RESUMO

Boron-doped diamond thin films exhibit extensive applications in chemical sensing, in which the performance could be further enhanced by nano-structuring of the surfaces. In order to discover the relationship between diamond nanostructures and properties, this paper is dedicated to deep learning target detection methods. However, great challenges, such as noise, unclear target boundaries, and mutual occlusion between targets, are inevitable during the target detection of nanostructures. To tackle these challenges, DWS-YOLOv8 (DCN + WIoU + SA + YOLOv8n) is introduced to optimize the YOLOv8n model for the detection of diamond nanostructures. A deformable convolutional C2f (DCN_C2f) module is integrated into the backbone network, as is a shuffling attention (SA) mechanism, for adaptively tuning the perceptual field of the network and reducing the effect of noise. Finally, Wise-IoU (WIoU)v3 is utilized as a bounding box regression loss to enhance the model's ability to localize diamond nanostructures. Compared to YOLOv8n, a 9.4% higher detection accuracy is achieved for the present model with reduced computational complexity. Additionally, the enhancement of precision (P), recall (R), mAP@0.5, and mAP@0.5:0.95 is demonstrated, which validates the effectiveness of the present DWS-YOLOv8 method. These methods provide effective support for the subsequent understanding and customization of the properties of surface nanostructures.

RESUMO

The mechanical and physicochemical properties of diesel engine exhaust particles before and after diesel oxidation catalyst (DOC) treatment are analyzed. It is considered important to explore the interrelationships between these attributes in order to understand their relevance. Understanding of these properties provides insights into the deposition characteristics of particles within the system and the evolution of the particles after the DOC treatment, which may help the selection of appropriate aftertreatment strategies. In this paper, particle samples were collected before and after the DOC to explore the variations in the mechanical and physicochemical properties of the particles under different operating conditions. Atomic force microscopy, thermogravimetric analysis, transmission electron microscopy, and Raman spectroscopy were employed to investigate the attraction force, adhesion force, adhesion energy, oxidative reactivity, primary particle size, nanostructure, and graphitization degree of the particles. The results indicated that under post-injection conditions, the attraction force, adhesion force, and adhesion energy of the particles increased significantly. However, when the particles passed through the DOC, these properties decreased to varying degrees. By analyzing the combination of physicochemical properties, it was determined that the attraction force of the particles was primarily influenced by the primary particle size and the particle's graphite structure. The adhesion force was found to be closely related to the content of soluble organic matter. Additionally, the soluble organic matter affected the degree of particle agglomeration by altering the adhesion energy of the particles.

RESUMO

Boron-doped diamond (BDD) thin film electrodes have great application potential in water treatment. However, the high electrode energy consumption due to high resistance directly limits the application range of existing BDD electrodes. In this paper, the BDD/graphene/BDD (DGD) sandwich structure electrode was prepared, which effectively improved the conductivity of the electrode. Meanwhile, the sandwich electrode can effectively avoid the degradation of electrode performance caused by the large amount of non-diamond carbon introduced by heavy doping, such as the reduction of the electrochemical window and the decrease of physical and chemical stability. The microstructure and composition of the film were characterized by scanning electron microscope (SEM), atomic force microscopy (AFM), Raman spectroscopy, and transmission electron microscopy (TEM). Then, the degradation performance of citric acid (CA), catechol, and tetracycline hydrochloride (TCH) by DGD electrodes was systematically studied by total organic carbon (TOC) and Energy consumption per unit TOC removal (ECTOC). Compared with the single BDD electrode, the new DGD electrode improves the mobility of the electrode and reduces the mass transfer resistance by 1/3, showing better water treatment performance. In the process of dealing with Citric acid, the step current of the DGD electrode was 1.35 times that of the BDD electrode, and the energy utilization ratio of the DGD electrode was 2.4 times that of the BDD electrode. The energy consumption per unit TOC removal (ECTOC) of the DGD electrode was lower than that of BDD, especially Catechol, which was reduced to 66.9% of BDD. The DGD sandwich electrode, as a new electrode material, has good electrochemical degradation performance and can be used for high-efficiency electrocatalytic degradation of organic pollutants.

RESUMO

In order to qualitatively evaluate the impact of urea-in-water solution on the particle number emission, five different urea types were chosen under diesel engine bench test. The results show that compared to the instantaneous particle concentrations without injection, the instantaneous particle concentrations with injection increase distinctly, which are larger around 0.3-1.2 times than that without urea-in-water solution. At high speed phase, the instantaneous particle concentrations with urea-in-water solution injection rise obviously, especially for C and E solutions. In addition, the particle size distribution characteristic does not change with the urea-in-water solution dosing. The PN emission factors follow the sequence of C solution > E solution > A solution > B solution > D solution. It is deduced that the metallic element contents in the urea-in-water solution play a key role for the PN emission factor. In the future, the particle formation due to urea-in-water solution injection should be given more attention.

RESUMO

Broca's area, made up of Brodmann areas (BA) 44 and 45 in the ventrolateral frontal region, is associated with language production and articulation. A comprehensive network analysis of Broca's area is necessary for understanding language function, which is still lacking. In this study, we attempted to investigate the association fiber tracts related to Broca's area using both diffusion spectrum imaging (DSI) and postmortem fiber dissection. DSI was performed on 10 healthy subjects and an atlas comprising the average data of 842 healthy subjects from the Human Connectome Project. Fiber dissection was implemented in 10 cerebral hemispheres of cadaver donors. The following five association fiber tracts related to Broca's area were identified: first, the distinct fasciculus of the inferior fronto-occipital fasciculus (IFOF), from Broca's area (BA44, BA45) and pars orbitalis (BA47) to the parietal and occipital lobes; second, the ventral superior longitudinal fasciculus (SLF-III), from the supramarginal gyrus (BA40) to the ventral precentral gyrus (PreG, BA6) and posterior Broca's area (BA44); third, the arcuate fascicle (AF), from the superior, middle, and inferior temporal gyrus (BA20, BA21, BA22) to Broca's area (BA44, BA45) and ventral PreG; fourth, the frontal aslant tract (FAT), from Broca's area (BA44, BA45) to the lateral superior frontal gyrus (SFG), medial SFG, and supplementary motor area (BA6, BA8, BA9); and fifth, the frontal longitudinal fasciculus (FLF), a novel intralobar frontal association fiber tract, from the anterior part of the middle frontal gyrus (MFG, BA46) and Broca's area (BA45) to the caudal MFG (BA8), caudal SFG, and dorsal PreG (BA6). Moreover, compared with the left FAT, the right FAT covered almost the entire inferior frontal gyrus (BA44, BA45, BA47). The cross validation between DSI and fiber dissection revealed a good consistence in the association fiber tracts of Broca's area. Combining DSI and fiber dissection, this study first identified five association fiber tracts related to Broca's area and characterized their structure and anatomy comprehensively. The frameworks provided key elements for functional research in Broca's area.

RESUMO

Short-term power load forecasting is essential in ensuring the safe operation of power systems and a prerequisite in building automated power systems. Short-term power load demonstrates substantial volatility because of the effect of various factors, such as temperature and weather conditions. However, the traditional short-term power load forecasting method ignores the influence of various factors on the load and presents problems of limited nonlinear mapping ability and weak generalization ability to unknown data. Therefore, a short-term power load forecasting method based on GRA and ABC-SVM is proposed in this study. First, the Pearson correlation coefficient method is used to select critical influencing factors. Second, the gray relational analysis (GRA) method is utilized to screen similar days in the history, construct a rough set of similar days, perform K-means clustering on the rough sets of similar days, and further construct the set of similar days. The artificial bee colony (ABC) algorithm is then utilized to optimize penalty coefficient and kernel function parameters of the support vector machine (SVM). Finally, the above method is applied on the basis of actual load data in Nanjing for simulation verification, and the results show the effectiveness of the proposed method.

RESUMO

This study mainly focused on a strategy for reducing acrylamide(AM) content in wheat breads by combining Lactobacilli and Saccharomyces cerevisiae in sourdough, in comparison with natural fermentation. The results showed that acrylamide levels in breads using sourdough were much lower (102.02-129.37 µg/kg) than control group (204.79 µg/kg). The pH value of sourdough directly influenced the formation of acrylamide in breads (P < 0.01). Furthermore, significant (P < 0.05) correlations were also found between protein and acrylamide contents. There no significant correlations were observed between acrylamide and reducing sugar contents. According to the different effects of strains, it could be concluded that the acrylamide reducing potential of strains was strain-specific, with Pediococcus pentosaceus being the most effective. This suggests that sourdough fermentation with appropriate strains can be used as an advantageous technology to reduce the acrylamide content of wheat breads.

RESUMO

This study analyzed performance of public hospitals and regional differences in performance following reform of medical service prices in Guangdong province, China. From three cities in four regions, we randomly selected a total of 12 traditional Chinese medicine hospitals and 12 general tertiary hospitals. Six questionnaires were completed by the hospitals, using 2014-2018 internal data. Principal components analysis was used to compare performances of the hospitals and regions following price reform. The extent to which medical service prices were adjusted varied considerable for different procedures in the same region and for the same category of procedures among regions. After reform, compensation for medical services in public hospitals reached the target of 80%, except in the Western region. However, annual growth of costs to patients was generally above 4%; the burden on patients was not alleviated by fee control. Reforms were more effective for comprehensive than Chinese traditional medicine hospitals. Performance scores of general hospitals in the Pearl River Delta, Eastern, Western, and Northern regions were 1.24, 1.16, -0.22, and -1.01, respectively. This is consistent with ranking by level of economic development of each region. The government should implement a regional medical service pricing mechanism. Additionally, comprehensive and traditional Chinese medicine hospitals should each have appropriate pricing policies. Future policies should focus on controlling costs incurred by patients.

RESUMO

Compared with traditional craniotomy, the expanded endoscopic endonasal approach (EEEA) may have some advantages for tuberculum sellae meningioma (TSM) treatment. We described our experience of the therapeutic effect of endoscopic TSM treatment. From August 2015 to December 2019, 40 patients with a TSM were treated by the EEEA in our institution. EEEA outcome in TSM treatment was analyzed. Among 39 patients with visual impairment, 38 (97.4%) improved their visual function to some extent after the EEEA, and one case had no significant change in visual acuity. Among all patients, 38 (95.0%) achieved gross total resection (GTR) and 2 (5.0%) achieved near-total resection (NTR). Cerebrospinal fluid (CSF) leakage occurred in three patients (7.5%) and meningitis (post-CSF leakage) in two patients (5.0%). Eight patients (20.0%) suffered postoperative hyposmia, three of whom developed long-term hyposmia. One patient (2.5%) suffered from bleeding of the branch of the anterior cerebral artery intraoperatively leading to postoperative acute cerebral infarction. The EEEA is a safe and reliable minimally invasive method for TSM removal. Compared with traditional craniotomy, the EEEA may have better visual outcomes and a higher prevalence of GTR, but carries the risk of CSF leakage.

RESUMO

OBJECTIVE: Alcohol abuse can cause severe injury to human brain. Astrocytes are the most abundant nonneuronal cells that function to maintain the brain homeostasis. In present study, we aimed to investigate the role of ROCK2 in astrocytes exposed to alcohol. METHODS: Astrocytes were transfected with lentivirus (LV)-anti-ROCK2 vector to downregulate the expression of ROCK2. The ROCK2 expression in mRNA and protein level was analyzed by real-time PCR and Western blotting, respectively. Cytokines or indicators involved in inflammation and oxidative stress were determined by assay kits. Proteins involved in nuclear factor kappa B (NF-κB) signaling pathway and NOD-like receptor protein 3 (NLRP3) inflammasome were analyzed by Western blotting. RESULTS: Alcohol exposure dramatically upregulated ROCK2 expression and lactate dehydrogenase (LDH) activity in astrocytes. On the contrary, transfecting with LV-anti-ROCK2 vector downregulated ROCK2 expression and LDH activity in astrocytes, demonstrating that downregulation of ROCK2 alleviated alcohol-induced astrocytic injury. Furthermore, downregulation of ROCK2 attenuated alcohol-induced inflammation by reducing the levels of pro-inflammatory cytokines (tumor necrosis factor (TNF)-α and interleukin (IL)-6) and enhanced the level of anti-inflammatory IL-10. Downregulation of ROCK2 also attenuated alcohol-induced oxidative stress by reducing the reactive oxygen species (ROS) production, as well as enhancing the activity of anti-oxidative superoxide dismutase (SOD) and glutathione (GSH). More importantly, downregulation of ROCK2 inhibited the activation of NF-κB signaling pathway and NLRP3 inflammasome. CONCLUSION: Therefore, ROCK2 could be a potential target to treat alcohol-induced astrocytic injury and the downregulation of ROCK2 might be a promising approach to protect against alcohol-induced astrocytic injury.

RESUMO

While the proximal dentatothalamocortical tracts are considered pivotal in the occurrence of cerebellar mutism syndrome (CMS) after medulloblastoma resection, how the cerebellum participates in motor-speech networks through direct structural connectivity is still unclear. Via tractography, we provide evidence of cerebellar streamlines projecting into the left inferior frontal gyrus majorly connecting Broca's complex and the bilateral Crus lobes. The streamlines, named Crus-Broca tracts, originated from the bilateral Crus lobes, synapsed onto the dentate nucleus, ascended into the superior cerebellar peduncle (where these streamlines were closely superior to the superior border of the supratonsillar cleft and the superolateral roof of the fourth ventricle), surprisingly bypassed the left red nucleus and the left thalamus, and ended at the subregions of Broca's complex. The streamlines, named Broca-Crus tracts, originated from the subregions of Broca's complex and ended predominantly at the right Crus lobes. If verified, the existence of these connections would support the notion of the bilateral cerebellums' participation in motor-speech planning, and the anatomical relationship of Broca-Crus tracts with the supratonsillar cleft would merit consideration for further studies aimed at further elucidating CMS mechanisms.

RESUMO

Chronic alcohol consumption leads to hippocampal neuronal impairment, which related to neuronal death, oxidative stress, and inflammatory response. Rho-associated protein kinase 2 (ROCK2) is a major regulator in the central nervous system injury. However, the effects of ROCK2 in ethanol-induced brain injury have not been explored. In this work, we investigated the neuroprotective effects and the mechanism of ROCK2 inhibition in vivo. Wistar rats were exposed to 37% ethanol for 8 weeks to establish brain injury models. Morris water maze test was performed to evaluate cognitive function, and we found that the down-regulation of ROCK2 reduced the escape latency and increased the passing times and percentage of time spent in the target quadrant of rats. The results of H&E staining and Nissl staining showed that ROCK2 inhibition alleviated the pathological injury induced by ethanol. PI staining and Western blot confirmed that inhibiting ROCK2 attenuated the neuronal death and apoptosis as reflected by the reduced PI-positive neurons and the decreased expression of cleaved-caspase-3 and cleaved-caspase-9. Furthermore, the down-regulation of ROCK2 ameliorated the oxidative stress and inflammatory response induced by ethanol in rats as reflected by the up-regulation of IL-10, SOD, and GSH and reduction of TNF-α, IL-6, and MDA respectively. Additionally, Western blot and EMSA analysis revealed that the down-regulation of ROCK2 suppressed the nuclear transfer of NF-κB p65. In conclusion, our data suggested that ROCK2 inhibition ameliorated ethanol-mediated hippocampal neuronal impairment by anti-apoptotic, anti-inflammatory, anti-oxidative effects at least partially through the suppression of the NF-κB pathway.

RESUMO

INTRODUCTION: The existing literature on mobile phone use while driving (MPUWD) mainly targets the participants from general population and the young adults, however, few studies pay attention to this form of distracted driving with samples in professional contexts. The present study aims to bridge the gap by identifying the extent of and the motives behind making use of mobile phones while driving for food dispatch among deliveryman. METHOD: The snowball sampling was used to collect the data (N = 317) through a self-reported questionnaire, including demographics, personality traits, risk perception, driving self-efficacy, and mobile phone use while driving. RESULTS: Descriptive analysis for the assessed MPUWD behaviors showed that 96.3% (N = 315) of food deliveryman undertook the MPUWD behaviors, though disproportionate distribution among these behaviors existed. Structural equation modeling analysis displayed that psychoticism and driving self-efficacy directly predicted the MPUWD behaviors. The mediating role of driving self-efficacy was verified with the findings that driving self-efficacy completely mediated the relationships that between risk perception and MPUWD behaviors and that between extraversion and MPUWD behaviors, as well as partially mediated the correlation between psychoticism and MPUWD behaviors. CONCLUSIONS: The present study confirms the prevalence of MPUWD behaviors among food deliveryman. The SEM estimates and bootstrap estimates suggest that personality traits and perceived risk perception per se display limited predicting utility to MPUWD behaviors among food deliveryman, whereas driving self-efficacy and the proposed predictors together well illustrate the assessed MPUWD behaviors among food deliveryman. Practical Applications: These findings imply that developing and implementing intervention efforts in a concerted way would curb these behaviors effectively.

RESUMO

BACKGROUND: Invasive growth of pituitary macroadenomas to the suprasellar region occurs commonly. Pituitary adenomas show varying growth patterns when the sellar diaphragm is absent, and they are often confused with other common tumors in the sellar region. This article explores the clinical features of suprasellar pituitary adenomas with defects of the sellar diaphragm (SPADSD) and evaluates the efficacy of the endoscopic endonasal approach (EEA) for treatment of such tumors. METHODS: We performed a detailed examination of records from 19 patients collected prior to surgery. After relevant diseases were excluded, the tumor properties were evaluated according to imaging characteristics. Diagnoses were verified using EEA surgery. The concept of SPADSD was put forward. Postoperative recovery was followed to determine whether EEA is suitable for the treatment of such tumors. RESULTS: In the 19 patients with SPADSD, we found that the tumors were less stressed on the pituitary, and tumors in the suprasellar region often had irregular shapes. During surgery, we took extended supra-saddle approaches and confirmed that unrestricted growth of the tumor was caused by defects in the diaphragm of the sella turcica to the suprasellar region. Recovery was good after surgery, confirming the efficacy of EEA for treatment of these tumors. CONCLUSION: SPADSD has different clinical features from those of other pituitary tumors and requires careful screening prior to surgery. Endoscopic surgery is the preferred procedure for this type of tumor.

RESUMO

To meet the industrial needs for SSZ-13, variable sizes of SSZ-13 with different Si/Al ratios were firstly obtained by conventional hydrothermal synthesis using the seed method. Using a set of characterizations, like X-ray fluorescence (XRF), X-ray diffraction (XRD), and scanning electron microscopy (SEM), the physicochemical structure and size distribution could be traced. After the specific Si/Al ratio of SSZ-13 zeolites was optimized, synthesized by changing the amounts of structure-directing agents (SDAs) and NaOH, the obtained SSZ-13 showed a high degree of crystallinity. With the limitation of the pH values, the variation of the alkalinity and water content was not helpful to generate different grain sizes of SSZ-13 materials. With the help of ground seed, the various grain sizes of SSZ-13s from 0.4 to 4 µm had a similar degree of crystallinity and size distribution. Moreover, due to the identical intensity of the Al peak in the NMR results, the different grain sizes of SSZ-13s had the same acidity. Our study revealed that using the seed method was an easy and efficient way to synthesize SSZ-13s with different sizes.

RESUMO

Endoscopic endonasal approaches (EEAs) are ideal for most chordomas, but there is little information regarding the practical clinical classification of clival chordomas to guide surgery with EEAs. This article investigates a relatively concise and practical clinical classification system for clival chordomas and summarizes the clinical characteristics and operative key points of different clinical types. Here, 55 patients with clival chordomas treated through EEAs from 2012 to 2017 were retrospectively reviewed. Depending on the origin of the notochord and the growth pattern of the tumor, with our introduced Wang's line, these cases of clival chordoma were divided into types I-IV. There were 14 cases of type I-A, 7 cases of type I-B, 10 cases of type II, 10 cases of type III-A, 7 cases of type III-B, and 7 cases of type IV. The gross total resection (GTR) rate of primary and recurrent type I tumors was 64% and 25%, and residual tumors were found mainly in cases with involvement of the cavernous sinus or the posterior upper part of the dorsum sella. The GTR rate of primary and recurrent type II tumors was 85% and 66.6%, respectively. Residual tumors were found in cases with involvement of the petrous apex. The GTR rate of primary and recurrent type III tumors was 75% and 20%, and residual tumors were found in cases with involvement of the parapharyngeal space and dorsal side of C1-2. Residual type I-B and type III-B tumors were found when there was BA or VA adhesion or brain stem invasion. Our new classification method proposed here can be used to guide the resection of clival chordomas through EEAs.

RESUMO

BACKGROUND: Fertility desire for a second child has been a lively topic since the implementation of the two-child policy in China. Chinese researchers have explored various factors influencing the fertility desire for a second child. However, there have not been studies on the individual differences in the relative fertility costs and fertility benefits and their effects on fertility desire for a second child. METHODS: A total of 396 participants rated four kinds of relative fertility costs, four kinds of fertility benefits and their fertility desire for a second child. Latent profile analysis (LPA) was used to explore the individual differences in the relative fertility costs and fertility benefits and their effects on fertility desire for a second child. RESULTS: Stepwise regression analysis showed that parenting joy, health risks, mutual care among siblings, the flourishing of family, and time pressure and opportunity cost significantly predicted the fertility desire for the second child. According to the latent profile analysis, the participants were classified into four classes. Participants in the lowest-cost/lowest-benefit and high-cost/medium-benefit classes had low fertility desire for a second child, while those in the low-cost/high-benefit and highest-cost/highest-benefit classes had high fertility desire. CONCLUSION: Fertility benefits have a stronger effect on the fertility desire for a second child than relative fertility costs. Fertility benefits should be paid more attention to than relative fertility costs.

RESUMO

OBJECTIVE: The aim of this study was to discuss the relationship between Ki-67 values and the degree to which chordoma invade the clivus and to certify that the prognosis of chordoma is worse when it invades the middle and lower clivus than when it does not. METHODS: We collected 56 cases of first-time chordoma illness in which patients received no treatment before surgery. Patients underwent craniocerebral magnetic resonance imaging and skull-base 3-dimensional computed tomography scans before the operation. We divided patients into 2 groups depending on the extent to which the middle and lower clivus were invaded. We classified patients with chordoma that did not significantly invade the middle and lower clivuses into a "noninvasive" group and the others into an "invasive" group. Ki-67 values were extracted from the pathological report after surgery. We use an independent χ2 test to indicate that Ki-67 values for the invasive group were higher than those for the noninvasive group. RESULTS: We grouped the data and did a statistical analysis. We found that the Ki-67 values are >5% for most patients in whom chordoma have eroded the middle-lower clivus, whereas it is ≤5% for patients in whom the middle-lower clivus region has not been invaded. Therefore, there is a correlation between Ki-67 value and the region of chordoma invading the clivus. CONCLUSIONS: Statistical analysis revealed that Ki-67 values when the chordoma invaded the middle and lower clivus were significantly higher than when it did not. Thus, we can conclude that the prognosis is worse when chordoma invade the middle and lower clivus.

RESUMO

PURPOSE: The aim of this study was to discuss the advantages of GQI reconstruction in the imaging of nerve fibers at crossing regions. Compared with DTI, the paper also discussed the advantages of GQI in imaging principles. METHODS: 3-T MRI data from five normal participants were reconstructed using GQI and DTI. After adjusting the parameters, we compared the differences in reconstructed nerve fibers at the crossing regions between the two methods. To complete this study, we chose four obvious examples (the optic nerve, the Superior cerebellar peduncles, the intersection of the pyramidal tract, the corpus callosum and the arcuate fibers and the intersection of the supplementary motor area (SMA) and the anterior part of arcuate fasciculus) to illustrate. RESULTS: By reconstructing nerve fibers in three regions, we can find that crossing-area images of nerve fibers significantly differed between DTI and GQI reconstruction. Although crossing fibers could be clearly and completely visualized after GQI reconstruction, they showed artifacts, incompleteness, deletions, and fractures after DTI reconstruction. After GQI reconstruction, we can find that there were two or more nerve fibers in each voxel. However, only one nerve fiber was present in each voxel after DTI reconstruction. CONCLUSION: The imaging of crossing fibers is more complete, consistent, and accurate when they are reconstructed by GQI than when they are reconstructed by DTI.

RESUMO

OBJECTIVE: Intracranial aneurysm (IA) is a life threatening cerebrovascular disease characterized by phenotypic modulation of vascular smooth muscle cells (VSMCs) and loss of vessel cells. In addition to environmental factors, genetic factors have been proposed to be a critical factor in the onset and progression of IA. The present study investigates the effects of serine-arginine protein kinase 1 (SRPK1) on VSMC proliferation and apoptosis both in vivo and in vitro, as well as its role in vascular remodeling in vivo through PI3 K/Akt signaling in IA. METHODS: Differentially expressed genes related to IA were initially identified using microarray analysis. Immunohistochemistry was conducted to determine SRPK1 expression in the vascular walls in IA and normal cerebral vascular walls. TUNEL staining were applied to observe cell apoptosis patterns of VSMCs. VSMC proliferation and apoptosis in vitro were detected by cell counting kit-8 (CCK8) assay and flow cytometry. The expressions of SRPK1, PI3 K/Akt signaling pathway- and apoptosis-related genes were evaluated by RT-qPCR and Western blot analysis. RESULTS: Microarray data of GSE36791 and GSE54083 were analyzed to determine the selection of SRPK1 gene. The vascular walls in IA rat models produced high levels of SRPK1 expression and an activated PI3 K/Akt signaling pathway. VSMCs treated with siRNA-SRPK1 exhibited enhanced cell proliferation, repressed cell apoptosis, and increased vascular remodeling, all of which suggest the inhibition of the PI3 K/AKT pathway. Notably, PI3 K/AKT pathway reversed the effect of SRPK1 silencing. CONCLUSION: Our results show that siRNA-mediated silencing of SRPK1 gene inhibits VSMC apoptosis, and increases VSMCs proliferation and vascular remodeling in IA via the PI3 K/Akt signaling pathway. Our findings provide a novel intervention target for the molecular treatment of IA.
