RESUMO

The COVID-19 pandemic has caused significant mobility restrictions and generated profound impacts on global socio-economic development. Mobility restrictions can generate significant impacts on the demand and supply sides of the rental housing market. By taking 77 large Chinese cities as cases, this research establishes a stepwise mediation effect test to evaluate the impacts of the pandemic on the rental housing market during Q1 2020. The results show that the confirmed cases were negatively associated with rental unit transactions, and the inter-city and intra-city movement played a significant role of mediating effects. Meanwhile, the impact of pandemic on rents lagged behind rental transaction in China's large cities, and the strict mobility controls caused the high vacancy rate of rental housing, leading to the bankruptcy of many housing rental agencies. Our research add to the burgeoning literature examining the mediating effect of mobility control between confirmed case and housing rental market. It demonstrates that the change of housing rental market induced by pandemic in China is the short-term influence on rental unit transaction, which is different from western countries. In China, a country with the most strict mobility control, the challenges come from the impact of pandemic on housing rental agencies.

RESUMO

The COVID-19 pandemic has resulted in millions of deaths and affected socioeconomic structure worldwide and the search for new antivirals and treatments are still ongoing. In the search for new drug target and to increase our understanding of the disease, we used large scale immunofluorescence to explore the host cell response to SARS-CoV-2 infection. Among the 602 host proteins studied in this host response screen, changes in abundance and subcellular localization were observed for 97 proteins, with 45 proteins showing increased abundance and 10 reduced abundances. 20 proteins displayed changed localization upon infection and an additional 22 proteins displayed altered abundance and localization, together contributing to diverse reshuffling of the host cell protein landscape. We then selected existing and approved small-molecule drugs (n =123) against our identified host response proteins and identified 3 compounds - elesclomol, crizotinib and rimcazole, that significantly reduced antiviral activity. Our study introduces a novel, targeted and systematic approach based on host protein profiling, to identify new targets for drug repurposing. The dataset of [~]75,000 immunofluorescence images from this study are published as a resource available for further studies.

RESUMO

The mechanisms underlying exercise-induced neuroprotective effects after traumatic brain injury (TBI) remained elusive, and there is a lack of effective treatments for TBI. In this study, we investigated the effects of an integrative approach of exercise and Yisaipu (TNFR-IgG fusion protein, TNF inhibitor) in a mouse TBI model. Male C57BL/6J mice were randomly assigned to a sedentary group or a group that followed a voluntary exercise regimen. The effects of 6-week prophylactic preconditioning exercise (PE) alone or in combination with post-TBI Yisaipu treatment on moderate TBI associated deficits were examined. The results showed that combined treatments of PE and post-TBI Yisaipu were superior to single treatments on reducing sensorimotor and gait dysfunctions in mice. These functional improvements were accompanied by reduced systemic inflammation largely via decreased serum TNF-α, boosted autophagic flux, and mitigated lesion volume after TBI. Given these neuroprotective effects, composite approaches such as a combination of exercise and TNF inhibitor may be a promising strategy for facilitating functional recovery from TBI and are worth further investigation.

RESUMO

Objective:To investigate the correlation between 131I uptake and therapeutic efficacy in metastatic differentiated thyroid carcinoma (DTC). Methods:The clinical data of 138 patients with metastatic DTC (42 males, 96 females, age range: 8-74 years) treated with 131I in nuclear medicine departments of 31 centers all over China were retrospectively analyzed. The lesional 131I uptake was quantitatively analyzed with target-to-nontarget (T/NT) ratio through the regions of interest in metastatic lesions confirmed by either planar or tomographic 131I SPECT/CT imaging. The efficacies of 131I treatment on the metastatic DTC were divided into complete remission (CR), partial remission (PR), stable disease (SD) and progress disease (PD) based on the change of the lesion diameter before and after the treatment. Factors which may affect therapeutic efficacy were assessed by the univariate (Kruskal-Wallis rank sum test, χ2 test and one-way analysis of variance) and binary logistic regression analyses. The receiver operating characteristic (ROC) curve of lesional T/NT ratio to predict the ineffectiveness of 131I therapy was performed. Results:A total of 1 165 efficacies were evaluated. The planar imaging results ( n=653) showed that there was no statistically significant difference of T/NT ratio among CR, PR, SD and PD groups ( χ2=4.15, P>0.05). The tomographic imaging results ( n=512) suggested CR, PR, SD and PD in 7.6%(39/512), 65.8%(337/512), 22.9%(117/512), and 3.7%(19/512) of individuals, respectively, and the T/NT ratio among the four groups was significantly different ( χ2=30.46, P<0.01). The univariate analysis also showed that age, stimulated thyroglobulin(sTg), 131I dose were the factors affecting therapeutic efficacy ( F or χ2 values: 2.561, 7.095 and 8.799, all P<0.05). Furthermore, binary logistic regression analysis revealed that older patients (odds ratio ( OR)=1.034, P=0.022) or patients with lower lesional T/NT ( OR=1.086, P=0.006) had a higher probability of ineffectiveness. The area under ROC curve for T/NT ratio to predict ineffectiveness was 0.726, and the cut-off value was 6.2, with a sensitivity of 78.7%(107/136) and a specificity of 73.1%(275/376). Conclusions:131I therapy is an effective treatment for metastatic DTC. The age at the time of metastatic diagnosis and the lesional T/NT ratio are independent influential factors for ineffectiveness of 131I therapy. When the leisonal T/NT ratio is lower than 6.2, the inefficiency of 131I is higher.

RESUMO

Mesenchymal stem cells (MSCs) have gained increasing attention as a potential approach for the treatment of bone injuries due to their multi-lineage differentiation potential and also their ability to recognize and home to damaged tissue sites, secreting bioactive factors that can modulate the immune system and enhance tissue repair. However, a wide gap between the number of MSCs obtainable from the donor site and the number required for implantation, as well as the lack of understanding of MSC functions under different in vitro and in vivo microenvironment, hinders the progression of MSCs toward clinical settings. The clinical translation of MSCs pre-requisites a scalable expansion process for the biomanufacturing of therapeutically qualified cells. This review briefly introduces the features of implanted MSCs to determine the best strategies to optimize their regenerative capacity, as well as the current MSC implantation for bone diseases. Current achievements for expansion of MSCs using various culturing methods, bioreactor technologies, biomaterial platforms, as well as microtissue-based expansion strategies are also discussed, providing new insights into future large-scale MSC expansion and clinical applications.

RESUMO

Prostate cancer is one of the most common malignancies in men worldwide. Altered expression of ARHGAP10, a member of the Rho GTPase activating protein (RhoGAP) family, has been found in several human cancers. However, its clinical significance in prostate cancer remains unknown. In the current study, we found that mRNA levels of ARHGAP10 were significantly higher in prostate cancer tissues than in the non-cancerous controls. Gene set enrichment analysis (GSEA) revealed that ARHGAP10 expression was negatively correlated with the Wnt signaling pathway. Immunohistochemical staining results showed that 62.2% (56/90) and 65.5% (59/90) of prostate cancer tissues displayed low expression of ARHGAP10 and high expression of ß-catenin, respectively. ARHGAP10 protein expression was significantly correlated with histologic grade (P < 0.0001), tumor stage (P = 0.0298), preoperative prostate specific antigen level (P = 0.0261), vital status (P = 0.0017) and ß-catenin expression (P < 0.0001). Kaplan-Meier survival analysis indicated that patients with low levels of ARHGAP10 and high levels of ß-catenin had poor overall survival. Multivariate analyses revealed that ARHGAP10 and ß-catenin expression was independent prognostic factor for prostate cancer. In summary, the current study suggests that ARHGAP10 in association with ß-catenin may play a role in the development of prostate cancer and serve as a prognostic factor for this disease.

RESUMO

With the rapid development of Internet, the number of online medical service review websites has gradually increased, whereby more and more patients use those engines to share their experience in health treatment, discuss the quality of medical services or rate physicians, and make more researches on medical institutions and doc-tors just to detailed information on the remedies of their health cases. This paper selected the UK and US as typical cases of countries that have rapidly developed in this field, and focused on UK’s NHS Choices and the United States’ RateMDs, Healthgrades and Yelp websites. Their current status, organization structure, content, characteristics, ex-perience and problems were analyzed and summarized. At present, China’s medical service online review is still in the ascendant, but the relevant policies and academic research are facing scarcity. Based on the situation in China and the experience from the UK and US, this paper proposed policy suggestions as follows: (1) Establishing a na-tional comprehensive medical service platform led by the government or professional and competent association which integrates health information query, retrieval and medical service review functions. (2) Set up a scientific and pro-fessional organization structure for standardized and orderly governance. (3) Combining the subjective reviews with objective quality indicators. (4) Adopting performance evaluation, and so on. All of these concerns are proposed in order to improve the medical service transparency and responsiveness, and to promote the medical service quality im-provement.

RESUMO

The study is to establish the two-dimension HPLC fingerprints of Dihuang (Rehmannia glutinosa), by HPLC-PDA and HPLC-ELSD methods. The separations were performed on Waters Atlantis®T3(4.6 mm× 250 mm,5 μm)and Welch Ultimate®Hilic-NH₂(4.6 mm× 250 mm,5 μm)columns with the gradient elution of acetonitrile-0.01% phosphoric acid and acetonitrile-water, respectively. The chromatographic display wavelength for PDA detector was set at 203 nm. For HPLC-ELSD, the nebulizer was set as cooling mode, the drift tube temperature was set at 60 °C and the gas pressure was 35.0 psi. Based on similarity evaluation system for chromatographic fingerprint of traditional Chinese medicine, 26 and 10 chromatographic peaks were determined as common components for HPLC-PDA and HPLC-ELSD fingerprints, respectively. Chemometrics analyses, such as similarity analysis; cluster analysis and principal component analysis, were performed on the common peak areas in two-dimension fingerprints for 41 batches of Dihuang from multiple sources. The results showed that the HPLC-PDA fingerprint could distinguish dried rehmannia root between different sources, and HPLC-ELSD fingerprint could differentiate dried rehmannia root from prepared rehmannia root. The two-dimension fingerprints were established with advantages of a good degree of separation, abundant chemical information and multi-components identified including two nucleosides (adenosine and uridine),four iridoid glycosides (catalpa alcohol, rehmaionoside D,rehmaionoside A and leonuride),two phenylethanoid glycosides (acteoside and cistanoside A) and nine sugars. The method is simple and practical, which could be used for the identification and quality assessment for Dihuang.

RESUMO

OBJECTIVE: This study investigated the effects of calcium on fluorosis-induced impairment in learning and memory of offspring rats. Methods Seventy-five newly weaned female Sprague-Dawley (SD) rats were randomly divided into five groups as follows: Control group (Control) drank tap water, and ate the normal diet (calcium content of 0.79%); fluoride group (F) drank 100 mg/L NaF solution, and ate the normal diet; low calcium group (LCa) drank tap water, and ate the low calcium diet (calcium content of 0.063%); low calcium fluoride group (F+LCa) drank 100 mg/L NaF solution, and ate the low calcium diet; high calcium fluoride group (F+HCa) drank 100 mg/L NaF solution, and ate the high calcium diet (calcium content of 7%). After exposing rats to fluoride for three months, male and female rats were mated and 14 and 28 days old offspring were obtained as experimental subjects. Examinations determined the submicroscopic parameters of the synaptic interface and expression levels of specific proteins: doublecortin (DCX) and synaptophysin (p38). RESULTS: (1) High fluorosis significantly reduced synapse density, length of synaptic active zone, thickness of postsynaptic density, and led to abnormal changes in the structural parameter of synaptic gap width, which was significantly reduced or increased. High dietary calcium significantly reversed the abnormal changes in structural parameters, and low calcium aggravated these variations. (2) Dietary calcium resulted in nonsignificant effect on expression levels of DCX and p38. CONCLUSION: The results suggested that dietary calcium significantly affected hippocampal synaptic plasticity of offspring of mothers exposed to water fluorosis, but its molecular mechanism may not be related to the expression of DCX and p38 in the brain. The findings also demonstrate the important effects of maternal exposure to water fluorosis on offspring brain functions before water improvement.

RESUMO

Objective To evaluate the association between iodine intake and treatment outcomes of radioiodine remnant ablation in patients with papillary thyroid cancer (PTC),and to investigate the determinants related to the ablation efficacy. Methods A total of 95 PTC patients (28 males,67 females;average age 39.8 years) without distant metastases from January 2013 to May 2014 were enrolled in this retrospective study. All patients underwent total thyroidectomy and 2-4 weeks of low iodine diet (LID) before initial 131I therapy.Patients were divided into 3 groups according to urinary iodine excretion (UIE):moderate-severe iodine deficiency group (0<UIE<50 μg/L,n =30),mild iodine deficiency group (50 μg/L≤UIE<100 μg/L,n =26),adequate iodine group (100 μg/L ≤ UIE < 300 μg/L,n =39).Patients were followed up for 3-6 months after radioiodine ablation, successful ablation was defined as no visible uptake in the thyroid bed on diagnostic 131I whole body scan and Tg level <2 μg/L (with negative TgAb),or no visible uptake in thyroid bed on posttreatment 131I whole body scan.x2 test,two-sample t test,Mann-Whitney u test and logistic regression analysis were performed. Results In all,84.2% (80/95)of patients were successfully ablated. The successful rates in the three iodine intake groups were 96.7% (29/30),84.6% (22/26) and 74.4% (29/39),respectively,with significant difference (x2=7.374,P<0.05).Univariate analysis revealed that UIE,pre-treatment TSH,pre-treatment Tg and the amount of remnant thyroid tissue at ablation affected ablation efficacy (x2 =7.374,t =2.037,z =-2.966,x2 =4.144,all P<0.05).Logistic regression showed that the level of pre-treatment Tg (P < 0.05) and iodine intake (P < 0.05) were independent factors of ablation efficacy. Conclusion Iodine intake before 131I remnant ablation is one of the important factors affecting treatment outcomes.Thyroid remnant could be more successfully ablated if reasonable LID protocols be adopted according to the iodine nutritional status before treatment.

RESUMO

The soil respiration, nitrification and denitrification processes play an important role on soil nitrogen transformation and diffuse nitrogen loading. These processes are also the chains for soil circle. In this study, the Zhegao watershed located north of Chaohu Lake was selected to explore the interactions of these processes with diffuse nitrogen pollution. The BaPS (Barometric Process Separation) was applied to analyze the soil respiration, nitrification and denitrification processes in farmland and forest. The SWAT (Soil and Water Assessment Tool) simulated the temporal and spatial pattern of diffuse nitrogen loading. As the expanding of farmland and higher level of fertilization, the yearly mean loading of diffuse nitrogen increased sustainably from 1980-1995 to 1996-2012. The monthly loading in 1996-2012 was also higher than that in the period of 1980-1995, which closely related to the precipitation. The statistical analysis indicated that there was a significant difference between two periods. The yearly averaged loading of the whole watershed in 1996-2012 was 10.40 kg x hm(-2), which was 8.10 kg x hm(-2) in 1980-1995. The variance analysis demonstrated that there was also a big difference between the spatial distributions of two periods. The forest soil had much higher soil respiration than the farmland soil. But the farmland had higher nitrification and denitrification rates. The more intensive nitrogen transformation in the farmland contributed to the less diffuse nitrogen loading. As the nitrification rate of farmland was higher than denitrification rate, agricultural diffuse nitrate nitrogen loading would increase and organic nitrogen loading would reduce. The analysis of soil respiration, nitrification and denitrification is helpful for the study of soil nitrogen circle form the aspect of soil biology, which also benefits the control of agricultural diffuse nitrogen pollution.

RESUMO

Objective To investigate the application effect of anterior clinoid process drilled off via epidural approach in posterior communicating artery aneurysm ( PCoAA) clipping. Methods The clinical data of 42 patients with PCoAA who underwent craniotomy from January 2012 to January 2014 were analyzed retrospectively, including 22 patients performed anterior clinoid process drilled off and 20 did not. The difficult or easy degree of intraoperative aneurysm clipping and postoperative efficacy were analyzed. Results The aneurysms in 22 patients underwent anterior clinoid process were clipped satisfactorily. The brain retractor was not used during the procedure. Only one patient had cerebral infarction after procedure. No patients had oculomotor nerve paralysis and incomplete clipping of aneurysms. Of the 20 patients without the anterior clinoid process drilled off,3 aneurysms were clipped incompletely because it was difficult to implant aneurysm clips, 2 had cerebral infarction, and 1 had oculomotor nerve paralysis. Conclusion Removing the anterior clinoid process drilled off via extradural approach may bring convenience for PCoAA clipping. It can effectively avoid the difficulty of implanting aneurysm clips during the procedure. Its application is safe and can reduce postoperative complications.

RESUMO

OBJECTIVETo assess the value of (18)F-FDG dual head coincidence imaging in the prediction of the efficacy of radioiodine therapy in patients with cervical lymph node (LN) metastasis of papillary thyroid carcinoma (PTC).

METHODSThirty-six patients undergoing thyroidectomy and radioiodine ablation of the residual normal thyroid tissue received (18)F-FDG dual head coincidence imaging and then therapeutic (131)I-whole body imaging ((131)I-WBI) in the same week. According to those imaging results, the patients were divided into group I with positive results of both imaging examinations and group II with positive results by (131)I-WBI but negative results by(18)F-FDG dual head coincidence imaging. All the patients were followed up for 6 months.

RESULTSIn group I (14 patients), a total of 49 lesions were diagnosed as cervical LN metastases, and the total sensitivity differed significantly between (18)F-FDG dual head coincidence imaging and (131)I- WBI (67.3% vs 89.8%, P=0.027). In both groups, the total sensitivity of (18)F-FDG dual head coincidence imaging and (131)I-WBI showed a significant difference (26.0% vs 94.5%, P<0.001). The target and non-target ratio (T/NT) was identified as one of the factors affecting the radioiodine efficacy (P<0.001). In group II (22 patients), 76 lesions were diagnosed as cervical LN metastases. The effective rates of groups I and II were 35.7% and 81.8%, respectively, showing a significant difference between them (P=0.011).

CONCLUSION(131)I-WBI is more sensitive than (18)F-FDG dual head coincidence imaging in detecting cervical LN metastasis in patients with PTC. Patients with cervical LN metastases who have positive results in both (131)I-WBI and (18)F-FDG dual head coincidence imaging tend to have a poorer response to the therapy than the patients with negative results in (18)F-FDG dual head coincidence imaging. The T/NT of the cervical LN metastases in (18)F-FDG dual head coincidence imaging is associated with the efficacy of radioiodine therapy.

RESUMO

OBJECTIVETo evaluated the safety and efficacy of hyperthermic intraperitoneal perfusion chemotherapy(HIPC) in the prevention and treatment of pseudomyxoma peritonei (PMP) recurrence after cytoreductive surgery(CRS).

METHODSStudies published in English before 2010 on HIPC after CRS for PMP were searched in PubMed database. Each study was carefully evaluated based on pre-determined criteria. Study results were comprehensively displayed in a form. A descriptive systematic review was performed.

RESULTSA total of 11 studies were included. The median survival time of patients in these studies ranged from 25.6 months to 156 months. The ranges of 1-year, 2-year, 3-year, 5-year, and 10-year survival rates were 72%-100%, 55%-96%, 59%-96%, 52%-96%, and 55%-96%, respectively. The overall complication rate ranged from 2%-15%, and the total perioperative mortality were from 0 to 7%.

CONCLUSIONHIPC after CRS is effective and safe for patients with PMP.
