateral corner of the knee. Larger studies are needed to confirm our findings.

RESUMO

The rediscovery of the enigmatic subterranean characiform Stygichthys typhlops is reported almost a half-century after the collection of the holotype, the only specimen previously known. Thirty-four specimens were collected in two shallow hand-dug wells at the region of the type locality, c. 13 km south-west of the town of Jaíba, Minas Gerais, Brazil. These specimens provide new information on the morphology of this species, and for the first time on its life history. The conservation status of S. typhlops is discussed. The species is severely threatened by habitat loss caused by exploitation of the aquifer.

The meniscotibial ligaments (LMT) of humans are capsular fibers with proximal origin in the lateral border of the menisci and distal insertion in the lateral border of the tibial condyle, also called coronary ligament of the knee. However, few studies describe the presence, course and anatomical variations of this ligament. Our objective was to evaluate the LMT through quantitative and descriptive methods. Eighty-five knees were examined(42 right knees [RK] and 43 left knees [LK]), dissected, evaluated, catalogued and photographed by four examiners. Twenty knees (23.53%) presented LMT, 6 (30%) in RK and 14 (70%) in LK. In two RKs, the LMT presented proximal insertion in the medial menisci and distal insertion in medial tibial condyle; in the other ones, the LMT presented proximal insertion in the lateral menisci and distal insertion in the lateral tibial condyle. In the LK, four LMT presented proximal insertion in the medial menisci and distal insertion in the medial tibial condyle, while the other ones presented proximal insertion in the lateral menisci and distal insertionin the lateral tibial condyle. One LMT in the LK presented proximal and distal insertion in both menisci and tibial condyle. The analysis of human corpses in our study suggests that the presence of the LMT is much more common in the LK than in the RK, with more frequent insertion in the proximal region of the lateral menisci and distal insertion in the lateral tibial condyle.

Trichomycterus itacarambiensis is a troglobitic (cave-restricted) catfish found in the Olhos d'Agua Cave, Itacarambi Co., Minas Gerais, south-eastern Brazil. The cave is subject to a pronounced seasonality, and is flooded during part of the rainy season. The pigmentation degree is variable in T. itacarambiensis, a third of the population being true albinos; the eyes vary from normal to externally invisible. As expected for a cave-dwelling fish, T. itacarambiensis is a carnivorous generalist, which preys mainly on autochthonous insects and earthworms. It is a chemically oriented predator of bottom and surface animals, using foraging tactics also used by other cave catfishes, such as the pimelodids Pimelodella kronei and Imparfinis sp. However, bottom feeding seems to be more important for T. itacarambiensis than for the latter. There is a severe feeding stress during the dry season, when few individuals are able to get food. Reproduction is seasonal, with increased reproductive activity at the end of the rainy period, as recorded for Imparfinis sp., also a species living in a semi-arid region with a well-defined dry season. It is estimated that up to 50% of the female T. itacarambiensis can reproduce every year, a high proportion for a troglobitic fish species. At least during the study period, pigmented individuals reproduced more frequently than the albinos.

Blind pimelodid catfishes Pimelodella kronei from a karstic area in Southeastern Brazil, were paired in the laboratory to study the agonistic behaviour. P. kronei, a recent troglobite, displays an intense and complex aggressive behaviour, presenting dominance based primarily on size. This dominance is established after bouts of duration inversely proportional to the size difference between the opponents. The aggressive repertoire of these pimelodids is similar to that of other studied siluriforms, and consists of varied and articulate components that include oscillations, shoves and bites. Once dominance is established, the subordinates recognize and avoid immediately the dominants after body contact or close proximity. High level of aggressivity in the blind catfishes may be related to competition for food in the cave habitat, where agonistic interactions were frequently observed.
