AIMS: The aim of this study was to verify whether there is significant relationship between stages of mandibular canine calcification and skeletal maturation and to determine whether the same can be used as a reliable diagnostic aid to assess skeletal maturity. MATERIALS AND METHODS: The sample consisted of 50 males and 50 females aged between 8 and 15 years. Two radiographs namely orthopantomograph and handwrist radiograph were taken for this study. Skeletal age was ascertained from handwrist radiographs according to the method introduced by Fishman L.S. The developmental stages of mandibular canine were assessed according to Demirjian's stages of dental calcification. RESULTS: The relationship between canine stage calcification and skeletal maturity index was statistically significant in all stages. INTERPRETATION AND CONCLUSION: Mandibular canine calcification stages can be recommended as a supplemental diagnostic aid. Similar studies may be conducted in different population to rule out any possible racial and regional influences on growth characteristics.

OBJECTIVE: Bilateral sagittal split osteotomy (BSSO) is a routinely used surgical step for the correction of a class III with mandibular prognathism. Many factors influence the stability of the surgical correction achieved. This study was designed to access the role of growth pattern in the surgical stability after a BSSO correction. MATERIALS AND METHODS: A total of 18 individuals (6 vertical growers, 6 horizontal growers, and 6 normal growing individuals) were considered for the study. Five parameters, horizontal plane (HP)-pogonion (POG) angle, HP-occlusal plane angle, POG height, POG depth, and Point B depth, were measured and compared postsurgically and in the follow-up phase. RESULT: Vertical growing individuals showed greater tendency for relapse and clockwise rotation of mandible postsurgically. No major difference was observed in the normal and horizontal growing individuals. When planning BSSO on vertical growing patient, utmost care should be taken to prevent posttreatment relapse to ensure better surgical stability.

Consequences of a catastrophic flood on the habitat quality and the concurrent responses of the bentho-pelagic community were studied in Cochin estuary, a eutrophic estuary along the southwest coast of India. The episodic flood in 2018 led to a marked decline in the dissolved nutrients and heavy metal concentrations in water and sediments of the estuary. The pre-flood phytoplankton abundance dominated by a bloom-forming species Cerataulina bicornis experienced a significant drop after the flood. Contrarily, zooplankton and macrobenthos responded favorably towards the flood-imposed habitat alterations. Higher susceptibility to heavy metal pollution and increased grazing pressure from gelatinous carnivores restricted the abundance of Copepoda, the dominant zooplankton taxon during pre-flood. The lower heavy metal concentration in the sediment after the flood favored higher macrobenthic abundance and diversity with a conspicuous change in the community structure from opportunistic polychaetes, indicators of pollution to molluscans and crustaceans, indicators of the healthy benthic zones.

BACKGROUND: Very few studies have compared the safety and efficacy of antiplatelets and anticoagulants in the treatment of extracranial carotid and vertebral artery dissection. Our study was aimed at comparing the two types of antithrombotic treatment in extracranial dissection and to study the predictors of outcome in these patients. MATERIALS AND METHODS: Prospective data of 200 consecutive patients with a confirmed diagnosis of extracranial carotid (n = 132) or vertebral (n = 68) artery dissection (76% males; mean age, 43.5 ± 13 years) treated with antiplatelets (n = 136) or anticoagulants (n = 64) were analyzed retrospectively. The presenting symptom was stroke in 74.5%, transient ischemic attack (TIA) in 18.5%, and local symptoms in 7% of the patients. Follow-up was done at three and six months. Primary outcome measures were TIA or stroke and symptomatic intracerebral hemorrhage (SICH) at three months. RESULTS: At the three-month follow-up, 106 (53%) patients had an excellent outcome. Recurrent ischemic events occurred in 7 (3.5%) and SICH in 11 (5.55%) patients. Six (4.41%) patients in the antiplatelet group and 1 patient (1.56%) in the anticoagulant group had recurrent ischemic events (P = 0.434); SICH was more frequent in the anticoagulant group (9.4% vs 3.7%, P = 0.185). On multivariate analysis, significant predictors of a poor three-month outcome were stroke as the presenting event and severity of stroke at onset. CONCLUSIONS: The risk of recurrent ischemic events in carotid and vertebral artery dissection is low and is irrespective of the type of antithrombotic treatment. Stroke as the presenting event and severity of stroke at onset were significant predictors of a poor three-month outcome.

Multiple sequence alignment (MSA) is an integral part of molecular biology. But handling massive number of large sequences is still a bottleneck for most of the state-of-the-art software tools. Knowledge driven algorithms utilizing features of input sequences, such as high similarity in case of DNA sequences, can help in improving the efficiency of DNA MSA to assist in phylogenetic tree construction, comparative genomics etc. This article showcases the benefit of utilizing similarity features while performing the alignment. The algorithm uses suffix tree for identifying common substrings and uses a modified Needleman-Wunsch algorithm for pairwise alignments. In order to improve the efficiency of pairwise alignments, a knowledge base is created and a supervised learning with nearest neighbor algorithm is used to guide the alignment. The algorithm provided linear complexity O(m) compared to O(m2). Comparing with state-of-the-art algorithms (e.g., HAlign II), SPARK-MSNA provided 50% improvement in memory utilization in processing human mitochondrial genome (mt. genomes, 100x, 1.1. GB) with a better alignment accuracy in terms of average SP score and comparable execution time. The algorithm is implemented on big data framework Apache Spark in order to improve the scalability. The application of nanotechnology has created high impact in diagnosis and prognosis of various disorders including cardiovascular diseases. In the present study, we investigated the beneficial effect of selenium incorporated guar gum nanoparticles (SGG) compared to nascent selenium (Se) and guar gum nanoparticles (GGN) against ischemiareperfusion (IR) induced alterations in oxidative phosphorylation and energy metabolism in H9c2 cardiac cells. Ischemia and reperfusion were induced for 1h. The alterations in activities of various complexes (complex 1, II, III and IV) of mitochondrial electron transport chain (ETC), aconitase activity, oxygen consumption rate, and the ATP content were seen. The role of heat shock protein, hypoxia inducible factor-1α and atrial natriuretic factor (ANP) were also analyzed. Then the beneficial properties of various particles like Se, GGN and SGG were evaluated. Among these, SGG treatment (1 and 5ng) was found to be more beneficial compared to other particles. Overall results reveal that SGG nanoparticles are effective in protecting H9c2 cardiac cells from IR injury via improving the efficiency of ETC in H9c2 cells.

Nanotechnology for the treatment and diagnosis has been emerging recently as a potential area of research and development. In the present study, selenium incorporated guar gum nanoparticles have been prepared by nanoprecipitation and characterized by transmission electron microscopy and particle size analysis. The nanoparticles were screened for antioxidant potential (metal chelation, total reducing power and hydroxyl radical scavenging activity) and were evaluated against the cell line based cardiac ischemia/reperfusion model with special emphasis on oxidative stress and mitochondrial parameters. The cell based cardiac ischemia model was employed using H9c2 cell lines. Investigations revealed that there was a significant alteration (P ≤ 0.05) in the innate antioxidant status (glutathione↓, glutathione peroxidase↓, thioredoxin reductase↓, superoxide dismutase↓, catalase↓, lipid peroxidation↑, protein carbonyl↑, xanthine oxidase↑ and caspase 3 activity↑), mitochondrial functions (reactive oxygen species generation, membrane potential, and pore opening) and calcium homeostasis (calcium ATPase and intracellular calcium overload) during both ischemia and reperfusion. For comparative evaluation, selenium, guar gum and selenium incorporated guar gum nanoparticles were evaluated for their protective properties against ischemia/reperfusion. The study reveals that selenium incorporated guar gum nanoparticles were better at protecting the cells from ischemia/reperfusion compared to selenium and guar gum nanoparticles. The potent antioxidant capability shown by the sample in in vitro assays may be the biochemical basis of its better biological activity. Further, the nanodimensions of the particle may be the additional factor responsible for its better effect.

The present study evaluated the antihypertrophic potential of the ethanolic extract of Boerhaavia diffusa (BDE), a well-known edible cardiotonic plant reported in Ayurveda against angiotensin II-induced hypertrophy in H9c2 cardiac myoblast cells. Markers of hypertrophy such as cell size, protein content and the concentrations of atrial natriuretic peptide (ANP) and B-type natriuretic peptide (BNP) were analysed for the confirmation of hypertrophy induction. Angiotensin II (100 nM) caused an increase in cell volume (69·26 (SD 1·21)%),protein content (48·48 (SD 1·64)%), ANP (81·90 (SD 1·22)%) and BNP (108·57 (SD 1·47)%). BDE treatment significantly reduced cell volume, protein content and the concentrations of ANP and BNP (P#0·05) in H9c2 cells. The activity of various antioxidant enzymes and the concentration of reduced glutathione, which was lowered due to hypertrophy, were increased in BDE-treated cells. The BDE treatment also reduced intracellular reactive oxygen species generation, lipid peroxidation and protein carbonyls in cells. In addition,the expression patterns of NF-kb and transforming growth factor b1 were found to be increased during hypertrophy, and their expressions were reduced on BDE treatment. In vitro chemical assays showed that BDE inhibits angiotensin-converting enzyme and xanthine oxidase in a dose-dependent manner with an estimated 50% effective concentration (EC50) value of 166·12 (SD 2·42) and 60·05 (SD 1·54) mg/ml,respectively. The overall results clearly indicate the therapeutic potential of B. diffusa against cardiac hypertrophy, in addition to its nutritional qualities.

Arsenic trioxide (ATO) has been long used as a chemotherapeutic agent because of its significant anticancer property. Unfortunately, the use of ATO is limited due to its cardiotoxic effects. The present study evaluates the protective property of ethanolic extract of Boerhavia diffusa (BDE) against ATO-induced toxicity on various cell organelles in H9c2 cardiomyocytes. The effects of different concentrations of ATO (5, 7.5 and 10 µM) on cell organelles like mitochondria, endoplasmic reticulum (ER), lysosome and actin, generation of reactive oxygen species, antioxidant enzyme status and intracellular calcium overload were evaluated. ATO significantly (P ≤ 0.05) altered mitochondrial transmembrane potential, intracellular calcium level, ER, lysosomal activity and F-actin network in addition to induction of oxidative stress. Co-treatment with BDE protected the cardiomyocytes from the adverse effects of ATO, especially at 5 µM concentration, which was evident from decreased activity of lactate dehydrogenase (5 µM ATO + 20 µg/mL BDE: 6.61 ± 1.97 µU/mL, respective control group: 16.15 ± 1.92 µU/mL), reduced oxidative stress, calcium influx and organelle damage. Results obtained from the present study allow for a better characterization of the effects of ATO on H9c2 myoblasts. In conclusion, our data suggest that cell organelles are also the targets of ATO-induced cardiotoxicity in addition to other reported targets like ion channels, and BDE has the potential to protect the cardiotoxicity induced by ATO.
