RESUMO

Numerous studies have shown that different subtypes of breast cancer (BC) have significant differences in terms of the tumor microbiome, host gene expression, and histopathological image, whereas the biological links between these cancer-associated indicators are still unknown. Here, we performed a comprehensive analysis with 610 patients of the four subtypes of BC with matched tissue microbiota, host transcriptome, and histopathological image samples. Correlation analysis showed that the composition of intratumoral viruses shaped the tumor microenvironment (TME) of patients with BC, and the TME was further reflected in the histopathological images. Of the four subtypes, patients with triple-negative breast cancer (TNBC) had unique intratumoral viral community composition, non-cancer cell infiltration in the TME, and histopathological image characteristics. Furthermore, we detected multiple virus-cell-image association axes in TNBC, in which tumor-associated macrophages (TAMs) have clinical prognostic implication. This study provides a comprehensive map of the associations between the intratumoral virome, TME, and histopathological image of TNBC, as well as insights into disease prognosis that can be crucial for precise therapeutic intervention strategies.

RESUMO

Black-odorous water, which exists widely in rural areas of China, affects the living environments of residents and causes a loss of ecosystem functions, and improper treatment may even cause secondary pollution. Moreover, the transformation relationships among the components of humus in the sediments of black-odorous water are largely unknown. Therefore, it is necessary to select appropriate methods for treating black-odorous water in rural areas and to understand the characteristics of humus in sediment during the treatment process. In this study, the use of aeration in treating black-odorous water and interstitial water in rural areas was studied and the effects of different treatment methods on the contents and spectral characteristics of humus components in sediments were explored. It was found that the quality of the treated water improved from severe black-odorous to nonblack-odorous, the colour of the sediment changed from black to greyish-brown to turquoise, and the odour changed from strong to weak. The removal rates of ammonia nitrogen, total nitrogen, total phosphorus, and chemical oxygen demand in the sediment aeration group reached more than 50% in the upper water, and more than 70% in the interstitial water. After treatment, the content of fulvic acid, the main black substance in sediment, decreased by 0.36-1.58 g/kg, and the molecular structure of the humus was simplified.

RESUMO

The Poplar Ecological Retreat (PER) project is a significant environmental protection initiative implemented to protect the Dongting Lake wetlands ecosystem in China, and the ecological impacts of this project have gradually become a hot topic. In this study, water conservation was selected as an indicator of ecosystem function to explore the impact of the project by using the water yield module of the Integrated Valuation of Ecosystem Services and Tradeoffs (InVEST) model and geographic statistical methods. Water yield and land-use changes were quantitatively evaluated, and significant changes in the wetland landscape configuration occurred after the PER project because forest beach, moss marshland and mudflat areas were transformed into a poplar-retreat area. Thus, the proportion of the poplar-retreat area and average water yield increased. By using the principal component and cluster analysis methods, climate change and landscape patterns were shown to be the dominant driving factors affecting water conservation under stable meteorological conditions. Moreover, the landscape heterogeneity that resulted from the landscape patterns had a greater impact than climate change, which means that reducing human activities can enhance water conservation. Additionally, the correlation between landscape heterogeneity and water yield indicated that reducing landscape fragmentation and improving aggregation also benefit water conservation. Therefore, the PER project is successful at enhancing ecological functions and protecting wetland environments, and it represents a good example of maintaining ecological sustainability despite local economic performance restrictions. This project serves as an important reference for the establishment of sustainable wetland ecological policies by national governments, and the results can provide theoretical support for landscape ecology and eco-hydrology research.

RESUMO

The hsp70 and hsp90 expression patterns and catalase (CAT) activity in the freshwater planaria Dugesia japonica exposed to cadmium (Cd) under laboratory conditions were investigated. Planaria were exposed to a range of Cd concentrations (0-150 µg Cd/L) for 24 h. The expression levels of hsp70 and hsp90 were determined by relative quantitative real-time polymerase chain reaction. Within the overall dose range in the experiment, the expression level of hsp70 and the activity of CAT in D. japonica were altered significantly. Hsp70 was induced in D. japonica upon Cd exposure concentrations as low as 9.375 µg Cd/L. No significant effect on the expression level of hsp90 was observed. Our findings demonstrated that stress gene hsp70, but not hsp90, was responsive to Cd contamination in D. japonica CAT activity was significantly induced at concentrations of 18.75, 37.5, and 75 µg Cd/L after 24-h exposure. We recommend that the use of hsp70 as a biomarker should be complemented by evidence of changes in other parameters, such as CAT activity, in D. japonica.
