Research attention is growing for biochar as amendment for anaerobic digestion (AD), as it may improve both the AD process and digestate properties. In this study, two biochars (from insect frass or the woody fraction of green waste, both pyrolyzed at 450 °C) were added (5 % w/w) during semi-continuous AD of organic kitchen waste and chicken manure. Biochar was mixed either during the AD process or to the digestate post-treatment. The impact of biochar addition on the AD process, NH3 and greenhouse gas emissions, as well as C and N mineralization following soil application, was examined. Biochar did not affect the biogas yield, but did lead to a reduction in NH3 levels in the biogas and NH4+-N sorption onto the biochars, reducing the risk of AD inhibition. N sorbed as NH4+-N onto the biochar was more available for mineralization than N sorbed as NH3. N sorption on biochar-amended digestates did not result in trade-offs when these digestates were applied to soil: the sorbed N was not released as NH3 or N2O after soil application and was not released as mineral N in either the short or medium term. The C-rich biochars increased the OC content of the digestate, which enhanced its value as soil improver. Additionally, mixing biochar with digestate decreased the soil CO2 emissions up to 33 % when it was added after AD; no priming effects were observed. These findings highlight the potential benefits of incorporating biochar into AD systems and the subsequent application of biochar-amended digestate in agricultural practices.

Cattle slurry storage is a major source of gaseous N emissions. The aim of this study was to evaluate the effects of biochar, clinoptilolite and elemental sulfur (S°) on (1) NH3 and greenhouse gas emissions during storage of cattle slurry and (2) after soil application of the enriched solid fractions; and (3) on the agronomic quality of the solid and liquid fractions. In the first phase, biochar was added to the slurry (10 g L-1); subsequently in the second phase, clinoptilolite (50 g L-1), S° (1 g L-1) and 40 g L-1 extra biochar were added. Gaseous emissions were monitored by a semi-continuous multi-gas analyzer and the agronomic quality of solid and liquid fractions was assessed after separation. The enriched solid fractions were applied to soil to study the effects on gaseous emissions, N and C mineralization. Amendment of biochar reduced NH3 emissions during cattle slurry storage by 12% during the first 7 days. Extra amendment of biochar, clinoptilolite and S° in combination with biochar resulted in a decrease of NH3 emissions of approximately 20%. The N sorbed from the slurry by the biochar was not released as NH3 during soil application of the solid fractions and was not released as mineral N in the short term (within 28 days). A short-term positive priming effect of biochar on the C mineralization of manure and biochar-manure mixture applied to soil was observed. The biochar-enriched solid fractions contained more C, total and organic N and water-available P with a slow release.

Biochar amendment during biomass processing can improve those processes and products, and reduce the emissions of greenhouse gases and NH3, resulting in ecologic and economic benefits. The potential positive effects of biochar are related to NH4+-N and NH3 sorption, which in turn are depending on different biochar characteristics. By knowing the relationship between biochar characteristics and NH4+-N and NH3 sorption, biochar production can be steered towards a higher N sorption or existing biochars can be selected for targeted applications for high N sorption. Therefore, this study aims to develop fast screening tests to estimate the potential for both NH4+-N and NH3 sorption of biochar, before application in biomass processing. Further, the effects of feedstock, pyrolysis temperature, biochar characteristics and biochar treatments on N sorption are studied. The results show that NH4+-N sorption varied between 0 and 1.54 mg NH4+-N/g fresh biochar and was highest for manure-based biochars with a high nutrient content and cation exchange capacity, produced at lower temperatures (300-450 °C). For some biochars, the feedstock itself had a higher NH4+-N sorption than the biochar. Grinding and washing increased the NH4+-N sorption. In addition, a general linear model was proposed to predict the NH4+-N sorption based on three chemical characteristics· NH3 sorption varied between 0 and 100 % of the negative control and showed a linear positive relationship with the NH4+-N sorption, moisture retention factor and cation exchange capacity. Pyrolysis temperature and feedstock type did not significantly affect NH3 sorption. NH4+-N and NH3 desorption varied amongst biochar type.

We investigated the potential of C-rich byproducts to replace wood chips as bulking agent (BA) during composting. The impact of these alternatives on the composting process and on compost stability and characteristics was assessed. Three BA (chopped heath biomass and spent growth media used in strawberry and tomato cultivation) were used for processing leek residues in windrow composting. All BA resulted in stable composts with an organic matter (OM) content suitable for use as soil amendment. Using chopped heath biomass led to high pile temperatures and OM degradation and a nutrient-poor compost with high C/P ratio appropriate for increasing soil organic carbon content in P-rich soils. Spent substrates can replace wood chips, however, due to their dense structure and lower biodegradation potential, adding a more coarse BA is required. Generally, the nutrient content of the composts with growth media was higher than the composts with wood chips and chopped heath biomass.

Maintaining and increasing soil quality and fertility in a sustainable way is an important challenge for modern agriculture. The burgeoning bioeconomy is likely to put further pressure on soil resources unless they are managed carefully. Compost has the potential to be an effective soil improver because of its multiple beneficial effects on soil quality. Additionally, it fits within the bioeconomy vision because it can valorize biomass from prior biomass processing or valorize biomass unsuitable for other processes. However, compost is rarely used in intensive agriculture, especially in regions with high manure surpluses. The aim of this research is to identify the barriers to on-farm composting and the application of compost in agriculture, using a mixed method approach for the case of Flanders. The significance of the 28 identified barriers is analyzed and they are categorized as market and financial, policy and institutional, scientific and technological and informational and behavioral barriers. More specifically, the shortage of woody biomass, strict regulation, considerable financial and time investment, and lack of experience and knowledge are hindering on-farm composting. The complex regulation, manure surplus, variable availability and transport of compost, and variable compost quality and composition are barriers to apply compost. In conclusion, five recommendations are suggested that could alleviate certain hindering factors and thus increase attractiveness of compost use in agriculture.

Mallotus and Phyllanthus genera, both containing several species commonly used as traditional medicines around the world, are the subjects of this discrimination and classification study. The objective of this study was to compare different discrimination and classification techniques to distinguish the two genera (Mallotus and Phyllanthus) on the one hand, and the six species (Mallotus apelta, Mallotus paniculatus, Phyllanthus emblica, Phyllanthus reticulatus, Phyllanthus urinaria L. and Phyllanthus amarus), on the other. Fingerprints of 36 samples from the 6 species were developed using reversed-phase high-performance liquid chromatography with ultraviolet detection (RP-HPLC-UV). After fingerprint data pretreatment, first an exploratory data analysis was performed using Principal Component Analysis (PCA), revealing two outlying samples, which were excluded from the calibration set used to develop the discrimination and classification models. Models were built by means of Linear Discriminant Analysis (LDA), Quadratic Discriminant Analysis (QDA), Classification and Regression Trees (CART) and Soft Independent Modeling of Class Analogy (SIMCA). Application of the models on the total data set (outliers included) confirmed a possible labeling issue for the outliers. LDA, QDA and CART, independently of the pretreatment, or SIMCA after "normalization and column centering (N_CC)" or after "Standard Normal Variate transformation and column centering (SNV_CC)" were found best to discriminate the two genera, while LDA after column centering (CC), N_CC or SNV_CC; QDA after SNV_CC; and SIMCA after N_CC or after SNV_CC best distinguished between the 6 species. As classification technique, SIMCA after N_CC or after SNV_CC results in the best overall sensitivity and specificity.

Neural-tube defects (NTDs) are considered to be the most common congenital malformations. As Shanxi Province, a poor region in the North of China, has one of the highest reported prevalence rates of NTDs in the world, folate fortification of rice is an excellent alternative to low intake of folate acid pills in this region. This paper investigates the relations between socio-demographic indicators, consumer characteristics (knowledge, consumer perceptions on benefits, risks, safety and price), willingness-to-accept and willingness-to-pay genetically modified (GM) rice. The consumer survey compromises 944 face-to-face interviews with rice consumers in Shanxi Province, China. Multivariate analyses consist of multinomial logistic regression and multiple regression. The results indicate that consumers generally are willing-to-accept GM rice, with an acceptance rate of 62.2%. Acceptance is influenced by objective knowledge and consumers' perceptions on benefits and risks. Willingness-to-pay GM rice is influenced by objective knowledge, risk perception and acceptance. Communication towards the use of GM rice should target mainly improving knowledge and consumers' perceptions on high-risk groups within Shanxi Province, in particular low educated women.

Converting the complete genome sequence of Candida albicans into meaningful biological information will require comprehensive screens for identifying functional classes of genes. Most systems described so far are not applicable to C. albicans because of its difficulty with mating, its diploid nature, and the lack of functional random insertional mutagenesis methods. We examined artificial gene suppression as a means to identify gene products critical for growth of this pathogen; these represent new antifungal drug targets. To achieve gene suppression we combined antisense RNA inhibition and promoter interference. After cloning antisense complementary DNA (cDNA) fragments under control of an inducible GAL1 promoter, we transferred the resulting libraries to C. albicans. Over 2,000 transformant colonies were screened for a promoter-induced diminished-growth phenotype. After recovery of the plasmids, sequence determination of their inserts revealed the messenger RNA (mRNA) they inhibited or the gene they disrupted. Eighty-six genes critical for growth were identified, 45 with unknown function. When used in high-throughput screening for antifungals, the crippled C. albicans strains generated in this study showed enhanced sensitivity to specific drugs.

To develop better molecular genetic tools for the diploid yeast Candida albicans, the suitability of the MET15 gene as a visual selection marker was studied. Both MET15 alleles of C. albicans CAI-4 were isolated by functional complementation of a Saccharomyces cerevisiae strain lacking the MET15 gene. Growth of this complemented strain on Pb(2+)-containing medium was associated with a colour shift of brown into white colonies. The MET15 alleles of C. albicans were located on chromosome 4 by pulsed-field gel electrophoresis and Southern blotting. A met15-deficient strain of C. albicans CAI-4 was generated using the ura blaster technique. This strain showed a brown colony colour on Pb(2+)-containing medium, which corresponded with the colony colour of a S. cerevisiae strain lacking the MET15 gene. Unexpectedly, the met15-deficient strain of C. albicans still grew on methionine-depleted medium. However, this growth was severely delayed. In addition, complementation of this strain with an integrative or replicative plasmid containing either of the MET15 alleles resulted in the formation of white transformants on Pb(2+)-containing medium. These transformants grew very well on methionine-depleted medium. Colony sectoring was obtained with the replicative plasmid and not with the integrative one. This study demonstrates that the MET15 gene of C. albicans is suitable as a visual marker and therefore can be used to identify transformants and study plasmid stability. A multi-site field study was conducted to evaluate an inactivated Mycoplasma hyopneumoniae (Mh) vaccine in 14 pig herds infected by Mh and practising an all-in/all-out production system. In each herd, a vaccinated and control group of 250 pigs each were compared during the growing/finishing period with respect to performance parameters (major variables) and by means of clinical, serological and pathological parameters (ancillary variables). Mh vaccination significantly (P < 0.05) improved daily weight gain (+22 g), feed conversion ratio (-0.07), medication costs (-0.476 ECU/pig) (1 ECU = US$1.0269542), prevalence of pneumonia lesions (-14%) and severity of pneumonia lesions (-3%). Mortality rate, severity of coughing and carcass quality were not significantly influenced by Mh vaccination. Serological results of Mh and other respiratory pathogens are presented and discussed. A cost-benefit analysis based on significantly improved performance parameters demonstrated that Mh vaccination was economically attractive as it resulted in an increase of the net return to labour with 1.300 ECU per finishing pig sold. The sensitivity of the economic benefit was illustrated towards fluctuations in pig finishing prices.

This article focuses on the impact of mass media meat-health information on consumer perception, attitude, and behavior toward fresh meat in Belgium. In a situation similar to that which occurred in most other European countries, Belgian fresh meat consumption fell considerably during 1995-1999. A multitude of messages linking meat consumption to human health risks were reported by mass media. Bovine Spongiform Encephalopathy (BSE) since 1996 and dioxin in 1999 constituted the major issues. Empirical research, conducted in April 1998, revealed the tremendous negative impact of mass media coverage of meat-health issues on consumer risk perception, health concern, and attitude and behavior toward fresh meat. Oppositely, personal communication through butchers had only a small effect on consumer decision-making in this era dominated by alarming meat-health press. Implications are threefold. First, mass media should be aware of its social responsibilities, which include spreading reliable and correct information to the society. This is especially the case as human health risks are involved. Second, the meat industry urgently needs to reorient itself toward quality, safety, and transparency. Finally, future communication dealing with similar crises situations requires cooperation across the meat chain, government, and those who are responsible for public health promotion and communication.

Changes at consumer level, as well as an image decline of the meat sector, resulted in considerable decreases of fresh meat consumption. Consumer orientation is considered as a prerequisite to slow down or reverse the adverse fresh pork consumption evolution. Consumer perception of pork is taken as the starting point of this article. Focus is first on assessing differences between facts related to nutritional value and healthiness aspects of pork and their perception by Belgian meat consumers. Second, possibilities of improving pork characteristics are identified. Pork is perceived worst as compared to beef and poultry on the attributes leanness, healthiness, taste and tenderness. Consumer-oriented response strategies should focus at improving these intrinsic quality characteristics, before other elements like traceability, labelling or marketing can be implemented successfully. The possibilities to improve nutritional value, healthiness and sensory characteristics pertain to selection, pig diet composition, transport, slaughter and post-slaughter circumstances. A successful adoption of consumer orientation urges for co-operation throughout the entire pork production chain. The identified topics are key attention points for adequate production and marketing by the pork sector, as well as for consequent communication by government and public services.

In this study, we showed that the cell wall anchor of protein A from Staphylococcus aureus is functional in the food-grade organism Lactococcus lactis. A fusion protein composed of the lactococcal Usp45 secretion signal peptide, streptavidin monomer, and the S. aureus protein A anchor became covalently attached to the peptidoglycan when expressed in L. lactis. The streptavidin moiety of the fusion protein was functionally exposed at the cellular surface. L. lactis cells expressing the anchored fusion polypeptide could be specifically immobilized on a biotinylated alkaline phosphatase-coated polystyrene support.
