RESUMO

Objective To evaluate the efficacy and risks of autologous blood patch pleurodesis in patients with persistent air leak(PAL)after lung resection. Methods A total of 97 patients with PAL after lung resection in Beijing Shijitan Hospital from October 2014 to October 2019 were retrospectively reviewed, including 53 treated by autologous blood patch pleurodesis and 44 by the conventional way.The therapeutic effect, adverse reactions and complications were analyzed. Results All the patients with PAL were cured with autologous blood patch pleurodesis. Most air leaks(81.1%)ceased within 48 hours after treatment, and the left 18.9% patients got cured after a repeat. The mean tube retention time and the mean in-hospital stay were 8.4 days and 10.0 days in the autologous blood patch pleurodesis group and 13.5 days and 15.3 days in the conventional treatment group.A prolonged drainage time(P=0.00)and in-hospital stay(P=0.00)were observed in the conventional treatment group. No severe complications were observed except two patients developed slight fever and cutaneous emphysema. Conclusion In our experience, the autologous blood patch pleurodesis is an effective way with low risk of adverse reactions in the treatment of PAL.

RESUMO

In the present study, an inductively-coupled plasma-mass spectrometry (ICP-MS)-based triple-amplification system, by combination of nicking-displacement, rolling circle amplification (RCA) and bio-bar-code probes, was fabricated for the detection of DNA target. By using this system, hepatitis B virus (HBV) DNA target down to 3.2×10(-17)M was detected by DNA probes labeled with Au nanoparticles (AuNPs). Single nucleotide polymorphisms in genes can also be effectively discriminated. In addition, we proved that this strategy is capable of detecting the target in complicated biological samples and holds great potential application in biomedical research.

RESUMO

On the basis of strong preference for cleaving double-stranded DNA or DNA in DNA:RNA heteroduplexes of duplex-specific nuclease (DSN), a dual-amplified electrochemical detection of mRNA was developed in this article, by coupling the enhancement of DSN and bio-bar-code conjugates. Capture probe was linked with magnetic nanoparticles (MNPs) at its 5' end and bio-bar-code at its 3' end. In the presence of target surviving mRNA, all hybridized S1 strands were cleaved off the biosensor by the DSN, and the bio-bar-code probe with CdS nanoparticles (CdS NPs) was released into the solution. The metal sulfide nanoparticles were measured by anodic stripping voltammetry (ASV) subsequently. This assay exhibited high sensitivity and selectivity with a detection limit of 0.48fM. In addition, we proved that this simple and cost-effective strategy is capable of detecting the target in complicated biological samples and holds great potential application in biomedical research and clinical diagnostics.

RESUMO

A new analysis method using ultra-performance liquid chromatography (UPLC)-quadrupole-time-of-flight mass spectrometry (Q/TOF MS) was developed for screening and confirmation of 12 ß-agonists in feeds. Under the optimized gradient elution conditions, it takes only 6 min to separate all 12 ß-agonists, and the critical pair of Ractopamine and Isoxsuprine isomers was almost completely separated. Based on the over 10,000 full-width at half maximum (FWHM) mass resolution and high mass accuracy features of TOF MS with 20 mDa mass window, accurate mass chromatograms were reconstructed for individual compounds. The fragmentation pathways of 12 ß-agonists were also studied using Q-TOF MS. The potential of UPLC-Q/TOF MS for confirmatory analysis was shown by determining the accurate mass of all compounds and fragment ions upon collision-induced-dissociation (CID) at different energies. The extra mass measurement errors for all ß-agonists were found to be within 5 ppm. The limits of detection (LODs) and limits of quantitation (LOQs) were determined for all ß-agonists in feeds and found to be 1-5 ng/mL and 5-50 µg/kg, respectively. The method of UPLC-Q/TOF MS developed in this study was initially applied to the research of feeds for analysis of 12 ß-agonists and proved to be accurate, sensitive, convenient and practical.

RESUMO

<p><b>OBJECTIVE</b>To investigate the impact of sub-chronic Aluminium-maltolate [Al(mal)3] exposure on the catabolism of amyloid precursor protein (APP) in rats.</p><p><b>METHODS</b>Forty adult male Sprague-Dawley (SD) rats were randomly divided into five groups: the control group, the maltolate group (7.56 mg/kg BW), and the Al(mal)3 groups (0.27, 0.54, and 1.08 mg/kg BW, respectively). Control rats were administered with 0.9% normal saline through intraperitoneal (i.p.) injection. Maltolate and Al(mal)3 were administered to the rats also through i.p. injections. Administration was conducted daily for two months. Rat neural behavior was examined using open field tests (OFT). And the protein expressions and their mRNAs transcription related with APP catabolism were studied using enzyme-linked immunosorbent assay (ELISA) and real-time polymerase chain reaction (RT-PCR).</p><p><b>RESULTS</b>The expressions of APP, β-site APP cleaving enzyme 1 (BACE1) and presenilin-1 (PS1) proteins and their mRNAs transcription increased gradually with the increase of Al(mal)3 doses (P<0.05). The enzyme activity of BACE1 in the 0.54 and 1.08 mg/kg Al(mal)3 groups increased significantly (P<0.05). The expression of β-amyloid protein (Aβ) 1-40 gradually decreased while the protein expression of Aβ1-42 increased gradually with the increase of Al(mal)3 doses (P<0.05).</p><p><b>CONCLUSION</b>Result from our study suggested that one of the possible mechanisms that Al(mal)3 can cause neurotoxicity is that Al(mal)3 can increase the generation of Aβ1-42 by facilitating the expressions of APP, β-, and γ-secretase.</p>

RESUMO

<p><b>OBJECTIVE</b>To study whether the effects of Leflunomide were different in treating patients with rheumatoid arthritis (RA) of different Chinese medical syndrome patterns.</p><p><b>METHODS</b>Totally 150 RA patients were recruited and assigned to 5 Chinese medical syndrome types, i.e., the heat-dampness blocking collateral type, cold-dampness blocking collateral type, Shen-qi deficiency cold type, Gan-Shen yin deficiency type, and stagnant blood blocking collateral type according to Chinese medical syndrome typing standards. They were treated with Leflunomide, 3 months as one therapeutic course. The parameters including numbers of joint tenderness and swelling, morning stiffness time, scores estimated by Visual Analog Scale (VAS), as well as laboratory indices involving rheumatoid factor (RF), erythrocyte sedimentation rate (ESR), C-reactive protein (CRP), American College of Rheumatology 20% improvement (ACR20), and American College of Rheumatology 50% improvement (ACR50) were observed before and after treatment, and statistically analyzed.</p><p><b>RESULTS</b>After treatment the numbers of joint tenderness, numbers of joint swelling, VAS scores, ESR, CRP, and RF all decreased, showing statistical difference when compared with those before treatment (P < 0.05). The morning stiffness time was shortened in the heat-dampness blocking collateral type, cold-dampness blocking collateral type, and stagnant blood blocking collateral type, showing statistical difference (P < 0.05). Of them, the numbers of joint tenderness, the numbers of joint swelling, the morning stiffness time, RF, VAS scores, and the improvement of the total effective rate were obviously better in the heat-dampness blocking collateral type, cold-dampness blocking collateral type, and stagnant blood blocking collateral type than in the Shen-qi deficiency cold type and Gan-Shen yin deficiency type, showing statistical difference (P < 0.05).</p><p><b>CONCLUSIONS</b>Leflunomide showed significant effects in treating RA. Of them, its effects were obviously better in the heat-dampness blocking collateral type, cold-dampness blocking collateral type, and stagnant blood blocking collateral type than in the Shen-qi deficiency cold type and Gan-Shen yin deficiency type.</p>

RESUMO

Glioblastoma, originating from glial cells, is the most commonly seen primary intracranial tumors in adults with extremely poor prognosis. miRNAs are a class of endogenous 18-26 nt non-coding RNAs playing important roles in a variety of biological processes. Recent studies have shown that miRNAs are closely associated with the development and progression of glioblastoma. In this paper, we summarize the regulatory roles of miRNAs in the progression of glioblastoma.

RESUMO

Eighteen novel triazole compounds containing thioamide were designed and synthesized. Their structures were confirmed by elemental analysis, (1)H NMR, IR, and MS. The title compounds exhibited certain antifungal activity. And the geometry structures of the title compounds were optimized by means of the density functional theory (DFT) method at B3LYP/6-31G( *) level. The quantitative structure-activity relationship (QSAR) of the title compounds was systematically investigated. A correlative equation between FA and DELH, V was well established by using the multiple linear regression (MLR).
