This study examined the effects of including live black soldier fly (BSF, Hermetia illucens) larvae in the diet of laying hens on gut microbiota, and the association between microbiota and fearfulness. A total of 40 Bovans White laying hens were individually housed and fed 1 of 4 dietary treatments that provided 0, 10, 20%, or ad libitum daily dietary portions of live BSF larvae for 12 wk. Cecum microbiota was collected at the end of the experiment and sequenced. Behavioral fear responses to novel objects and open field tests on the same hens were compared against results from gut microbiota analyses. The results showed that the bacteria genera Enterococcus, Parabacteroides, and Ruminococcus torques group were positively associated with increased dietary portion of live larvae, while Lactobacillus, Faecalibacterium, Bifidobacterium, Subdoligranulum, and Butyricicoccus were negatively associated with larvae in the diet. Inclusion of larvae did not affect fear behavior, but the relative abundance of Lachnospiraceae CHKCI001 and Erysipelatoclostridium was associated with fear-related behaviors. Further studies are needed to determine whether the change in gut microbiota affects fearfulness in the long-term.

Introduction: Lactobacilli may prevent broilers from colonization with Campylobacter spp. and other gram-negative zoonotic bacteria through lactic acid production and modulation of the intestinal microbiota. This study evaluated the effects of daily intake of Lactiplantibacillus plantarum 256 (LP256) on Campylobacter jejuni (C. jejuni) loads in ceca and feces of C. jejuni challenged broilers, together with the changes in the gut microbiota. Methods: Two experiments were conducted using the broilers Ross 308 (R-308; Experiment 1) for 42 days and Rowan Ranger broilers (RR; Experiment 2) for 63 days. The LP256 strain was administered either via silage inoculated with LP256 or direct supplementation in the drinking water. Concurrently, haylage as a forage similar to silage but without any inoculum was tested. C. jejuni loads in fecal matter and cecal content were determined by plate counts and qPCR, respectively. The cecal microbiota, in response to treatments and the challenge, were assessed by 16S rRNA sequencing. Results and Discussion: Culturing results displayed a significant reduction in C. jejuni colonization (2.01 log) in the silage treatment in comparison to the control at 1 dpi (day post-infection) in Experiment 1. However, no treatment effect on C. jejuni was observed at the end of the experiment. In Experiment 2, no treatment effects on C. jejuni colonization were found to be statistically significant. Colonization load comparison at the peak of infection (3 dpi) to that at the end of the trial (32 dpi) revealed a significant reduction in C. jejuni in all groups, regardless of treatment. Colonization dynamics of C. jejuni in the cecal samples analyzed by qPCR showed no difference between any of the treatments in Experiment 1 or 2. In both experiments, no treatment effects on the cecal microbiota were observed. However, proportional changes in the bacterial composition were observed after the C. jejuni challenge, suggesting that colonization affected the gut microbiota. Overall, the daily intake of LP256 was not effective in reducing C. jejuni colonization in either broiler type at the end of the rearing period and did not cause any significant changes in the birds' cecal microbiota composition.

Osteoporosis and bone fractures are a severe problem for the welfare of laying hens, with genetics and environment, such as housing system, each making substantial contributions to bone strength. In this work, we performed genetic analyses of bone strength, bone mineral density, and bone composition, as well as body weight, in 860 commercial crossbred laying hens from 2 different companies, kept in either furnished cages or floor pens. We compared bone traits between housing systems and crossbreds and performed a genome-wide association study of bone properties and body weight. As expected, the 2 housing systems produced a large difference in bone strength, with layers housed in floor pens having stronger bones. These differences were accompanied by differences in bone geometry, mineralization, and chemical composition. Genome scans either combining or independently analyzing the 2 housing systems revealed no genome-wide significant loci for bone breaking strength. We detected 3 loci for body weight that were shared between the housing systems on chromosomes 4, 6, and 27 (either genome-wide significant or suggestive) and these coincide with associations for bone length. In summary, we found substantial differences in bone strength, content, and composition between hens kept in floor pens and furnished cages that could be attributed to greater physical activity in pen housing. We found little evidence for large-effect loci for bone strength in commercial crossbred hens, consistent with a highly polygenic architecture for bone strength in the production environment. The lack of consistent genetic associations between housing systems in combination with the differences in bone phenotypes could be due to gene-by-environment interactions with housing system or a lack of power to detect shared associations for bone strength.

Rest and sleep are important for the welfare of mammals and birds. A large part of the daily time budget of broiler chickens is taken up by resting behavior and the quality of resting is important. However, in intensive broiler production systems, disruptions of resting behaviors are common. These disruptions of resting behavior could be negative for the health and growth of the birds. This study investigated if artificial brooders that provide a delimited and darker resting place, away from active birds, reduce disruptions of resting behavior compared to a control situation without artificial brooders. Six pens of each treatment were used in the same building, keeping 60 chickens (Ross 308) per pen. The artificial brooders were removed at 21 days of age. Data on disturbances and duration of resting bouts and activity between resting bouts were collected on 20 and 34 days of age. Also, as an indicator of the quality of rest, the animals' cognitive performance was evaluated in a spatial learning test that was performed at 11 days of age. The results showed that birds housed in pens with access to brooders have longer resting bouts (260.7 ± 5.2 vs. 132.8 ± 5.3s, p < 0.001) and are less likely to be disturbed during resting by other individuals (0.15 vs. 0.48, p < 0.001). The effect of the artificial brooders on both the duration of resting bouts and the proportion of disturbances remained after the removal of the brooders at 21 days of age. The duration of activity between resting bouts was shorter if the resting bout was ended by a disturbance (9.98 ± 1.0 vs. 61.0 ± 2.4s, p < 0.001). Birds reared with brooders were more likely to solve the spatial learning task (0.5 vs. 0.27, p < 0.01), but those succeeding were not faster at solving it. Broilers may be exposed to disrupted rest due to the lack of a dedicated resting place separated from areas with high activity. Using artificial brooders reduces disturbances but does not eliminate them. Therefore, additional changes to the housing conditions or management will be needed to prevent disturbances.

Hatching concepts such as on-farm hatching provide an opportunity to supply newly hatched chickens with optimal nutrition that support growth and development of a healthy gut. Brown algae contain bioactive compounds, especially laminarin and fucoidan that may improve intestinal health and immune responses. This study aimed to examine the effects of early access to feed and water posthatch and feed supplementation with algal extract rich in laminarin from Laminaria digitata, on growth performance, organ and microbiota development and antibody production. A total of 432 Ross 308 chicks were allotted to 36 rearing pens in a 2 × 3 factorial design with two hatching treatments and three dietary treatments. During chick placement, half of the pens were directly provided access to feed and water (Early) while half of the pens were deprived of feed and water for 38 h (Late). The chicks were fed three different starter diets until day 6; a wheat-soybean meal-based control diet, a diet with low inclusion of algal extract (0.057%) and a diet with high inclusion of algal extract (0.114%). Feed intake and BW were registered on pen basis at placement, days 1, 6, 12, 19, 26, 33 and 40. To induce antibody responses, all chicks were vaccinated against avian pneumovirus on day 10. Three chicks per pen were selected as focal animals and used for blood sampling on days 10 and 39. On days 6, 19, and 40, two birds per pen were killed and used for organ measurement and caecal digesta sampling for gut microbiota analysis using the Illumina Miseq PE 250 sequencing platform. Results showed that algal extract did not influence gut microbiota, gut development or vaccine-induced antibody responses. However, during the first 38 h, early-fed chicks consumed on average 19.6 g of feed and gained 27% in BW, while late-fed chicks lost 9.1% in BW which lowered BW and feed intake throughout the study (P < 0.05). Late chicks also had longer relative intestine, higher relative (g/kg BW) weight of gizzard and proventriculus but lower relative bursa weight on day 6 (P < 0.05). No effects of hatching treatment on microbiota or antibody response were detected. The microbiota was affected by age, where alpha diversity increased with age. In conclusion, this study showed that early access to feed but not algal extract improved the growth performance throughout the 40-day growing period, and stimulated early bursa development.

The use of insects in animal feed has the potential to reduce the demand for soybean production and reduce the deforestation and loss of natural resources. In particular, the black soldier fly (BSF, Hermetia illucens) larvae have received attention due to their ability to convert organic waste into high-value biomass. Several studies have investigated the effects of providing BSF larvae to both broilers and laying hens. However, knowledge gaps regarding hens' voluntary intake of live larvae and the effects of larvae consumption on egg production still remain. Therefore, the aim of the present study was to determine the effects of the consumption of 4 different amounts of live BSF larvae on laying hen feed consumption, hen health and fearfulness, and egg production and quality. To this end, 40 Bovans White laying hens were housed individually and provided with 0, 10, 20% or ad libitum daily portions of live larvae from 18 to 30 wk of age. The larvae consumption and concentrate consumption, hen weight, egg production, and egg quality were monitored. Overall, differences were found between the hens given ad libitum access to larvae compared to the other treatments. Ad libitum hens, consumed 163 ± 41 g larvae/hen/day, consumed less concentrate (P = 0.03) and gained more weight (P = 0.0002) than all other treatments. They also had an overall higher consumption of protein, fat and energy (P < 0.03). There was no effect of larvae consumption on egg production, egg weight, shell thickness, shell breaking strength, or Haugh unit (P > 0.05). There was also no effect on hen behavior toward a novel object or in an open field test. This study is the first to provide different amounts of live BSF larvae, including an ad libitum portion to laying hens. The 20% diet could promote sustainability in the egg industry and be economically advantageous if BSF larvae can be bought in bulk for less than 40% of the cost of the concentrate.

Artificial commercial lighting used in animal production facilities can have negative influences on visual abilities, behaviour and welfare of domestic fowl. This study examined the effects of natural-derived light spectrums on behaviour, production and welfare of laying hens reared from hatching into adulthood. Comparisons were made of frequency of a range of behaviours associated with activity, aggression and comfort in birds kept in control light (commercial standard), daylight (full spectrum, including ultraviolet (UV) wavelengths and forest light (forest understorey, including UV). In addition, bird preferences for different lights, feather damage and egg production were monitored. The results showed that the behavioural repertoire of birds changed with age, while the effects of light treatment were subtle. Some evidence was found that birds preferred either daylight or forest light to control light, suggesting that inclusion of UV contributed to the preference. Daylight and forest light were associated with more active behaviours, and daylight with better plumage and later start of lay. Thus natural-like light may have beneficial effects on domestic fowl, but the differences between broad-spectrum light sources are rather small.

A total of 70 broiler chickens were used to evaluate the apparent ileal digestibility coefficient (AIDC) of protein and amino acids, and apparent digestibility coefficient (ADC) of energy in the protein rich Fungal Biomass of Neurospora intermedia (FBN) obtained from bioethanol production. The chickens were housed in 10 pens with seven chickens per pen and fed one of two experimental diets between day 28 and 35 of age. The experimental diets were wheat-soybean meal-based control diet and a diet composed of 70% control diet and 30% of FBN. There were no difference (P > 0.05) between the control and FBN diet on chick feed intake and body weight at day 35. However, the AIDCs of crude protein and amino acids were significantly (P < 0.01) higher in control diet than in the FBN diet except for proline and phenylalanine. The AIDC of CP (0.74), cysteine (0.68), methionine (0.70), AME (15.6), and threonine (0.69) in FBN were comparable to the corresponding values in other protein-rich feedstuffs such as soybean meal and fish meal. The results from this study show that FBN may be an alternative protein source for poultry.

BACKGROUND: During the production period of laying hens, the number of cracked eggshells increases and the skeleton becomes brittle. Both these problems are related to ageing of the hen and cause economic problems for egg producers and impaired animal welfare. This study investigated key factors in the shell gland and duodenum related to eggshell quality and bone strength in laying hens during the production period. Five Lohmann Selected Leghorn (LSL) and five Lohmann Brown (LB), common hybrids in commercial egg production, were euthanized at 21, 29, 49 and 70 weeks (wk) of age. Blood samples for analysis of total calcium were taken at euthanization. Right femur and humerus were used for bone strength measurements and tissue samples from shell gland and duodenum were processed for morphology, immunohistochemical localisation of oestrogen receptors (ERα, ERß), plasma membrane calcium ATPase (PMCA) and histochemical localisation of carbonic anhydrases (CA). Eggs were collected for shell quality measurements. RESULTS: At age 49 week, shell and bone strength had both deteriorated, but the hens were then able to maintain the level until 70 week of age and femur bone strength even improved. The main physiological findings associated with the effects seen at 49 week were reduced gland density and a shift in balance between ERα and ERß in the shell gland, which coincided with a reduction in CA activity in the duodenum. Somewhat surprisingly, capillary density and capillaries with CA activity both increased in the shell gland over time, the latter possibly mediated via ERß. These findings were independent of hybrid. PMCA was found in both shell gland and duodenum, but appeared unrelated to the age-related changes in shell and bone quality. CONCLUSIONS: In hens around half-way through the production period, both shell quality and bone strength had deteriorated. Decreased gland density and a shift in the balance between ERα and ERß in the shell gland, co-occurring with a dramatic drop in duodenal CA activity, are suggested as possible factors involved in age-related changes in shell and bone quality.

INTRODUCTION: HANS KAI is a unique health promotion intervention to improve participants' health by focussing on interrelated chronic disease prevention behaviours through peer support and strengthening of social support networks. The study objective was to determine the effectiveness of HANS KAI in an urban Canadian setting. METHODS: We used a mixed methods intervention research design that involved multiple sites from November 2010 to April 2015. Data was obtained from participant surveys as well as in-person interviews at zero, 6, 12 and 24 months. Participants met in groups at least once a month during the research period, to self-monitor health indicators, prepare and share a healthy snack, participate in a physical activity, set a healthy lifestyle goal (optional) and socialize. RESULTS: There were statistically significant mental health improvements from pre- to post-program, and 66% of the participants described specific behaviour changes as a result of HANS KAI participation. Additional positive health impacts included peer support; acquiring specific health knowledge; inspiration, motivation or accountability; the empowering effect of monitoring one's own health indicators; overcoming social isolation and knowing how to better access services. CONCLUSION: The need to identify innovative ways to address chronic disease prevention and management has been the driver for implementing and evaluating HANS KAI. While further research will be required to validate the present findings, it appears that HANS KAI may be an effective approach to create environments that empower community members to support each other while promoting healthy lifestyle choices and detecting early changes in health status.


INTRODUCTION: Le modèle HANS KAÏ est une intervention de promotion de la santé originale qui vise à améliorer la santé des participants en ciblant les comportements interdépendants de prévention des maladies chroniques grâce au soutien par les pairs et au renforcement des réseaux de soutien social. L'objectif de notre étude était de mesurer l'efficacité du modèle HANS KAÏ en contexte urbain canadien. MÉTHODOLOGIE: Nous avons utilisé un modèle de recherche interventionnelle à méthodes mixtes appliqué à plusieurs sites entre novembre 2010 et avril 2015. Les données ont été recueillies au moyen de sondages auprès des participants et d'entrevues en personne au début de l'étude puis à 6, 12 et 24 mois. Les participants se sont réunis en groupes au moins une fois par mois durant la période de recherche afin de surveiller eux-mêmes leurs indicateurs de santé, de préparer et de partager une collation saine, de participer à une activité physique, d'établir un objectif en matière de mode de vie sain (facultatif) et d'avoir une activité de convivialité. RÉSULTATS: Des améliorations statistiquement significatives de la santé mentale ont été constatées avant et après le programme, et 66 % des participants ont fait état de changements de comportement précis résultant de la participation au projet HANS. D'autres effets positifs sur la santé ont été constatés : un soutien par les pairs, l'acquisition de connaissances précises en matière de santé, l'inspiration, la motivation et la prise de responsabilité, l'autonomisation découlant du suivi de ses propres indicateurs de santé, la possibilité de rompre l'isolement social et enfin une meilleure connaissance de la façon d'avoir accès aux services. CONCLUSION: C'est la nécessité de trouver des moyens innovants pour favoriser la prévention et la gestion des maladies chroniques qui a motivé la mise en oeuvre et l'évaluation du modèle HANS KAÏ. Bien que d'autres recherches soient nécessaires pour valider nos résultats, il semble que le modèle HANS KAÏ puisse être efficace pour créer un contexte permettant à la population locale de s'entraider en favorisant des choix de vie sains et en détectant de façon précoce les changements d'état de santé.

BACKGROUND: Cracked shells, due to age related reduction of shell quality, are a costly problem for the industry. Parallel to reduced shell quality the skeleton becomes brittle resulting in bone fractures. Calcium, a main prerequisite for both eggshell and bone, is regulated by estrogen in a complex manner. The effects of estrogen, given in a low continuous dose, were studied regarding factors involved in age related changes in shell quality and bone strength of laying hens. A pellet containing 0.385 mg estradiol 3-benzoate (21-day-release) or placebo was inserted subcutaneously in 20 birds each of Lohmann Selected Leghorn (LSL) and Lohmann Brown (LB) at 70 weeks of age. Eggs were collected before and during the experiment for shell quality measurements. Blood samples for analysis of total calcium were taken three days after the insertion and at sacrifice (72 weeks). Right femur was used for bone strength measurements and tissue samples from duodenum and shell gland were processed for morphology, immunohistochemical localization of estrogen receptors (ERα, ERß), plasma membrane calcium ATPase (PMCA) and histochemical localization of carbonic anhydrase (CA). RESULTS: Estrogen treatment increased shell thickness of both hybrids. In addition, shell weight and shell deformation improved in eggs from the brown hybrids. The more pronounced effect on eggs from the brown hybrid may be due to a change in sensitivity to estrogen, especially in surface epithelial cells of the shell gland, shown as an altered ratio between ERα and ERß. A regulatory effect of estrogen on CA activity, but not PMCA, was seen in both duodenum and shell gland, and a possible connection to shell quality is discussed. Bone strength was unaffected by treatment, but femur was stronger in LSL birds suggesting that the hybrids differ in calcium allocation between shell and bone at the end of the laying period. Plasma calcium concentrations and egg production were unaffected. CONCLUSIONS: A low continuous dose of estrogen improves shell strength but not bone strength in laying hens at the end of the laying period.

The ability to perceive rapid movement is an essential adaptation in birds, which are involved in rapid flight, pursuing prey and escaping predators. Nevertheless, the temporal resolution of the avian visual systems has been less well explored than spectral sensitivity. There are indications that birds are superior to humans in their ability to detect movement, as suggested by higher critical flicker frequencies (CFFs). It has also been implied, but not properly tested, that properties of CFF, as a function of light intensity, are affected by the spectral composition of light. This study measured CFF in the chicken, Gallus gallus L., using four different light stimuli - white, full-spectrum (white with addition of UV), yellow (590 nm) and UV (400 nm) - and four light intensity levels, adjusted to relative cone sensitivity. The results showed significantly higher CFF values for full-spectrum compared with white light, as well as a steeper rate of increase with intensity. The presence of UV wavelengths, previously demonstrated to affect mate choice and foraging, appears to be important also for detection of rapid movement. The yellow and UV light stimuli yielded rather similar CFFs, indicating no special role for the double cone in flicker detection.

BACKGROUND: Automated hematology instruments commonly are used for mammalian blood analysis, but there is a lack of accurate automated methods available for avian leukocyte analysis. OBJECTIVE: The aim of this study was to validate differential leukocyte counts in blood from chickens using the Cell-Dyn 3500 hematology system and avian-specific software. METHODS: Blood samples were collected in lithium-heparin tubes from 2 groups (n = 84 and n = 139) of laying hens. Manual 200-cell differential counts were done on routinely-stained blood smears, and manual total granulocyte counts (heterophils and eosinophils) were done using an eosinophil stain in a counting chamber. Automated differential counts were done using VET 2.3, a research and development version of avian-specific software for the Cell-Dyn 3500. Results were analyzed using Pearson's correlation and difference plots. RESULTS: Automated granulocyte counts from the Cell-Dyn were in good agreement with manual granulocyte counts (r = 0.93 and 0.80 for the 2 study groups). No correlation was found between automated and manual lymphocyte counts. Correlation coefficients for monocyte counts were 0.70 and 0.43. CONCLUSION: Automated leukocyte results from the Cell-Dyn using VET 2.3 software were not fully accurate. Total granulocyte counts may be of clinical usefulness, but results obtained for other parameters were unreliable.
