RESUMO

The influence of thyroxine on activity of enzymes of energy metabolism (hexokinase, phosphofructokinase, pyruvate kinase, laktate dehydrogenase, glucose-6-phosphate dehydrogenase, NADP-isocitrate dehydrogenase, cytochrome-c oxidase) and antioxidative system (glutathione peroxidase, glutathione reductase, superoxide dismutase) of neonatal piglet neutrophils was investigated. It has been found, that after durable injections of hormone (4 mg/kg body weight) the increase of glycolytic enzymes activities as well as aerobic energy pathway catalyzers took place. Simultaneously the augmentation of superoxide dismutase reaction occurred after the thyroxine treatment. Such effect might represent an important link in compensatory mechanism, which prevents the destructive action of reactive oxygen species.

RESUMO

The influence of thyroxine on the activities of enzymes of energy metabolism (hexokinase, 6-phosphofructokinase, pyruvate kinase, lactate dehydrogenase, glucose-6-phosphate dehydrogenase, NADP-isocitrate dehydrogenase, cytochrome c oxidase) was investigated in bone marrow myeloid cells and blood neutrophils of 3-10-day old neonatal piglets. Data obtained suggest different responsiveness of energy metabolism enzymes to thyroxine action. Repeated hormone injections resulted in the preferential stimulation of enzymes involved in oxidative stages of carbohydrate catabolism in animal myelocaryocytes, while the activities of anaerobic enzymes in these cells were less affected. At the same time glycolytic enzymes in neutrophil granulocytes showed higher sensitivity to thyroxine action than enzymes catalyzing oxidative stages of energy metabolism.

RESUMO

Changes in the concentration of haptoglobin-glycoprotein-globulin fraction of blood serum were studied in patients with lung cancer. It was found, that haptoglobin concentration increased in accordance to a disease stage. Formation of haptoglobin hemoglobin (Hp-Hb) complex in the sera of patients may be of great importance for neutralization of superoxidative products.

RESUMO

Changes in the concentration of the lipid peroxidation products in the blood of 152 patients in stroke were studied. 18 patients with LPO processes activation received supplementary treatment with antioxidant preparation (tocopheroli acetas, ascorbic acid, pyridoxalphosphate, glutaminic acid), which lowered significantly the blood content of LPO products; in 12 patients with reduced concentration of LPO products treatments with membrane-stabilizing preparations (lipostabyl, sodium hydroxybutyrate, ascorbic acid, unithiol) were administered, and restoration of LPO processes was established to have occurred. With the purpose of improving methods and assessment of the cerebral insult treatment efficacy, it is recommended that specific features of changes in the concentration of LPO products be used.

RESUMO

Studied after stomach resection for ulcer disease in 161 patients were lipid peroxidation (LPO), antioxidant system (AOS) and related parameters characterizing metabolism of carbon (Lactat), protein (piruvat) and fat (fat acids, lipoproteins), an indicator of the dystrophic change degree (acid phosphatase). Revealed in these patients was syndrome of peroxidation, which leads to disturbances of protein, carbon and fat metabolism, destruction of cell membranes. Increased, "normal", weakened LPO was identified in 67.7%, 9.94%, 22.36% of the patients respectively. In patients with an increased LPO statistically significant activation of AOS was established, lactate having increased more than twice, pyruvate remaining unchanged; the level of fat acids and lipoproteins almost doubled. In such a situation it is reasonable to use preparations which activate aerobic glycolysis, and antioxidants of a direct, mild degree, action. In group II patients with more prominent increase in the acid phosphatase activity preference should be given to membrane-stabilizing preparations. In patients with a decreased level of LPO considerable increase in lactate and acid phosphatase was noted. It is recommended that preparations promoting normalization of carbon and lipid metabolism, enhancement of redox processes in the respiratory chain should be included into complex therapy as should be stabilizers of membranes of cell organelles, with biogenic stimulators to follow.

RESUMO

A study is presented of 89 inpatients with cerebral stroke. Lipid peroxidation was determined during the first days of the diseases (1-7 days). The state of the patients was evaluated by the clinical picture, manifestation of neurological symptoms, biochemical indices of the blood, coagulogram, ECG, REG, state of lipid peroxidation. The obtained data suggest a major role played by lipid peroxidation of the cell membranes, its elevation in of the activity in stroke. The concentration of lipid peroxidation depends on the severity of the condition and, thus, it may be used for evaluation of treatment efficacy and prognosis of the disease.

RESUMO

A new method for measuring serum and tissue antioxidative activity is based on a relationship between accumulation of lipid peroxidation (LPO) products in incubation medium and the amount of the substrate during LPO activation. The ratio of malonic dialdehyde accumulation during LPO activation in two samples of different volumes is proposed as an index of antioxidative activity.

The authors discussed the results of changes in the intensity of lipid peroxidation and antioxidant activity in 70 patients with type I diabetes mellitus (severe forms) before and in 32 patients after the transplantation of human fetal pancreatic islet cell cultures. Lipid peroxidation, antioxidant activity, indices of glycolysis and carbohydrate metabolism were investigated. In some cases metabolic derangement was noted against a background of a decrease in the end products of lipid peroxidation but in a majority of diabetic patients an increase in LPO intensity was noted against a background of raised antioxidant activity, high activity of glycolytic reactions and inhibition of Krebs' cycle. The greatest effect of transplantation was noted in a group of patients who had demonstrated high levels of lactate and the lactate-pyruvate ratio before transplantation. Therefore LPO indices can serve as a criterion of the selection of patients with insulin dependent diabetes mellitus for pancreatic endocrine tissue transplantation.
