Extended drought affects the production and quality of wheat (Triticum aestivum L.), one of the world's most important food crops. Breeding for increased drought resistance is becoming increasingly important due to the rising demand for food production. Four old traditional Croatian wheat cultivars were used in the present study to examine the early antioxidant response of flag leaves to desiccant-stimulated drought stress and to identify drought-tolerant cultivars accordingly. The results indicate that the enzymatic antioxidant system plays the most significant role in the early response of adult wheat plants to drought stress and the removal of excessive H2O2, particularly GPOD and APX. Nada and Dubrava cultivars revealed the strongest activation of the enzymatic defense mechanism, which prevented H2O2 accumulation and lipid peroxidation. Additionally, the Nada cultivar also showed increased synthesis of proline and specific phenolic compounds, which both contribute to the increased stress tolerance. Among the cultivars investigated, cultivar Nada has the broadest genetic base, which may explain why it possesses the ability to activate both enzymatic and non-enzymatic defense mechanisms in an early response to drought stress. This suggests that old traditional wheat cultivars with broad genetic bases can be a valuable source of drought tolerance, which is especially important given the current climate change.

Alfalfa (Medicago sativa L.) is one of the most important forage species and is recently more in focus for human consumption mainly due to its content of bioactive phenolics. Samples of the seventeen alfalfa cultivars/populations were collected at the Agricultural Institute Osijek with the aim to evaluate their forage quality, phenolic profiles, and antioxidant potential. Significant differences (p < 0.05) existed among studied alfalfa in all analyzed traits. The cultivar OS 99 and populations L7 and L20 were characterized by high crude protein content (22.5−24.7%) and the lowest neutral (40.2−42.9%) and acid detergent fibres (33−35.5%). The soluble-free phenolics from alfalfa leaves were extracted by methanol while insoluble cell-wall bound phenolics were released by alkaline hydrolysis. The bound phenolic extract showed a stronger DPPH scavenging capacity (20.8 mg TE/g dm) than the soluble (11.4 mg TE/g dm). The HPLC data revealed that more phenolics were found in the bound (3638.0 µg/g dm) than in the soluble form (912.3 µg/g dm). In the soluble extract of the alfalfa leaves, the major compound was catechin (338.3 µg/g dm), while rutin, epicatechin, and ferulic acid were minor ones. In the bound phenolic extract, the most abundant was ferulic (2198.2 µg/g dm) and p-coumaric acid (983.7 µg/g dm), followed by myricetin, apigenin, and quercetin. The principal component analysis revealed that alfalfa cultivars/populations were better discriminated based on the data on phenolics, rather than on forage quality. The cultivars/populations Florida 66, OS 66, L 40, L 42, Seed Force 4, and Torlesse were the most interesting in terms of phenolic health-promoting characteristics.

The selection of drought-tolerant sour cherry genotypes is essential for developing sustainable fruit production in today's climate-change conditions. The phenotypic heterogenic population of sour cherry Oblacinska, with high and regular yield suitable for mechanical harvesting and industrial processing, is a traditional and predominant cultivar in northern Croatia (Pannonian region) and Serbia commercial orchards. In this context, 2-year old virus-free sour cherry plants of 4 isolated Oblacinska sour cherry ecotypes (OS, 18, D6, and BOR) produced by micropropagation were exposed to severe drought in a greenhouse under semi-controlled conditions to evaluate its photosynthetic intra-varietal variability. Relative water content (RWC), chlorophyll fluorescence (ChlF), and photosynthetic pigments were evaluated during the ten days of the experiment. As a visible symptom of stress, the withering of plants was followed by a diminution of RWC and photosynthetic pigments in the drought exposed leaves of sour cherry ecotypes compared to the control treatment. ChlF elucidated variability in the photosynthetic efficiency within studied sour cherry ecotypes, highlighting PIABS, PItotal, and ψE0 as the most sensitive and thus the most informative JIP parameters for drought screening. Among the investigated ecotypes, BOR proved to be the most sensitive. The Oblacinska sour cherry ecotype OS showed the highest tolerance to drought conditions and, therefore, can be used as a source of tolerance in sour cherry breeding programs.

Global warming and the associated climate change are imposing abiotic stress on plants. Abiotic factors are crucial for plant productivity, survival, and reproduction. Eight sunflower hybrids were tested in conditions of different water availability and with combinations of different temperatures and irradiation. The changes in the photosynthetic efficiency were measured in the morning (control conditions: 2013, 25.8 °C and 349.1 W m-2; 2014, 21.8 °C and 296.4 W m-2) and afternoon (the combination of increased temperatures and high irradiation: 2013, 34 °C and 837.9 W m-2; 2014, 29.4 °C and 888.9 W m-2) at a flowering stage in rainfed or irrigated conditions. The measurement time (morning and afternoon conditions) had a statistically significant effect on all the tested parameters. The performance index (PIABS) in 2013 and the maximum quantum yield of photosystem II (TR0/ABS) in 2014 are the only parameters significantly affected by the irrigation. As a result of the combined effect of increased temperatures and high irradiation, PIABS values decreased by 73-92% in rainfed conditions and by 63-87% in irrigated conditions in 2013, depending on the hybrid, while in 2014, the decrease varied between 70 and 86%. The TR0/ABS decrease was 7-17% in 2013, depending on the hybrid, and 6-12% in 2014, both in rainfed and irrigated conditions. The principal component analysis confirmed the effect of the combination of increased temperatures and high irradiation on hybrids, sorting them exclusively according to the time of measurement. All investigated parameters highly fluctuated between hybrids but without observable trends for the morning and afternoon conditions, as well as for irrigation. Plants' reaction to the combination of increased temperatures and high irradiation manifested as a change in their photosynthetic efficiency, i.e., the photosynthetic apparatus' functioning was impaired.

Genotype-dependent responses of apples to drought stress were evaluated between commercial and traditional apple cultivars. The results indicate different mechanisms of tolerance to investigated drought stress conditions. Chlorophyll fluorescence induction (OJIP) parameters, chlorophyll and carotenoid content, malondialdehyde (MDA), hydrogen peroxide (H2O2), proline, phenols and leaf water content (WC) were measured. The traditional cultivar "Crvenka" confirmed the best tolerance to a drought stress condition, presenting higher photosynthetic efficiency, higher leaf water content, higher levels of chlorophyll content and lower lipid peroxidation with greater membrane stability. The commercial cultivar "Golden Delicious Reinders" showed decreased water content in leaves, increased lipid peroxidation levels and photoinhibition. Considering all results, the commercial cultivar "Golden Delicious Reinders" was adversely affected by drought, while traditional cultivars exhibited better tolerance to drought stress.

Fusarium head blight (FHB) is found in both temperate and semi-tropical regions and causes losses in wheat (Triticum aestivum L.) resulting in reduced yield, deteriorated grain quality, and contamination of the grains with mycotoxins, primarily deoxynivalenol (DON). In this study, we focused on the identification of protein components in Fusarium-inoculated and non-inoculated wheat samples along with the major antioxidant enzymes that Fusarium can encounter during FHB infection process in six winter wheat varieties when FHB symptoms started to occur. Our hypothesis was that wheat antioxidants and H2O2 may play a role against Fusarium infections, consequently preserving protein grain components. Results showed that in more resistant varieties (Olimpija and Vulkan) DON content of inoculated flour was less accumulated and thus the major gluten network was not significantly attacked by Fusarium spp. The maximum increase in guaiacol peroxidase (POD) activity in response to FHB treatment was detected in the FHB-resistant varieties Olimpija and Vulkan, while the lowest increase in H2O2 content was detected in the FHB-susceptible variety Golubica. A particular reduction in the content of both total glutenin and high-molecular-weight glutenin subunits was detected in susceptible wheat varieties after serious artificial infection, along with increased DON accumulation. The defense mechanism in wheat varieties differed, where resistant varieties' protein composition depended on POD activity as a detoxification agent.

Very little is known about the physiological interactions between wheat quality and Fusarium head blight (FHB), which substantially reduces wheat grain yield and quality worldwide. In order to investigate stress-induced changes in flag leaves from plants artificially inoculated with Fusarium, we screened for chlorophyll a fluorescence transient at 1, 2, 4, 7 and 14 days after Fusarium inoculation. Our results indicate that the maximum quantum yield of photochemistry (Fv/Fm) and the performance index (PI) were not affected by FHB, but there were significant differences in those two traits between different varieties and measurement times. FHB caused a significant reduction in the percentage of glutenins (GLU), high-molecular-weight (HMW), and low-molecular-weight (LMW) subunits in 'Kraljica' and 'Golubica', unlike 'Vulkan', where the percentage of GLU increased.

Fusarium head blight (FHB) is a destructive fungal disease of wheat (Triticum aestivum L.) that causes significant grain yield losses and end-use quality reduction associated with contamination by the mycotoxin deoxynivalenol (DON). Three winter wheat varieties ('Vulkan', 'Kraljica' and 'Golubica') were screened for FHB resistance using artificial inoculation technique under field conditions. The aim of this study was to examine a relationship between FHB resistance and the effectiveness of enzyme antioxidant system of wheat varieties under different sampling times (3, 15, 24, 48, 96, 120 and 336 hai). In the time-course experiments FHB-resistant variety 'Vulkan' showed rapid induction of ascorbate peroxidase (APX) and polyphenol oxidase (PPO) activity in the early stages after infection (3 hai) and it seems that in 'Vulkan' FHB-resistance is associated with antioxidative enzymes activity. Moderately FHB resistant variety 'Kraljica' showed the higher guaiacol peroxidase (POD) activity and higher H2O2 content after 24 hai, increased malondialdehyde (MDA) content at the beginning of infection (3, 15 hai) while induction of catalase (CAT), APX and PPO was delayed. FHB-susceptible variety 'Golubica' involved antioxidant enzymes in defense response much later. Based on our results the activity of antioxidant enzymes (APX and PPO) was more pronounced in 'Vulkan' than in FHB-medium resistant variety 'Kraljica' and FHB-susceptible 'Golubica'. The differences in antioxidant response of wheat varieties under Fusarium infestation could be the result of genetic properties.
