RESUMO

To explore the changes and the reaction mechanisms between soil microecological environment and the content of secon-dary metabolites of plants under water deficit, this study carried out a pot experiment on the 3-leaf stage seedlings of Rheum officinale to analyze their response mechanism under different drought gradients(normal water supply, mild, moderate, and severe drought). The results indicated that the content of flavonoids, phenols, terpenoids, and alkaloids in the root of R. officinale varied greatly under drought stresses. Under mild drought stress, the content of substances mentioned above was comparatively high, and the content of rutin, emodin, gallic acid, and(+)-catechin hydrate in the root significantly increased. The content of rutin, emodin, and gallic acid under severe drought stress was significantly lower than that under normal water supply. The number of species, Shannon diversity index, richness index, and Simpson index of bacteria in the rhizosphere soil were significantly higher than those in blank soil, and the number of microbial species and richness index decreased significantly with the aggravation of drought stresses. In the context of water deficit, Cyanophyta, Firmicutes, Actinobacteria, Chloroflexi, Gemmatimonadetes, Streptomyces, and Actinomyces were the dominant bacteria in the rhizosphere of R. officinale. The relative content of rutin and emodin in the root of R. officinale was positively correlated with the relative abundance of Cyanophyta and Firmicutes, and the relative content of(+)-catechin hydrate and(-)-epicatechin gallate was positively correlated with the relative abundance of Bacteroidetes and Firmicutes. In conclusion, appropriate drought stress can increase the content of secondary metabolites of R. officinale from physiological induction and the increase in the association with beneficial microbe.

The extensive use of traditional cooking stoves to meet daily cooking and heating requirements has highlighted the serious problem of indoor and outdoor air pollution. This study evaluates seven improved coal-fired space-heating and cooking stoves and compares them with a widely used stove of an older design, selected as a baseline reference. The seven stoves were selected from a range of candidate improved stoves submitted by manufacturers for testing as part of the air quality improvement in the Hebei Clean Air Project, Hebei Province, China. Stove performance was evaluated when burning raw coal and coal briquettes during the high and low power stages respectively. All seven improved cooking stoves surpassed the baseline stove in combined heating and cooking thermal and emission performance. Among the improved cooking stoves, Model 2-TL was found to have the highest average thermal efficiency, 87.2 ± 0.5%, when burning coal briquettes at high and low power. The lowest emission of PM2.5 was 0.94 ± 0.5 mg MJNET -1, CO 0.55 ± 0.28 g MJNET -1, and CO/CO2 1.1 ± 0.6%, respectively. It is concluded that the use of these improved heating and cooking stoves should be promoted for daily cooking and heating requirements. This strategy will not only save fuel to the benefit of the household, but widespread adoption could contribute to significant reductions of CO and PM2.5 emissions in Hebei Province.

Celangulin I is an insecticidal component isolated from Chinese bittersweet Celastrus angulatus. The present study explored the possible effects of celangulin I on the calcium signaling pathway, especially on the L-type Ca(2+) channel and the calcium channels in the endoplasmic reticulum in the central neurons isolated from the third instar larvae of Spodoptera exigua using whole-cell patch-clamp and calcium imaging technique. The results showed that celangulin I could activate the high voltage-gated calcium channel at the concentration of 150µM. The peak currents were increased by 17% of the initial value at the end of the 10-min recording after treated with celangulin I. The rises of intracellular calcium ion concentration ([Ca(2+)]i) in neurons treated by celangulin I showed that the effects of celangulin I were concentration-dependent. Activation of the RyRs by ryanodine decreased the calcium release induced by celangulin I, indicating that celangulin I exerts effect on insect RyRs. Furthermore, we also provided evidence for the first time that celangulin I activates inositol 1,4,5-trisphosphate (IP3) sensitive intracellular calcium release channels in the endoplasmic reticulum third instar larvae neurons of S. exigua. Plausibly, these experimental results can explain the characteristic symptoms of anesthesia and paralysis in celangulin I treated insects.

A series of compounds containing dihydroquinazolinone moiety was designed and synthesized. Amine bridge part was changed in comparison with known anthranilic diamides insecticides. Their insecticidal activities against oriental armyworm (Mythimna separata) indicated that most of the compounds showed moderate to high activities at the tested concentrations. In particular, compounds 5a and 5k showed 80% larvicidal activities against oriental armyworm at the concentration of 5mg/L. The present study also explored the possible effects of target compounds on the high voltage-gated calcium channel and the calcium channels in the endoplasmic reticulum in the central neurons isolated from the third instar larvae of Spodoptera exigua using whole-cell patch clamp and calcium imaging technique. The results showed that compound 5a activated the high voltage-gated calcium channel in the central neurons of S. exigua weakly. The peak currents only increased by 6% of the initial value at the end of the 10-min recording after treated with 0.22µM 5a, while chlorantraniliprole has an opposite effect. The effects of 5a on the intracellular calcium ion concentration ([Ca(2+)]i) in neurons were well investigated. The experimental results indicated that these novel compounds have different mechanism compared with chlorantraniliprole.
