RESUMO

This experiment was performed in Wistar rats of both sexes exposed subchronically to 1:100, 1:500, 1:1000 and 1:1500 LD50. The evaluation was based on endpoints measured on the 30th and 90th after starting exposure and after a recovery period of 30 days: these included clinical signs, functional changes, hematological parameters, urine analysis, biochemical, histochemical, immunomorphological endpoints, electron microscopy of internal organs, chromosome examination of bone marrow. A high lethality was shown to occur with a characteristic clinical picture: interruption of weight gain, behavioural changes, leucopenia mainly involving neutrophil leucocytes, biochemical changes characteristic of liver, cardio-vascular system (myocardium and aorta) together with pathologic, biochemical, histochemical and ultrastructural changes in liver, brain, thyroid gland, myocardium, spleen and bone marrow. Endpoints were shown to be clearly dose-dependently related with small variations with the low dose, i.e. 1:1500 LD50 (5 mg/kg-1 bw).

RESUMO

Chloracetophone (0,0-dimethyl-2,2,2-trichloro-1-(1-chloroacetoxy)phosphonate) is a new organophosphorus insecticide. The LD50 for male rats is 420 mg/kg b.w. The mutagenic activity was evaluated with the method of cytogenetic analysis of rat bone marrow after acute and short-term oral exposure. In the acute study groups of male and female animals were treated with chloracetophone at a dose of 1/5 LD50 and then examined at periods of 6, 12, 24, 48 and 72 h following administration. Separate groups of animals were examined 24 h after single administration of the doses 1/5, 1/10 and 1/20 LD50. In the short-term study groups of male animals were treated for 5 successive days. The slides were prepared 6 or 24 h after the last administration. Concurrent negative and positive (cyclophosphamide 20 mg/kg b.w.) control groups were used. 50 cells at metaphase per animal were scored for chromosomal aberrations. The results did not show significant increase in the frequency of chromosomal breaks in the groups with chloracetophone.

RESUMO

The mutagenic activity of basfungin, when acting in combination with sodium nitrite, was studied in a subacute experiment, using the method of bone marrow cytogenetic analysis. The results were compared with those in groups in which basfungin and sodium nitrite were applied separately, as well as with the results of the parallel control. The mutagenic effect in the combined action group did not essentially differ from the effect of application of basfungin alone, both qualitatively and diaulitatively. Interesting results were obtained in the group receiving sodium nitrite alone - the percentage of cells with chromosomal aberrations was significantly than the control values.

The study on the embryotrophic action of phosalon insecticide preparation is conducted against the background of its comprehensive and thorough hygiene-toxicologic assay. Pregnant white rats undergo treatment per os with 1/10 and 1/100 LD50 throughout the entire gravidity period. Recordings are made of the quantity of corpora lutea, pre- and postimplantation lethality; the pedigree development is traced up to the end of the second postnatal month with readings being made of weight and length in the first postnatal day, presence or not of external malformations, weight increase and survival up to the end of the lactation period, day of eye cleft opening and hairing, and sex ratio. The presence of phosalon and phosalon-oxone in whole embryo homogenates is estimated. Enzyme activity of GOT, GPT, catalase, LDH, G-6-PD, RNA and DNA quantity are studied in the liver of newborns on the second postnatal day. Within 21 days of birth, the peripheral blood picture and visceral weight coefficient are also studied. Investigation of the peripheral blood picture, activity of catalase, GOT and GPT in the liver and serum, alkaline and acid phosphatase, and peroxidase in the serum is performed at the end of the second month. The results point to the presence of an embryotoxic effect at the level of the investigated hepatic indicators, detected on the 2nd day of the postnatal period, only at 1/10 LD50 dose.
