RESUMO

Perfluorocaproic acid (PFHxA), a short-chain substitute for the emerging contaminant perfluorinated compounds, has been detected in the aquatic environment. However, its aquatic toxicity and health risk assessment are mainly unknown. In this study, we compared the toxic doses of 0 mg/L, 5 mg/L, 15 mg/L, 45 mg/L and 135 mg/L on the pathological damage to tissue sections, antioxidant indexes and inflammatory factor expressions in liver, spleen, kidney, Prosogaster, Mid-gut, Hid-gut as well as the changes of IgM, C3, C4, LZM, GOT, GPT in serum of crucian carp. We determined the response of the intestinal microbial community to PFHxA stress by 16S. The results showed that the growth performance of crucian carp was slowed with the increase of PFHxA dose, which caused different degrees of damage to the tissues. Meanwhile, the indexes of SOD, GSH-Px, T-AOC, ACP, AKP and LZM in each tissue were reduced, and the indexes of IgM, C3, C4 and LZM in serum were reduced. The levels of MDA, GOT and GPT in tissues and GOT and GPT in serum were promoted. In addition, IL-1ß, TNF-α, NF-KB, and KEAP-1 in each tissue increased compared with the control group. The levels of IL-10, Nrf2, CAT, and GPx were decreased. The 16S rRNA gene sequencing results showed that PFHxA significantly reduced the abundance and diversity of the gut microbiota. It is suggested that PFHxA is likely to cause different degrees of damage to various tissues by disrupting the diversity of the intestinal flora. These results provide insights to facilitate the risk assessment of PFHxA contaminants in the aquatic environment.

BACKGROUND: Asthma belongs to chronic inflammatory respiratory diseases characterized by airway inflammation and remodeling. Circular RNAs (circRNAs) are promising therapeutic targets for various diseases, including asthma. In this work, we aim to investigate the role of circular RNA Erb-B2 receptor tyrosine kinase 2 (circERBB2) during progression of asthma. METHODS: Human airway smooth muscle cells (ASMCs) were treated with platelet-derived growth factor BB (PDGF-BB) to mimic cell remodeling. The expression of circERBB2, microRNA-98-5p (miR-98-5p), and insulin-like growth factor 1 receptor (IGF1R) was measured by qRT-PCR. Cell proliferation, migration and apoptosis were determined by cell counting-8 (CCK-8), transwell, and flow cytometry. Protein levels of PCNA, MMP-9, IGF1R were evaluated using Western blotting. The levels of tumor necrosis factor-α (TNF-α), interleukin-1ß (IL-1ß), and IL-6 were detected by enzyme-linked immunosorbent assay (ELISA). Luciferase reporter gene experiment was adopted to evaluate the targeting relationship between miR-98-5p with circERBB2 and IGF1R. Interaction between RNAs was determined by RNA pulldown and RIP assay. RESULTS: The depletion of circERBB2 attenuated the proliferation, migration, and levels of inflammatory factors induced by PDGF-BB and cell apoptosis. CircERBB2 was identified to directly interact with miR-98-5p, and overexpression of miR-98-5p abolished the function of circERBB2 on PDGF-BB-stimulated ASMCs. IGF1R was identified as a target of miR-98-5p, and knockdown of IGF1R relieved the PDGF-BB-induced ASMCs proliferation and migration. CONCLUSION: Our work disclosed that knockdown of circERBB2 suppressed PDGF-BB-caused proliferation, migration and inflammatory response of ASMCs, through regulating miR-98-5p/IGF1R signaling, presented circERBB2 as a promising therapeutic target for asthma.

Six kinds of impermeable underlying surface, cement tile roof, asbestos roof, cement flat roof, residential concrete pavement, asphalt pavement of restaurants, asphalt pavement of oil depot, and a combined sewer overflow canal in the Jiansheng town of Dadukou district in Chongqing city were chosen as sample plots to study the characteristics of nutritional pollutants and heavy metals in town runoff. The research showed that the average mass concentrations of TSS, COD, TN, TP in road runoff were (1681.2 +/- 677.2), (1154.7 +/- 415.5), (12.07 +/- 2.72), (3.32 +/- 1.15) mgL(-1), respectively. These pollutants were higher than those in roof runoff which were (13.3 +/- 6.5), (100.4 +/- 24.8), (3.58 +/- 0.70), (0.10 +/- 0.02) mg x L(-1), respectively. TDN accounted for 62.60% +/- 34.38% of TN, and TDP accounted for 42.22% +/- 33.94% of TP in the runoff of impermeable underlying surface. Compared with the central urban runoff, town runoff in our study had higher mass concentrations of these pollutants. The mass concentrations of TSS, COD, TDN, TN, TDP and TP in the combined sewer overflow were (281.57 +/- 308.38), (231.21 +/- 42.95), (8.16 +/- 2.78), (10.60 +/- 3.94), (0.38 +/- 0.23) and (1.51 +/- 0.75) mg x L(-1), respectively. The average levels of heavy metals in this kind of runoff did not exceed the class VI level of the surface water environmental quality standard. Most pollutants in the combined sewer overflow had first flush. However, this phenomenon was very rare for TSS. There was a significant positive correlation between TSS and COD, TP in the combined sewer overflow. And this correlation was significant between NH4+ -N and TP, TDP, TN, TDP. However, a negative correlation existed between NO3- -N and all other indicators.
