The aim of this study was to evaluate the perioperative analgesic and sedative effects of oral CBD in cats undergoing ovariohysterectomy. Twenty-two cats were assigned to receive either oral cannabidiol oil (2 mg/kg, CBD group, n = 12) or placebo oil (0.1 mL/kg, Placebo group, n = 10) 60 min before the premedication. The anesthetic protocol included dexmedetomidine/meperidine, propofol, and isoflurane. Intravenous fentanyl was given to control cardiovascular responses to surgical stimulation. Pain was assessed at 0.5, 1, 2, 4, 6 and 8 h post-extubation using the UNESP-Botucatu multidimensional composite pain scale and the Glasgow feline composite-measure pain scale. Sedation scores were assessed at the same timepoints and at 15 min after the premedication. Morphine was administered as rescue analgesia. Higher sedation scores were recorded in the CBD group at 15 min after premedication (p = 0.041). Intraoperatively, more cats required fentanyl in the Placebo group than in CBD group (p = 0.028). The pain scores did not differ between groups, except at 0.5 h post-extubation when lower scores were detected in the CBD group (p = 0.003-0.005). Morphine was required in 100% of the animals in both groups. CBD increased preoperative sedation and decreased intraoperative analgesic requirements, with minimal evidence of postoperative analgesic benefits over the placebo.
RESUMO
Pancreatic ductal adenocarcinoma (PDAC) represents one of the deadliest cancers among all solid tumors. First-line treatment relies on gemcitabine (Gem) and despite treatment improvements, refractoriness remains a universal challenge. Attempts to decipher how feedback-loops control signaling pathways towards drug resistance have gained attention in recent years, particularly focused on the role of phosphatases. In this study, a CRISPR/Cas9-based phenotypic screen was performed to identify members from the dual-specificity phosphatases (DUSP) family potentially acting on Gem response in PDAC cells. The approach revealed the atypical RNA phosphatase DUSP11 as a potential target, whose inhibition creates vulnerability of PDAC cells to Gem. DUSP11 genetic inhibition impaired cell survival and promoted apoptosis, synergistically enhancing Gem cytotoxicity. In silico transcriptome analysis of RNA-seq data from PDAC human samples identified NF-ĸB signaling pathway highly correlated with DUSP11 upregulation. Consistently, Gem-induced NF-ĸB phosphorylation was blocked upon DUSP11 inhibition in vitro. Mechanistically, we found that DUSP11 directly impacts nc886 expression and modulates PKR-NF-ĸB signaling cascade after Gem exposure in PDAC cells resulting in resistance to Gem-induced cell death. In conclusion, this study provides new insights on DUSP11 role in RNA biology and Gem response in PDAC cells.

OBJECTIVE: Pediatric cardiac tumors are rare and, among them, 90% are benign. Cardiac fibroma is the second most frequent tumor, after rhabdomyoma. The objective of this study is to report a case of cardiac fibroma diagnosed incidentally in a patient admitted with acute viral bronchiolitis. CASE DESCRIPTION: A 5-month-old male infant was admitted to the pediatric emergency department with acute viral bronchiolitis requiring hospitalization. He presented a detectable respiratory syncytial virus in oropharyngeal swab, blood test with lymphocytosis and a chest radiography revealed cardiomegaly. Further cardiologic testing was performed detecting elevation of cardiac biomarkers, an electrocardiogram with alteration of left ventricular repolarization and echocardiogram with a heterogeneous mass in the left ventricular, with areas of calcification. A chest angiotomography suggested rhabdomyosarcoma or cardiac fibroma and a magnetic resonance showed a mass, with characteristics suggesting fibroma. The final diagnosis was made after two cardiac catheterizations for biopsy of the lesion, confirming cardiac fibroma by anatomopathological examination. Because the patient had moderate to severe systolic dysfunction, he was submitted to heart transplant. COMMENTS: One third of cardiac fibromas are asymptomatic, generally diagnosed late through tests ordered for other reasons. The gold-standard test for definitive diagnosis is biopsy. Cardiac fibroma usually does not present spontaneous regression and, in most cases, partial or total surgical resection is necessary. When tumors are unresectable, heart transplantation should be indicated. It is essential to have detailed characterization of the cardiac mass to establish the most appropriate therapeutic approach for each patient.

RESUMO A saúde dos homens tem sido influenciada ao longo do tempo por complexas redes de normas culturais e sociais que delimitam o conceito de masculinidade. Este artigo teve como objetivo analisar a influência das representações de masculinidade na saúde dos homens, com ênfase nas implicações para diagnóstico, tratamento e prevenção do câncer de próstata. Explorou-se a construção social da masculinidade, destacando como os papéis designados aos homens moldam suas atitudes com relação à saúde. Esta pesquisa é um recorte de estudo realizado durante o mestrado acadêmico, o qual traçou os itinerários terapêuticos de pacientes com câncer de próstata do Hospital Universitário de Brasília, a fim de identificar as repercussões da pandemia. Trata-se de uma pesquisa qualitativa baseada no método dos itinerários terapêuticos. O estudo incluiu a participação de 15 homens adultos diagnosticados com câncer de próstata, e os resultados evidenciaram a influência da construção da masculinidade na busca de tratamento. Os participantes hesitaram em procurar assistência médica imediata devido ao receio de parecerem vulneráveis. Além disso, a principal compreensão entre os participantes estava relacionada à preocupação de deixar a família desprotegida, destacando como as expectativas tradicionais do gênero ainda influenciam a sociedade.
ABSTRACT Men's health has been influenced over time by complex networks of cultural and social norms that define the concept of masculinity. This article aimed to analyze the influence of representations of masculinity on men's health, with an emphasis on the implications for the diagnosis, treatment and prevention of prostate cancer. The social construction of masculinity is explored, highlighting how the roles assigned to men shape attitudes towards health. This research is part of a study carried out during the academic master's degree, which outlined the therapeutic itineraries of patients with prostate cancer at the University Hospital of Brasília, in order to identify the repercussions of the pandemic. This is qualitative research based on the therapeutic itinerary method. The study included the participation of 15 adult men diagnosed with prostate cancer, and the results highlighted the influence of the construction of masculinity in the search for treatment. Participants were hesitant to seek immediate medical assistance due to concerns about appearing vulnerable. Furthermore, the main understanding among participants was related to the concern of leaving the family unprotected, highlighting how traditional gender expectations still influence society.

RESUMO Objetivo: Tumores cardíacos pediátricos são raros e, entre eles, 90% são benignos. O fibroma cardíaco é o segundo mais frequente, após o rabdomioma. O objetivo deste trabalho é relatar um caso de fibroma cardíaco, cujo diagnóstico foi incidental em um paciente internado por bronquiolite viral aguda. Descrição do caso: Lactente do sexo masculino com cinco meses foi atendido em um pronto atendimento pediátrico com quadro de bronquiolite viral aguda, necessitando de hospitalização. Em exames complementares, apresentou vírus sincicial respiratório detectável, hemograma com linfocitose, além de radiografia de tórax com cardiomegalia. Em investigação cardiológica, as enzimas cardíacas encontravam-se elevadas, o eletrocardiograma apresentava alteração de repolarização de parede lateral de ventrículo esquerdo e o ecocardiograma demonstrou massa heterogênea em ventrículo esquerdo, com áreas de calcificação. A angiotomografia de tórax sugeriu rabdomiossarcoma ou fibroma cardíaco e a ressonância magnética demonstrou massa cuja localização e características sugeriram fibroma. O diagnóstico foi conclusivo após dois cateterismos para biópsia da lesão, confirmando fibroma cardíaco pelo anatomopatológico. Em razão da disfunção sistólica moderada a grave, o paciente foi submetido a transplante cardíaco. Comentários: Um terço dos fibromas cardíacos é assintomático, geralmente diagnosticado tardiamente por meio de exames solicitados por outro motivo. O padrão-ouro para o diagnóstico definitivo é a biópsia. O fibroma cardíaco não costuma apresentar regressão espontânea, sendo submetido, na maioria dos casos, a ressecção cirúrgica parcial ou total. Nos tumores irressecáveis, o transplante cardíaco deve ser indicado. A caracterização detalhada da massa cardíaca é fundamental para traçar a conduta terapêutica mais adequada para cada paciente.
RESUMO
This work aimed to detect Mycoplasma cynos, M. canis, M. edwardii, and M. molare in different types of kennels, in addition to evaluating their distribution in different colonization sites. The dogs belonged to different kennels from armed forces (n = 3), shelters (n = 3), and commercial purposes (n = 2). Samples of the oropharynx, genital mucosa, and ear canal were collected from each dog (n = 98), totaling 294 samples. Aliquots were submitted to isolation and the samples confirmed as Mycoplasma spp. were subjected to conventional PCR for M. canis and multiplex PCR for M. edwardii, M. molare, and M. cynos detection. Of the 98 dogs studied, 63.3% (62) were positive in at least one anatomical site evaluated for Mycoplasma spp. Among the 111 anatomical sites positive for Mycoplasma spp., M. canis, M. edwardii, and M. molare were detected in 29.7% (33/111), 40.5% (45/111), and 2.70% (3/111), respectively. No animal was positive for M. cynos.

Hypoparathyroidism remains the single endocrine deficiency disease that is not habitually treated with the missing hormone. In this article, we aim to provide a review of the conventional approach and the novel therapies as well as an overview of the perspectives on the treatment of this rare condition. We conducted a literature review on the conventional therapy using vitamin D analogs and calcium salts, indications for thiazide diuretics and phosphorus binders, PTH analogs history and usage, and the drugs that are currently being tested in clinical trials. Conventional treatment involves calcium salts and vitamin D analogs. Thiazide diuretics can be used to reduce hypercalciuria in some cases. A low-phosphate diet is recommended, and phosphate binders are rarely needed. During pregnancy, a careful approach is necessary. The use of PTH analogs is a new approach despite the limitation of high cost. Studies have included modified PTH molecules, calcilytics, microencapsulation of human parathyroid cells, and allotransplantation.

Esse estudo seccional, realizado no período 2014-2015, com aplicação de questionários, teve por objetivo conhecer a percepção sobre vertebrados em condição de sinantropia e a opinião sobre controle populacional para a população do município de Niterói, RJ. Dentre os 474 entrevistados, 76,8% aceitavam o controle populacional para ratos, 63,3% para pombos, 40,5% para cães, 33,8% para gatos, 25,5% para morcegos, 16,5% para serpentes e 14,1% para gambás. As serpentes foram os animais mais associados ao medo (59,1%) e risco de agressão (47,7%), pombos (89,2%) e morcegos ao risco de doenças (57,4%), ratos à nojo/repulsa (56,6%), gambás à natureza (73,8%), cães à companhia (79,8%), e gatos à amizade (57,4%). O estudo demonstrou que as relações dos humanos com os animais podem ser complexas, ambíguas e paradoxais, e fornece dados que poderão ser utilizados para políticas públicas de manejo dessas espécies.(AU)
This study (sectional investigation), in the 2014-2015, using questionnaires, aimed to understand the perception of vertebrates in synanthropic conditions and the opinion about their population control for the population of the municipality the Niterói, RJ. Among the 474 people interviewed, 76.8% accepted population control for rats, 63.3% for pigeons, 40.5% for dogs, 33.8% for cats, 25.5 % for bats, 16.5% for snakes and 14.1% for possums. Snakes were the animals most associated with the representation of fear (59.1%) and risk of aggression (47.7%), pigeons (89.2%) and bats with risk of disease (57.4%), rats with disgust and repulsion (56.6%), possums with nature (73.8%), dogs with company (79.8%), and cats with friendship (57.4%). In addition to demonstrating how complex human relations with animals can be ambiguous and paradoxical, this study provides data that can be used for public policies for the management of these species.(AU)
El objetivo de este estudio fue conocer la percepción de los vertebrados en condiciones de sinantropía y la opinión sobre el control poblacional de la población del municipio de Niterói, RJ. Entre los 474 entrevistados, el 76,8% aceptó el control de la población de ratas, el 63,3% de palomas, el 40,5% de perros, el 33,8% de gatos, el 25,5% de murciélagos, el 16,5% de serpientes y el 14,1% de zarigüeyas. One of the challenges in studying neuropsychiatric disorders is the difficulty in accessing brain tissue from living patients. Schizophrenia is a chronic mental illness that affects 1% of the population worldwide, and its development stems from genetic and environmental factors. In order to better understand the pathophysiology underlying schizophrenia, the development of efficient in vitro methods to model this disorder has been required. In addition to several in vitro models, induced pluripotent stem cells (iPSCs) arose as a powerful tool, enabling access to the genetic background of the donor. Moreover, genetic modification of these cells can improve studies of specific dysfunctions observed in the pathophysiology of several neuropsychiatric disorders, not only schizophrenia. Here, we summarize which in vitro models are currently available and their applications in schizophrenia research, describing their advantages and limitations. These technologies in the cell culture field hold great potential to contribute to a better understanding of the pathophysiology of schizophrenia in an integrated manner, in addition to testing potential therapeutic interventions based on the genetic background of the patient.

Schizophrenia is a complex and heterogeneous neurodevelopmental psychiatric disorder characterized by a variety of symptoms classically grouped into three main domains: positive (hallucinations, delusions, and thought disorder) and negative symptoms (social withdrawal, lack of affect) and cognitive dysfunction (attention, working and episodic memory functions, and processing speed). This disorder places an immense emotional and economic pressure on the individual and society-at-large. Although the etiology of schizophrenia is not completely known, it is proposed to involve abnormalities in neurodevelopmental processes and dysregulation in the signaling mediated by several neurotransmitters, such as dopamine, glutamate, and GABA. Preclinical research using animal models are essential in our understanding of disease development and pathology as well as the discovery and advance of novel treatment choices. Here we describe rodent models for studying schizophrenia, including those based on the effects of drugs (pharmacological models), neurodevelopmental disruption, demyelination, and genetic alterations. The advantages and limitations of such models are highlighted. We also discussed the great potential of proteomic technologies in unraveling the molecular mechanism of schizophrenia through animal models.

Schizophrenia is a psychiatric disorder of neurodevelopmental origin that is thought to result from the combination of genetic and socioenvironmental factors. Several studies have linked the endocannabinoid system with the pathophysiology of schizophrenia. Here, we provide a brief overview of the role of the endocannabinoid system (ECS) in the context of biological processes relevant to schizophrenia, such as neurodevelopment, synaptic plasticity, and brain energy metabolism. We also discuss alterations related to the ECS in schizophrenia and current efforts in both in vivo and in vitro studies that have provided a better understanding of the functioning of this system in the context of the disorder. Finally, we highlighted the modulation of the ECS as a potential for discovering novel therapeutic targets, suggesting new avenues for future research in the field.

Abstract Objective: To assess intra- and interobserver agreement among non-expert pathologists in identifying features of the eosinophilic esophagitis histologic scoring system (EoEHSS) in pediatric patients. Patients and methods: The authors used 50 slides from patients (aged 1-15 years; 72% male) with EoE. EoEHSS evaluates eosinophilic inflammation and other features including epithelial basal zone hyperplasia, eosinophilic abscesses, eosinophil surface layering, dilated intercellular spaces, surface epithelial alteration, dyskeratotic epithelial cells, and lamina propria fibrosis. Grade and stage of abnormalities are scored using a 4-point scale (0 normal; 3 maximum change). Four pathologists determined EoEHSS findings on two occasions. Intra- and interobserver agreement was assessed using Kappa (κ) statistics and intra-class correlation coefficients. Results: Intra- and interobserver agreement for the identification of eosinophil counts ≥ 15/high power field (HPF) was excellent, however varied when assessing additional features of the EoEHSS. For the more experienced pathologist, agreement for most EoEHSS items and the composite scores was substantial to excellent. For the less experienced pathologists, intraobserver agreement ranged from absent to substantial for individual features and ranged from moderate to substantial for the composite scores. Conclusion: Most items of the EoEHSS had substantial to excellent reliability when assessed by a pathologist experienced in the diagnosis of EoE but presented lower repeatability among less experienced pathologists. These findings suggest that specific training of pathologists is required for the identification of EoEHSS characteristics beyond eosinophil count, as these features are considered useful in the evaluation of response to treatment and correlation with clinical manifestations and endoscopic findings.

Adultez emergente é uma fase do desenvolvimento na qual os indivíduos se preparam para adentrar a vida adulta. Devido as suas características, é um período propício para estudar projeto de vida. Este estudo teve como objetivo investigar as concepções de estudantes universitárias sobre estágio extracurricular e sobre sua relação com desenvolvimento do projeto de vida. Foi realizado um grupo focal com três estudantes do curso de psicologia. Para a análise de dados, foi feita uma análise de conteúdo. Os resultados mostraram que projeto de vida e estágio extracurricular são percebidos como relacionados. Adultez emergente es una fase del desarrollo en la cual los individuos se preparan para adentrar a la vida adulta. Por sus características, es un período propicio para estudiar proyecto de vida. En este estudio se tuvo como objetivo investigar las concepciones de estudiantes universitarias sobre pasantía extracurricular y sobre su relación con desarrollo del proyecto de vida. Se realizó un grupo focal con tres estudiantes del curso de psicología. Para el análisis de datos, se hiso un análisis de contenido. Los resultados apuntan que proyecto de vida y pasantía extracurricular son percibidos como relacionados. Estas temáticas son evaluadas como benéficas para el desarrollo, ayudando en el establecimiento de la identidad, en la autopercepción de esas jóvenes como adultas y en la toma de decisiones.
Emerging adulthood is a stage of development in which individuals prepare to enter adult life. Due to its characteristics, it is a favorable period to study a purpose in life. This study aimed to investigate the conceptions of university students about extracurricular internship and its relationship with the development of the purpose in life. A focus group was carried out with three students from the psychology course. For data analysis, a content analysis was performed. The results showed that purpose in life and extracurricular internship are perceived as related. These themes are seen as beneficial for development, helping to establish identity, in the self-perception of these young people as adults and in decision-making.

Esse estudo seccional, realizado no período 2014-2015, com aplicação de questionários, teve por objetivo conhecer a percepção sobre vertebrados em condição de sinantropia e a opinião sobre controle populacional para a população do município de Niterói, RJ. Dentre os 474 entrevistados, 76,8% aceitavam o controle populacional para ratos, 63,3% para pombos, 40,5% para cães, 33,8% para gatos, 25,5% para morcegos, 16,5% para serpentes e 14,1% para gambás. As serpentes foram os animais mais associados ao medo (59,1%) e risco de agressão (47,7%), pombos (89,2%) e morcegos ao risco de doenças (57,4%), ratos à nojo/repulsa (56,6%), gambás à natureza (73,8%), cães à companhia (79,8%), e gatos à amizade (57,4%). O estudo demonstrou que as relações dos humanos com os animais podem ser complexas, ambíguas e paradoxais, e fornece dados que poderão ser utilizados para políticas públicas de manejo dessas espécies.

Dry eye disease (DED) is a multifactorial disease affecting tear quality and/or production and eye surface and is one of the most common eye disorders found in clinical practice. The association between psychiatric disorders and dry eye has been the subject of several studies since patients with this syndrome present a tendency toward a depressive mood. This narrative review aims to demonstrate the relationship between depression and DED, which is due to the side effects of psychotropic drugs or the tendency of the low pain threshold of the depressive patient. The work was produced from the analysis of 13 articles published during the last decade on this subject and demonstrated that the depressive state is linked to the appearance or worsening of DED resulting from chronic eye pain. Also, the treatment of depression with selective inhibitors of serotonin receptors causes inflammatory cytokine secretion with subsequent inflammation and apoptosis of cells on the ocular surface. The need for new studies on optimization of psychiatric treatment in patients with ophthalmic diseases, such as DED, was verified, aiming at the relief of symptoms and the reduction of psychological and eye damage caused by them.

Este estudo objetivou identificar fatores associados a quedas entre idosos adscritos à Estratégia de Saúde da Família (ESF). Estudo transversal observacional exploratório de base populacional, realizado com amostra de 571 idosos. A coleta de dados foi realizada por instrumento semiestruturado e testes validados GDS-Scale, FESI-Brasil, TUG test, e SPPB. As associações foram determinadas por Modelos de Regressão Logística. Em relação à ocorrência de quedas no ano anterior, 139 (26%) relataram ter sofrido queda. Foram associados a quedas maior número de comorbidades (OR1,56 IC95%1,007-2,430), uso de dispositivo auxiliar de marcha (OR2,18 IC95%1,054-4,556), relato de alteração auditiva (OR1,61 IC95%1,036-2,495), negativa de prática regular de exercício físico (OR0,571 IC95%0,359-0,895), presença de dor (OR1,650 IC95%1,073-2,558), e baixa autoeficácia em relação a quedas (OR1,568 IC95%1,010-2,442). Foram identificados ao final do estudo seis fatores associados a quedas entre idosos adscritos à ESF, que devem ser inseridos na avaliação desta população.
To identify fall-associated factors among the elderly enrolled in Family Health Strategy (FHS). Cross-sectional, exploratory, population-based study conducted with a sample of 571 elderly. Data were collected using a semi-structured instrument and validated tests GDS-Scale, FESI-Brasil, TUG test, and SPPB. Associations were determined by Logistic Regression Models. Regarding the occurrence of falls in the previous year, 139 (26%) reported at least one fall. Falls were associated with a greater number of comorbidities (OR1.56 95%CI 1.007-2.430), use of a walking aid (OR2.18 95%CI 1.054-4.556), hearing loss report (OR1.61 95%CI 1.036-2.495), negative practice of regular physical exercise (OR0.571 95%CI 0.359-0.895), presence of pain (OR1.650 95%CI 1.073-2.558), and low self-efficacy in relation to falls (OR1.568 95%CI 1.010-2.442). At the end of the study, six factors associated with falls were identified among elderly people enrolled in FHS, which should be included in the evaluation of this population.

Spirulina platensis is a "super-food" and has attracted researchers' attention due to its anti-inflammatory, antioxidant, and analgesic properties. Herein, we investigated the antinociceptive effects of Spirulina in different rodent behavior models of inflammatory pain. Male Swiss mice were treated with Spirulina (3-300 mg/kg, p.o.), indomethacin (10 mg/kg, p.o.), or vehicle (0.9% NaCl 10 mL/kg). Behavioral tests were performed with administration of acetic acid (0.6%, i.p.), formalin 2.7% (formaldehyde 1%, i.pl.), menthol (1.2 µmol/paw, i.pl.), cinnamaldehyde (10 nmol/paw, i.pl.), capsaicin (1.6 µg/paw, i.pl.), glutamate (20 µmol/paw, i.pl.), or naloxone (1 mg/kg, i.p.). The animals were also exposed to the rotarod and open field test to determine possible effects of Spirulina on locomotion and motor coordination. The quantitative phytochemical assays exhibited that Spirulina contains significant concentrations of total phenols and flavonoid contents, as well as it showed a powerful antioxidant effect with the highest scavenging activity. Oral administration of Spirulina completely inhibited the abdominal contortions induced by acetic acid (ED50 = 20.51 mg/kg). Spirulina treatment showed significant inhibition of formalin-induced nociceptive behavior during the inflammatory phase, and the opioid-selective antagonist markedly blocked this effect. Furthermore, our data indicate that the mechanisms underlying Spirulina analgesia appear to be related to its ability to modulate TRMP8 and TRPA1, but not by TRPV1 or glutamatergic system. Spirulina represents an orally active and safe natural analgesic that exhibits great therapeutic potential for managing inflammatory pain disorders.

The impact of cerium (Ce) and neodymium (Nd) rare-earth metal doping of TiO2 prepared by the hydrothermal method was investigated to tailor effective photocatalytic degradation of coloured wastewater under UV or visible illumination. The hydrothermal treatment of TiO2 decreased the pHpzc from 6.3 to 3.1-3.8 favouring the affinity for cationic water contaminants. Doping with Ce and Nd modified the crystallinity and the morphology of the photocatalysts and significantly increased the BET surface area and the adsorption capacity of cationic dyes. The photocatalytic activity under UV light irradiation decreased due to shielding of the catalyst active area by excessive amount of dye adsorbed. Conversely, the photocatalytic activity of the Ce and Nd doped TiO2 increased under visible light irradiation by 1.2 times as a result of the dye photosensitization effect. It was demonstrated that two-steps dark adsorption and photocatalytic reaction or one-step simultaneous adsorption and reaction can produce significantly different results for the photocatalytic degradation of dyes in coloured waters, the rate being controlled by the competitive adsorption of the reacting organics and the H2O/OH- species. The reaction is driven by the radical oxygen species (ROS) formed on the catalyst surface the nature of which, differs under UV or visible light irradiation. The Ce-doped TiO2 and Nd-doped TiO2 photocatalysts with 0.5% rare-earth content were found to be efficient in the degradation of MB in aqueous solution, removing the colour and reducing the toxicity of wastewaters.
