RESUMO

Pseudomonas aeruginosa vaccine (PV) containing cell proteins with molecular weight (Mr) 20,000-100,000 and up to 0.08% (w/v) admixture of lipopolysaccharide was obtained by water-salt extraction and subsequent ultrafiltration. PV protects mice against experimental P. aeruginosa infection, stimulates production of specific protective antibodies in rabbit and does not provoke obvious toxicity in laboratory animals.

RESUMO

In this research the experimental analytical method of balancing culture media, based on the determination of the yield of the target product, i.e. staphylococcal protein A, has been used. The medium, developed in the course of this research work on the basis of a hydrolysate of protein-containing waste products of the tanning industry, makes it possible to achieve the growth of producer strains similar to that achieved in peptone-yeast medium. The parameters of the growth of S. aureus strain A-676 and the biological activity of protein A obtained in the newly developed medium have been studied.

RESUMO

The possibility of using the experimental-analytical balance method (EABM) for development of balanced media, optimal and economic by their composition is shown. The method is based on the specific growth activity of the medium components and mathematical calculation of their concentrations. A balanced medium containing human placenta hydrolysate was developed. The medium has a high component consumption coefficient and provides maintenance of high levels of the pneumococcal culture biological properties such as growth, capsule formation, virulence and agglutinability. The quantities of nutrient yeast extract, cystine, glucose and sodium chloride in the balanced medium are 2, 18-20, 4-4.7 and 32-33 times lower respectively than those in the initial medium. The use of the balanced medium instead of the initial unbalanced one or instead of the meat medium based on the Hottinger broth will provide the total economy of 1010 or 2360-2580 roubles respectively per. 1000 liters of the medium. It is concluded that a wider use of this method in optimization of microbiological nutrient media in production of medical and biological preparations is promising.

RESUMO

A culture medium for the isolation and cultivation of pneumococci, produced in a solid or liquid form and based on raw material unsuitable for use as foodstuff (human placenta), has been developed. The amino acid composition of the medium has been studied. The medium has been found to contain 19 amino acids, to be free from ballast serum proteins and blood, and to ensure the good growth of pneumococci isolated from pathological material, the formation of the normal capsule, as well as active biological properties. The medium has proved to create elective and selective conditions enhancing the effectiveness of investigations and simplifying the isolation of pneumococci in the microbiological examination of patients.

RESUMO

The modification of a highly sensitive method for the determination of the concentration of dissolved oxygen in liquid culture media and in water is described. This modification permits the easy conversion of the relative readings of electrode membrane transducers for measuring the partial pressure of oxygen into the readings characterizing the absolute concentration of oxygen in the medium (mg/cu. dm). The content of dissolved oxygen in microbiological media and in water depends on the amount of dissolved mineral salts, amino acids and peptides (the products in incomplete proteolysis), as well as on temperature. The maximum level of oxygen saturation in liquid culture media at 37 degrees C is different for various types of media and constitutes their peculiar characteristic, determined in the process of aeration under experimental conditions.

RESUMO

The physiological activity of pneumococci under different physico-chemical conditions of deep cultivation in a liquid culture medium has been studied. The culture has been shown to achieve its maximum physiological activity under the conditions of neutral pHn-7.0) and the minimum content of dissolved oxygen in the medium. The addition of 5-11% of CO2 to the gas mixture has a positive effect on cultivation, the cultures thus obtained possessing high virulence and pronounced capsules.

RESUMO

For the first time the synthesis of proteolytic enzymes by Corynebacterium diphtheriae has been shown to precede the process of toxin formation. The process of controlling the proteolytic activity of enzymes seems to be a suitable basis for the evaluation of toxin formation in mathematically planned experiments with strictly controlled tasks.

RESUMO

Nutrient media prepared on the basis of microbial cells (Corynebacteria diphtheriae) proved to be no less nutrient in comparison with conventional media prepared on the full-value food products. Use of diphtheria bacilli (by-products of diphtheria toxoid production) as the basis for nutrient media permitted to use up to 30% less food products for this purpose.
