RESUMO

Imaging and serological approaches play an important role in the diagnosis and treatment of alveolar echinococcosis; however, they also suffer from some problems during their applications in clinical practices, which urges the identification of potential diagnostic markers. Novel serological, genomics and proteomics diagnostic markers alone or in combination may increase the sensitivity and specificity in early diagnosis of alveolar echinococcosis, which play vital roles in monitoring of disease courses and prognostic evaluation. This review mainly presents the advances in the studies on novel diagnostic markers for alveolar echinococcosis.

OBJECTIVE: Accumulating evidence determined that lncRNA plays important roles in the development and occurrence of cancers. Prostate cancer is the second most common type of cancer and one of the top five cancers for the cause of male death in the world. Therefore, this study was to explore the regulatory mechanism of lncRNA in chemoresistance of PC. MATERIALS AND METHODS: qRT-PCR was used to detect the mRNA expression of FEZF1-AS1, miR-25-3p and ITGB8. Western blot was applied to measure the protein expression of ITGB8 E-cadherin, N-cadherin, Vimentin, LC3I, LC3II, ATG5 and Beclin-1. In addition, CCK-8 assay was used to assess cell proliferation of transfected cells. Luciferase reporter assay and RIP assay were used to determine the relationship among FEZF1-AS1, miR-25-3p and ITGB8. RESULTS: In this study, the expression of FEZF1-AS1 and ITGB8 was upregulated, whereas the expression of miR-25-3p was downregulated in PC tumor tissues and PC/PTX cells. Luciferase reporter assay and RIP assay determined that miR-25-3p was a target of FEZF1-AS1 and ITGB8 was a target mRNA of miR-25-3p. Interestingly, knockdown of FEZF1-AS1 could inhibit cell viability and EMT and promoted cell autophagy in PC/PTX cells, but inhibition of miR-25-3p or promotion of ITGB8 could reverse the effects of si-FEZF1-AS1 on PC/PTX cells. CONCLUSIONS: In this study, we found that lncRNA FEZF1-AS1 promoted chemoresistance, autophagy and epithelial-mesenchymal transition (EMT) through regulation of miR-25-3p/ITGB8 axis in PC, providing a new regulatory mechanism of PC and a novel therapeutic target.

OBJECTIVE: To investigate the correlations of MC4R and MSH2 with adult obesity, a total of 46 patients with early-stage colon cancer were treated in our hospital between February 2008 and February 2009 and were enrolled. PATIENTS AND METHODS: Venous blood was regularly drawn from subjects of the observation group and 48 healthy subjects for 6 years. Expression levels of MC4R and MSH2 genes were tested using quantitative polymerase chain reaction analyses, and the ensuing proteins were determined using enzyme-linked immunosorbent assays and Western blotting and immunohistochemical analyses. Finally, correlations with body mass index (BMI) and the presence of colon cancer were identified using multivariate analyses. RESULTS: Compared with the control group, MSH2 mRNA and protein expression increased significantly over time (p < 0.05) in patients with colon cancer. Moreover, MSH2 expression was correlated with colon cancer progression, and MC4R mRNA and protein expression increase concurrently in comparison with the control group (p < 0.05). Also, the mean BMI among patients with colon cancer was 30.8, whereas that among control subjects was only 21.4. These data indicate a relationship between BMI and colon cancer. CONCLUSIONS: Expression of MSH2 in patients with colon cancer may promote the expression of the obesity gene MC4R, potentially contributing to body weight gains.

Taurine, which is found at high concentration in the heart, exerts several protective actions on myocardium. Physically, the high level of taurine in heart is maintained by a taurine transporter (TauT), the expression of which is suppressed under ischemic insult. Although taurine supplementation upregulates TauT expression, elevates the intracellular taurine content and ameliorates the ischemic injury of cardiomyocytes (CMs), little is known about the regulatory mechanisms of taurine governing TauT expression under ischemia. In this study, we describe the TonE (tonicity-responsive element)/TonEBP (TonE-binding protein) pathway involved in the taurine-regulated TauT expression in ischemic CMs. Taurine inhibited the ubiquitin-dependent proteasomal degradation of TonEBP, promoted the translocation of TonEBP into the nucleus, enhanced TauT promoter activity and finally upregulated TauT expression in CMs. In addition, we observed that TonEBP had an anti-apoptotic and anti-oxidative role in CMs under ischemia. Moreover, the protective effects of taurine on myocardial ischemia were TonEBP dependent. Collectively, our findings suggest that TonEBP is a core molecule in the protective mechanism of taurine in CMs under ischemic insult.

RESUMO

The automatic recognition of pulse images is the key in the research of computerized pulse diagnosis. In order to automatically differentiate the pulse patterns by using small samples, a fuzzy neural network to classify pulse images based on the knowledge of experts in traditional Chinese pulse diagnosis was designed. The designed classifier can make hard decision and soft decision for identifying 18 patterns of pulse images at the accuracy of 91%, which is better than the results that achieved by back-propagation neural network.

RESUMO

This paper proposes a new algorithm to classify pulse waveforms based on discrete wavelet network. This paper selects 4-order discrete Daubechies wavelet as the wavelet node of this wavelet network to classify six pulse patterns distinctive in shape. 600 pulse records are used to train this wavelet network and 300 pulse records are used to test the classifier's performance. The test results show that this approach has 83% agreement rate with the experienced experts. Compared with traditional classification methods, it needs not the experience in feature extraction.

Neutrophils are very abundant, short-lived leucocytes and their death by apoptosis is central to homoeostasis and the resolution of inflammation, yet the trigger for apoptosis is still a topic of debate. Depolarization of the mitochondrial membrane has been supposed to initiate neutrophil spontaneous apoptosis, as neutrophils gradually lose the anti-apoptotic protein Mcl-1 and Bax translocates and inserts into the mitochondrial membrane. However, other reports show that caspase 8 is required for neutrophil apoptosis, suggesting the involvement of DR (death receptor) signalling. As DR ligation is not required for neutrophil apoptosis, this raises the intriguing possibility that activation of caspase 8 during neutrophil apoptosis occurs via a novel mechanism. In the present paper, we discuss the current evidence for mechanisms occurring in neutrophil apoptosis, which could trigger DR signalling in the absence of DR ligation.

AIM: To observe the polymorphism of angiotensin-converting enzyme (ACE) gene in Northern Hans, Dahurs, and Ewenkis of China. METHODS: The polymerase chain reaction was used to type the insertion/deletion polymorphism at intron 16 of ACE gene among 90 Northern Hans, 84 Dahurs, and 64 Ewenkis individuals. The experiment displayed the distribution in three kinds of ACE genotype: ID (heterozygotes of insertion and deletion), DD (homozygotes of deletion), and II (homozytes of insertion). RESULTS: In Northern Hans the percentages of the distributing ACE I/D genotype were ID 27.8 %, DD 17.8 %, and II 54.4 %. The I/D genotype frequency of Dahurs individuals were ID 60.7 %, DD 26.2 %, and II 13.1 %. The Ewenkis genotype frequency were ID 70.3 %, DD 21.9 %, and II 7.8 %. CONCLUSION: The polymorphism of ACE gene of Northern Hans is different from that of Dahurs and Ewenkis in China.

To search for an effective drug to cure patients with chronic hepatitis B. Twenty eight patients with chronic hepatitis B were treated with prostaglandin E1. The comprehensive indexes, including glutamic pyruric tranasaminase (GPT), serum total bilirubin (TBIL), total bile acid (TBA), hyaluronic acid (HA) and precollagen type III (PCIII), were examined before and after treatment. The levels of GPT, TBIL, TBA, HA, PCIII after 1 month of the treatment were significantly lower than those before the treatment (P < 0.05), there were significant differences between the treated and the controlled group patients with chronic hepatitis B (P < 0.05). The results suggest that the treatment with PGE1 might improve hepatic function, and resist the hepatic fibrosis in patients with chronic hepatitis B.

Neutrophils play a central role in host defense and are recruited in vast numbers to sites of infection where they phagocytose and kill invading bacterial pathogens. Neutrophils have a short half-life that is extended at the inflamed site by pro-inflammatory cytokines and contact with bacterial cell walls. Normal resolution of inflammation involves the removal of neutrophils and other inflammatory cells by the induction of apoptosis. Spontaneous neutrophil apoptosis does not require Fas ligation, but is mediated by caspases 3, 8 and possibly caspase 9 and also involves activation of protein kinase C-delta. With chronic inflammatory disease, neutrophil apoptosis is delayed by pro-inflammatory cytokines, leading to persistence of neutrophils at the inflamed site and non-specific tissue damage. Here we discuss the evidence for inhibition of neutrophil apoptosis via signaling though PI-3-kinase and downstream pathways, including PDK-1 and PKB. Therapeutic strategies to resolve chronic inflammation could therefore usefully target neutrophil apoptosis and the PI-3-kinase or PKC-delta signaling pathways.

In order to find an effective drug to cure patients with chronic hepatitis B, cordyceps sinensis had been used to treat 25 patients with chronic hepatitis B. The comprehensive index, including T lymphocyte subsets (CD4, CD8), hyaluronic acid(HC) and precollagen type III(PC III), were observed before and after treatment. After 3 months of treatment, CD4 and CD4/CD8 ratio increased significantly(P < 0.05), while HA and PC III decreased significantly(P < 0.05) compared with the control. The results suggest that the beneficial effects might be obtained by using cordyceps sinensis to adjust the T lymphocyte subsets level and to treat hepatic fibrosis on patients with chronic hepatitis B.

In investigations of the process of bone destruction caused by chronic otitis media complicated with cholesteatoma, we proposed previously a hypothesis to explain its mechanism. In the present study, we substitute a synthetic auditory ossicle (Apaceram) for the bone to simplify the model system of experiments for our hypothesis. Its process was studied on a model simulation in vitro, in vivo and clinically at molecular level, using laser-Raman spectrometry. An increase of conductivity in both the saline solution and double-distilled water immersed Apaceram indicated demineralization depending on time lapse. The process of demineralization was revealed by the Raman spectral profiles as shown by narrowing a half-peak breadth (p < 0.01) of v1 signal (PO4(3-), 960 cm-1) and the disappearance of the shoulder at circa 950 cm-1 on the Apaceram surface implanted for 6 months. On the other hand, the process of remineralization was revealed in vivo by the spectral profiles: 1) a broader half-peak breadth (p < 0.01) of v1 signal on the Apaceram surface implanted for 10 months than that implanted for 6 months; 2) a weak reappearance of the shoulder at ca. 950 cm-1 on the Apaceram surface after 10 months; 3) four signals (v1 through v4) of PO4(3-) on the HOAP observed for inside the Apaceram shaft and 4) a weak v1 signal on both the granular fluorescent substance in the clinical case and the high density area of subcutaneous tissue after contact with the Apaceram in rats for 3 months. Judging from these investigations, it is proposed that the mechanism of bone destruction associated with cholesteatoma is a form of de- and remineralization.

RESUMO

The computed tomographic (CT) images of 52 patients of small peripheral lung nodule < 3 cm proved pathologically were reviewed. Air bronchogram (AB) was found in 14 nodules on thin-section CT images, in which well or moderately differentiated small adenocarcinomas were only seen with lipidic growth. Squamous-cell, large-cell carcinomas and small benign lung nodules had no AB sign both on CT image and in pathologic specimens.

With the development of the in vitro testing of heart valves, the standardization of the test methods becomes increasingly important and they should also be improved continuously. This paper discusses the problems of waveform comparison and average value dispositions. In the pulsatile model driven by pneumatics, the pressures before and after the valve, and the flow through it, are measured as three one-dimensional variates. The mean values are calculated according to the FDA and the ISO. A comparison and analysis of experimental waveforms indicate that, for basically the same ranges of pressures and flow rate, the flow curves of different types of valve are clearly different. The mean values and the waveforms in the time domain should be taken into consideration synthetically so that the pusatile characteristics of the valve can be more completely reflected. Using numerical filtering methods to treat the waveforms allows for better comparisons between the measured results taken from the different devices. By means of the constellation graphical method for treating mean values as multivariates, it is feasible to classify the valves and to judge their qualities under conditions of pulsatile flow.

The differences of serum lipid and lipoprotein (LP, profiles of animals susceptible (rabbit) and nonsusceptible (Beijing duck) to atherosclerosis as well as the distribution of apolipoprotein (apo) A-I and its catabolism in vivo were studied. Eight items, i.e. total cholesterol (TC), high density lipoprotein (HDL) cholesterol, triglyceride, percentage of alpha-LP and beta-LP, beta-LP concentration, lecithin cholesterol acyl transferase (LCAT), and agarose and polyacrylamide gel electrophoresis of both species were assayed before and after feeding a high fat, high cholesterol diet. Results indicate that the exogenous cholesterol consumed by Beijing ducks was carried and transported by HDL, while that in rabbits was transported by low density lipoprotein (LDL). The biological half lives of apo A-I in serum and in HDL were 41.1 +/- 2.4 and 42.8 +/- 1.7 h respectively, and its distribution in different organs was in the order of liver greater than kidney greater than spleen greater than lung greater than heart greater than intestine greater than muscles greater than aorta. These results show that the liver is the major organ for metabolizing HDL apo A-I, and the kidney is also important. The results also imply that in Beijing ducks the cholesterol carried by HDL may be catabolized through apo A-I receptors in the liver and kidney. The differences in cholesterol binding and in lipoprotein and apolipoprotein metabolism of the two species provide important clues for the elucidation of the pathogenesis of atherosclerosis.

A murine model system for observing the effect of Astragalus Membranaceus (AM) on experimental myocarditis caused by Coxsackie B-3 virus (CB3V) was developed in 4-week-old male BALB/C mice. Gross, histopathologic and ultrastructural examinations of the infected-AM treated group showed that the severity and involved area of the myocardial lesions became milder and smaller than those in the infected-NS treated mice. The total lesion area, and the total lesion area/total myocardial area examined (%) and virus titer in the former group were also smaller and lower than those in the latter group. The results suggest that AM is effective in the inhibition of Coxsackie B virus propagation and protection of myocardium in mouse myocarditis.
