RESUMO

The long-term inflammatory microenvironment is one of the main obstacles to inhibit acute spinal cord injury (SCI) repair. The natural adipose tissue-derived extracellular matrix hydrogel shows effective anti-inflammatory regulation because of its unique protein components. However, the rapid degradation rate and removal of functional proteins during the decellularization process impair the lasting anti-inflammation function of the adipose tissue-derived hydrogel. To address this problem, adipose tissue lysate provides an effective way for SCI repair due to its abundance of anti-inflammatory and nerve regeneration-related proteins. Thereby, human adipose tissue lysate-based hydrogel (HATLH) with an appropriate degradation rate is developed, which aims to in situ long-term recruit and induce anti-inflammatory M2 macrophages through sustainedly released proteins. HATLH can recruit and polarize M2 macrophages while inhibiting pro-inflammatory M1 macrophages regardless of human or mouse-originated. The axonal growth of neuronal cells also can be effectively improved by HATLH and HATLH-induced M2 macrophages. In vivo experiments reveal that HATLH promotes endogenous M2 macrophages infiltration in large numbers (3.5 × 105/100 µL hydrogel) and maintains a long duration for over a month. In a mouse SCI model, HATLH significantly inhibits local inflammatory response, improves neuron and oligodendrocyte differentiation, enhances axonal growth and remyelination, as well as accelerates neurological function restoration.

Saikosaponin a (Ssa) is an active ingredient of the Chinese herbal plant Radix Bupleuri (RB) and has severe hepatotoxicity. However, biomolecular mechanisms involved in Ssa-induced hepatotoxicity are not yet entirely clear. Previous studies reported that Ssd (an isomer of Ssa) as a sarcoplasmic/endoplasmic reticulum calcium ATPase (SERCA) inhibitor can induce autophagy in apoptotic defective cells, leading to autophagy-dependent cell death. Therefore, we speculate that endoplasmic reticulum (ER) stress and autophagy may also play an important role in Ssa-induced hepatocyte death. This study aimed to explore the connection between ER stress and autophagy and Ssa-induced hepatotoxicity. Experiments in vitro showed that the cell viability of L-02 cells in the Ssa treatment group decreased, the level of autophagy marker LC3-II/LC3-I and Beclin1 increased, the level of p62 decreased, the colocalization of autophagosome and lysosome increased, and the cell viability was significantly increased after the application of autophagy inhibitors 3-MA. In addition, SSa can induce ER stress in L-02 cells in vitro. Further studies demonstrated that SSa activated the PERK/eIF2α/ATF4/CHOP pathway, IRE1-TRAF2 pathway, ATF6 pathway, and AMPK/mTOR pathway associated with ER stress. Application of ER stress inhibitors 4-PBA can significantly down-regulate the level of autophagy and improve cell viability. Results of in vivo experiments showed that treatment with 150 and 300 mg/kg Ssa significantly elevated the liver/body weight ratio and caused histological injury in mice liver. Furthermore, Ssa treatment induced significantly downregulated p62 expression but upregulated LC3-II, CHOP, and GRP78 expression in mice livers. Taken together, our results showed that SSa can activate endoplasmic reticulum stress, promote toxic autophagy, and then induce cell death. We revealed an alternative mechanism involving autophagy and ERs, by which Ssa induced hepatotoxicity.

BACKGROUND: The most commonly ingested foreign body in Asians is fish bone. The vast majority of patients have obvious symptoms and can be timely diagnosed and treated. Cases of pyogenic cervical spondylitis and diskitis with retropharyngeal and epidural abscess resulting in incomplete quadriplegia due to foreign body ingestion have been rarely reported. The absence of pharyngeal or esophageal discomfort and negative computed tomography (CT) findings of fish bone have not been reported. We report the case of an elderly female patient with delayed cervical infection and incomplete quadriplegia who had a history of fish bone ingestion. CASE SUMMARY: A 73-year-old woman presented with right neck pain and weakness of four limbs for a week, and had a history of fish bone ingestion and negative findings on laryngoscopic examination one month previously. She did not complain of any pharyngeal or esophageal discomfort. Cervical magnetic resonance imaging showed C4/C5 spondylitis and diskitis along with retropharyngeal and ventral epidural abscesses. No sign of fish bone was detected on lateral cervical radiography and CT scans. The muscle strength of the patient's right lower limb receded to grade 1 and other limbs to grade 2 suddenly on the 10th day of hospitalization. Emergency surgery was performed to drain the abscess and decompress the spinal cord by removing the anterior inflammatory necrotic tissue. Simultaneously, flexible esophagogastroduodenoscopy was carried out and a hole in the posterior pharyngeal wall was found. The motor weakness of the right lower limb improved to grade 3 and the other limbs to grade 4 within 2 d postoperatively. CONCLUSION: This rare case highlights the awareness of the posterior pharyngeal or esophageal wall perforation in patients with cervical pyogenic spondylitis along with a history of fish bone ingestion, even though local discomfort symptoms are absent and the radiological examinations are negative.

OBJECTIVE: To explore and analyze the effect of liquid dressing in relieving pain and preventing incision adhesion after circumcision and its clinical application value. METHODS: Ninety male patients underwent circumcision in Hangzhou Third People's Hospital from September to November 2019, with the incision covered with liquid dressing + vaseline gauze (group A, n = 30), liquid dressing alone (group B, n = 30) or vaseline gauze only (group C, n = 30). At 2, 4 and 6 days after surgery, we compared the Visual Analogue Scale (VAS) pain intensity at dressing change, incision bleeding after dressing removal and incidence of postoperative complications among the three groups of patients. RESULTS: At 2, 4 and 6 days after surgery, the VAS pain score and incidence of incision bleeding were significantly lower in groups A and B than in C (P < 0.05). At 2 days, both the VAS pain score and incidence of incision bleeding were markedly decreased in group A as compared with those in group B (P < 0.05). At 4 and 6 days, the VAS pain score remained lower in group A than in B (P < 0.05), but the incidence rate of incision bleeding showed no significant difference between the two groups (P > 0.05). No statistically significant differences were observed in the incidence of postoperative complications among the three groups (P > 0.05). CONCLUSIONS: Liquid dressing can reduce pain intensity at dressing change, prevent incision adhesion and consequent dressing change-induced tearing and bleeding, and therefore promote incision healing after circumcision. Its combination with vaseline gauze can achieve an even better effect.

The aim of this study was to evaluate the safety and efficacy of percutaneous vertebroplasty (PVP) in Kambin triangle approach for the treatment of osteoporotic vertebral compression fractures (OVCFs).Between November 2017 and September 2018, 109 patients (144 vertebral bodies) with OVCFs, with a mean age of 76.7 ±â€Š9.9 years (55-96 years), underwent PVP in Kambin triangle approach. The time of operation, the volume of bone cement, the incidence of complication, the Visual Analog Scale (VAS) and Oswestry Disability Index (ODI) score, the position of puncture needles, and the spread of polymethylmethacrylate (PMMA) in vertebral body (VB) were recorded.All patients had been completed the operation successfully and were followed up 9.1 ±â€Š2.9 months. The average operation time of each VB was 24.0 ±â€Š3.5 minutes. The average volume of cement was 4.8 ±â€Š0.6 ml. The mean VAS scores were 8.4 ±â€Š0.7 preoperatively, 1.6 ±â€Š0.6 at the first day postoperatively, and 1.2 ±â€Š0.6 at the last follow-up. The mean ODI scores were 70.97 ±â€Š7.73 preoperatively, 27.99 ±â€Š4.12 at the first day postoperatively, and 19.65 ±â€Š3.49 at the last follow-up. The position of puncture needles in the VB was: 119 vertebral puncture needles reached the midline, 15 were close to the midline, and 10 exceeded the midline. The spread of PMMA in the VB was: type 1 in 81 levels (56.3%), type 2 in 37 (25.7%), type 3 in 18 (12.5%), type 5 in 8 (5.5%), and no case in type 4. One case developed pneumothorax after operation. No other complications (hematoma, cement embolism, spinal cord, and nerve injury) occurred.Kambin triangle approach in PVP, which can deliver the puncture needle to the midline of VB easily and with excellent cement distribution, is a safe and effective method.

Sulfur was impregnated into hyper-cross-linked porous polymer (HCP) with a high specific area and unique porous structure. Compared to its inorganic or carbon counterparts, the HCP has a relatively high specific surface area of 1980 m2 g-1 with a total pore volume of 2.61 cm3 g-1, resulting in sulfur content in HCP/S of as high as 80 wt %. As a benefit of the unique HCP structure, the HCP/S composite exhibits a high initial discharge specific capacity (1333 mA h g-1 at 0.2 C), high-rate property, and good cycling stability (658 mA h g-1 after 120 cycles at 0.5 C and 604 mA h g-1 after 80 cycles at 1 C). Furthermore, the capacity of cells loses less than 1% after the first 20 charge/discharge cycles, while the HCP/S cathode can be cycled with an excellent Coulombic efficiency of above 94% after 120 cycles. Compared with pristine sulfur, the superior electrochemical performance of HCP/S composite is related to the cross-linked porous framework. Such structure could provide short ionic/electronic conduction pathways and suppress the polysulfide shuttle during the discharge process.

We synthesized a series of pyrrolidinium based dicationic ionic crystals with high melting point and good thermal stability. Research on the crystal structure shows that there are ordered three-dimensional ionic channels in these crystals which is favorable for the ionic conductor to achieve high conductivity and diffusion coefficient. These ionic crystals are applied to electrolyte as matrix in dye sensitized solar cells, and the influence of crystal structure (including the alkylene chain separating two pyrrolidinium rings and anion) versus the device performances are studied by steady-state voltammography, current-voltage trace, and electrochemical impedance spectroscopy. As the solid state electrolyte, an optimized efficiency of 6.02% have achieved under full sunlight irradiation using ionic crystal [C6BEP][TFSI]2. And the device based on this solid electrolyte shows the excellent long-term stability, maintaining 92% of the initial efficiency after 960 h. This study elucidates fundamental the structure of dicationic crystal and provide useful clues for further improvement of solid-state electrolytes in DSSC.

Thiaozole/benzothiaozole-based dicationic conductors were synthesized and applied as solid-state electrolyte in dye-sensitized solar cells (DSSCs). X-ray diffraction, scanning electron microscopy, thermal gravimetric analysis, steady-state voltammogram, photocurrent intensity-photovoltage test, and electrochemical impedance spectroscopy are used to characterize the materials and the mechanism of the cell performance. Compared to the traditional monocationic crystals, the dicationic crystals have a larger size and can provide more opportunities to fine-tune their physical/chemical properties. As a consequence, this solid-state electrolyte-based DSSC achieved photoelectric conversion efficiency of 7.90% under full air-mass (AM 1.5) sunlight (100 mW·cm(-2)).

Titanium dioxide (TiO2)-layered fluorine doped tin oxide (FTO) powder was synthesized and applied as the photoanode in dye-sensitized solar cells (DSSCs). FTO powders are connected to form a direct electron pathway for the efficient extract of injected electrons, while the TiO2 layer serves as an energy barrier prohibiting the charge combination with oxidized dye or I3(-). The electrochemical impedance spectroscopy (EIS) analyses suggest that electrons have a longer combination lifetime (τe = 233 ms) than that of the electron in the DSSCs using traditional P25 photoanodes (τe = 28 ms). The DSSCs using 5 µm thick TiO2@FTO as photoanodes eventually give a respectable and long-term stable photovoltaic performance with a current density of 23.8 mA/cm(2), an open circuit voltage of 0.69 V, and power conversion efficiency of 7.4%. The results are received on a low dye loading level (0.25 × 10(-7) mol/cm(2)), which is (1)/10 of that for traditional photoanode (2.79 × 10(-7) mol/cm(2)).

RESUMO

One unusual aromatic monacolin analog, aromonacolin A (1), was isolated from the ethanolic extract of Monascus purpureus-fermented rice. Its structure was elucidated by extensive spectroscopic (HRESIMS, (1)H NMR, (13)C NMR, HSQC, HMBC, and NOESY) and chemical methods. The absolute configuration of the C-6 secondary alcohol was deduced via the circular dichroism data of the in situ formed [Rh(2)(OCOCF(3))(4)] complex.

RESUMO

Brain natriuretic peptide (BNP) and N-terminal brain natriuretic peptide (NT-proBNP) are important biomarkers for pediatric cardiovascular diseases. Peptide levels are associated with age and gender. Current studies have shown that BNP and NT-proBNP are valuable in the diagnosis of heart failure, with a high specificity and sensitivity. They also contribute to differentiating heart failure from acute respiratory distress induced by simple pulmonary factors. In addition, BNP and NT-proBNP are useful in the evaluation of disease severity and treatment guidance in children with pulmonary hypertension, cardiomyopathy and Kawasaki disease. Current limitations include the relatively small sample size of the study, the detection method and a range of normal values that are not completely uniform.

Bioassay-guided fractionation of an EtOH extract of Monascus purpureus-fermented rice led to the isolation of two new steroids (22S, 23R, 24S)-20ß,23α,25α-trihydroxy-16,22-epoxy-4,6,8(14)-trienergosta-3-one (1), the first example of a steroid possessing both a conjugated triene ketone system and a fused 4H-furan ring side chain within one molecule, and (22E, 24R)-3ß,5α-dihydroxyergosta-23-methyl-7,22-dien-6-one (2), as well as two known compounds (22E, 24R)-3ß,5α-dihydroxyergosta-7,22-dien-6-one (3) and (22E, 24R)-6ß-methoxy-ergosta-7,22-diene-3ß,5α-diol (4). Their structures were assigned by detailed interpretation of HRESIMS, 1D and 2D NMR spectroscopic data. The absolute stereochemistry of 1 was determined by single-crystal X-ray crystallography while the absolute stereochemistry of 2 was established by CD. Compounds 1-4 showed cytotoxic activity against the lung adenocarcinoma (A549) with IC(50) values of 0.08, 0.94, 12.6 and 13.5 µM, respectively. In addition, compounds 1 and 2 exhibited moderate activities against human ovarian cancer (A2780), with IC(50) values of 2.8 and 5.1 µM.

Sphere- and rod-shaped Zn(2)SiO(4):Mn(2+) phosphor nanocrystals were synthesized at 230 degrees C. The process consists of tuning the surfactant concentration in the oil/surfactant/ethanol system. Powder X-ray (XRD) and transmission electron microscopy (TEM) were used to characterize the phase purity, size and morphology. Photoluminescent (PL) spectra were collected and analyzed. Fourier transform infrared (FT-IR) spectra of the samples indicate the removal of surfactant in the phosphor nanoparticles. As a result, the sphere-shaped phosphor nanoparticles of 100 nm in size can be redispersed in ethanol ultrasonically. The suspension maintain stable for over 48 h.
