RESUMO

OBJECTIVE: To estimate the capabilities of computed tomography (CT) in revealing the anatomic causes of restrictive lung changes in patients with pulmonary histiocytosis X. SUBJECT AND METHODS: The results of examining 36 patients with pulmonary histiocytosis X, who underwent comprehensive functional study of external respiration (CRSER), CT, and high-performance CT (HPCT), were analyzed. RESULTS: According to the results of CRSER, the authors identified a group of patients with restrictive ventilation disorders, which included 7 men and 1 woman. The most common cause of restrictive disorders was generalized fibrous changes in lung tissue, which fails to result in its expansion. Multiple cysts were another cause of restrictive disorders in 2 patients. Fusion of individual cysts into large ones and the formation of a great number of paradoxically ventilated cysts were the third cause of restrictive disorders in pulmonary histiocytosis X, which was identified during HPCT. CONCLUSION: Comprehensive morphofunctional examination involving CRSER and high-resolution CT can reveal the unfavorable course of pulmonary histiocytosis X with the restrictive type of lesion.

RESUMO

The results of ultrasound (US) and X-ray studies were retrospectively studied in the diagnosis of circumscribed peritonitis in case of microperforation from duodenal ulcers at different stages of emergency medical care. Analysis of the findings has demonstrated that on admission and in its first hours the most effective diagnostic method is plain radiography that enables free gas accumulation to be found under the diaphragm and US study is of low informative value. Repeat targeted US study using expert-class scanners, with the well stated task based on clinical laboratory findings, is a high-informative diagnostic technique for circumscribed peritonitis in case of duodenal perforations.

RESUMO

This paper deals with the results of ultrasound (US) study in 34 patients with a history of colonic diverticulosis, who have been admitted to hospital for the clinical manifestations of diverticular microperforation. Diverticulosis was verified by X-ray irrigoscopy in 15 patients, by oral contrast-enhanced computed tomography in 6, and by colonoscopy in 18. Analysis of US findings revealed the major US syndromes of colonic diverticular microperforation, namely a change in the intestinal wall as its thickening and lower echogenicity, as well as perifocal infiltration and local accumulation of free fluid outside the altered bowel.

RESUMO

This paper deals with the results of ultrasound (US) and multislice computed tomography (MSCT) in patients with perforated duodenal ulcer in different periods after the onset of the disease, with varying clinical manifestations, and in the postoperative period. A comparative analysis of the results of radiation studies has indicated that X-ray study is a basic technique that defines treatment policy in the early development periods of perforation; US study (USS) reveals a change in the duodenum, fluid and infiltration outside the duodenum. As circumscribed peritonitis progresses, USS allows the diagnosis of abdominal abscesses and infiltrations. Out of the radiation techniques, MSCT is of most importance in the diagnosis of interintestinal abscesses and in the determination of syntopy of purulent cavities.

Daily oral administration to rats of probiotic strain Lactobacillus casei 114001 (L.c.) at dose 2,8 x 10(10) cfu/rat during 8 days reduced oxidative stress and liver lesions, induced by a single intraperitoneal administration of carbon tetrachloride (CCl4) at dose 0.5 ml/kg b.w. It was evidenced by several histopathological and biochemical markers, characteristic for CCl4 toroxicity. Membrane damage by toxin was reduced in rats, treated with L.c.: alanine aminotransferase activity in plasma and nonsedimentable activity of lysosomal enzymes in liver were significantly decreased. Treatment with L.c. resulted in partial recovery of activities of antioxidant enzymes and enzymes of xenobiotic metabolism and full recovery of antioxidant capacity of liver cytosol. High level of activity and expression of proteins heme oxygenase and Nrf2 were maintained.

Methodical variations of the computed tomography investigations are described in patients with localization of tumors in different parts of the intestine.

Open reading frame (orf) 129L of ectromelia (EV) and orf A30L of smallpox viruses (SPV) encoding fusion proteins were cloned and expressed in E. coli cells. The recombinant polypeptides (prA30L H pr129L) were purified from cell lysates by Ni-NTA chromatography. Recombinant polypeptides were able to form trimers in buffered saline and they destroyed under treatment with SDS and 2-mercaptoethanol. Reactivity of prA30L, pr129L and orthopoxvirus proteins was analyzed by ELISA and Western blotting with panel of 22 monoclonal antibodies (MAbs) against orthopoxviruses (19 against EV, 2 MAbs against vaccinia virus and 1 Mabs against cowpox virus). This data allowed us to conclude that there are 12 EV-specific epitopes of pr129L and EV fusion proteins, ten orthopox-specific epitopes of EV, VV, CPV fusion proteins, from them 9 orthopox-specific epitopes of prA30L and SPV fusion proteins. Five Mabs, which cross-reacted with orthopox-specific epitopes, were able to neutralize the VV on Vero cells and from them two MAbs has neutralizing activity against smallpox virus. Our findings demonstrate that 129L fusion protein have EV-specific epitopes, that EV 129L and SPV A30L fusion proteins have a several orthopox-specific epitopes to induce a neutralizing antibodies against human pathogenic orthopoxviruses.

The fragment of kinetoplast maxicircle (mitochondrial) DNA divergent region Leptomonas seymouri was localized containing promotor of 12S rRNA gene. The ability of this fragment to initiate transcription was tested with transcription system in organello in isolated mitochondria of L. seymouri.

An extensive collection of 125 rat hybridomas secreting monoclonal antibodies (Mabs) to ectromelia virus (EV) polypeptide (Poxviridae family, Orhtopoxvirus genus) was set up. A significant portion of Mabs (37 types) recognized epitopes of the 14 kDa polypeptide as well as the 37 and 35 kDa polypeptides. However, a majority of Mabs interacted with conformation-dependent epitopes, which were destroyed in immunoprecipitation. One hundred and thirteen of Mabs cross-interacted with antigenic determinants of vaccinia viruses (VV), cowpox virus (CPV) and smallpox virus (SPV); only 12 of them were found to be specific to EV. The Mabs antigenic activity was tested for 46 types of cross-reactivity Mabs in VV neutralization on Vero cells. Only the 112H12, 113D5, 113F8, 122H9 and 125G9 Mabs, which were specific to the kDa 14 polypeptide (gene A30L EV), had the neutralizing activity. The 122H9 and 125G9 Mabs were able to neutralize SPV. Therefore, it can be assumed that the 14 kDa polypeptide carries, on its surface, cross-reactivity neutralizing epitopes typical of orthopoxviruses.
