Objective@#To investigate the effect of adipose tissue-derived mesenchymal stem cell (ADSC) transplantation in the treatment of liver fibrosis rats and possible mechanism.@*Methods@#Subcutaneous adipose tissue in the inguinal region of rats was collected to isolate ADSCs. The rats with liver fibrosis induced by intraperitoneally injected carbon tetrachloride were divided into cell transplantation group and phosphate buffer saline (PBS) injection group, and the rats which were fed normally were enrolled as negative control group. The rats in the cell transplantation group were given tail vein injection of ADSCs, and those in the PBS injection group were given injection of 0.5 ml PBS. At 7 days after transplantation, blood samples were collected from the inferior vena cava to evaluate liver function; liver tissue was collected to measure the protein expression of hepatocyte growth factor (HGF) and alpha-smooth muscle actin (α-SMA); Masson trichrome staining was used to evaluate intrahepatic collagen deposition. Hepatic stellate cells (HSCs) were collected from the rats with liver fibrosis, and indirect co-culture of HSCs and ADSCs was performed in vitro to analyze the influence of ADSCs on the proliferation and apoptosis of HSCs. The independent samples t-test was used for comparison between groups, and an analysis of variance was used for comparison of means between multiple samples.@*Results@#ADSCs were found in liver tissue in the transplantation group, and compared with the PBS injection group, the transplantation group had significant alleviation in hepatocyte necrosis, vacuolization, and area of fibrosis and significant reductions in the serum levels of aminotransferases, while there was no significant difference in the level of albumin between the two groups. Compared with the PBS injection group, the transplantation group had significant upregulation in the protein expression of HGF and significant downregulation in the protein expression of α-SMA (both P < 0.05). In vitro co-culture for 72 hours showed that ADSCs inhibited the proliferation of HSCs, and there was a significant difference between the co-culture group and the control group with HSCs cultured alone. Caspase-3 immunostaining showed that after co-culture for 72 hours, there was a significant difference in the apoptosis rate of HSCs between the co-culture group and the control group with HSCs cultured alone (23.42% ± 3.02% vs 14.82% ± 3.93%).@*Conclusion@#ADSC transplantation can upregulate the expression of HGF in the liver, promote the apoptosis of HSCs, and thus alleviate liver fibrosis.

Objective To investigate the expression of DNM T1 and PDCD4 in Tongue Squamous cell carcinoma and their signif‐icance to Clinical .Methods The protein expression of DNM T1 and PDCD4 in 40 cases of TSCC tissues and The adjacent no tumor tissues were measured by Elivision Two Step immunohistochemical method(IHC) ,the relationship between DNM T1 ,PDCD4 and clinicopathological parameters were analyzed .Results There was positively over expression of DNM T1 while the expression of PD‐CD4 was at a low lever or lost in TSCC tissues ,the expression of two genes DNM T1 and PDCD4 in the adjacent no tumor tissues were in contrast ;there was a significant negative correlation between DNM T1 and PDCD4 expression in TSCC(r = - 0 .452 ,P<0 .05) .The expression of DNM T1 protein were associated with histopathological differentiation types ,regional lymph node metasta‐sis and TNM staging(P< 0 .05) ,it had nothing to do with age and gender ;the expression of PDCD4 protein were associated with regional lymph node metastasis and TNM staging(P< 0 .05) ,and it had nothing to do with age ,gender ,histopathological differenti‐ation types .Conclusion Implying the abnormal expression of DNM T1 and PDCD4 protein might be closely related to the develop‐ment and progression of TSCC .DNM T1 may be participates in the inactivation of PDCD4 expression ,and led to the development of the cancer at last .

Objective To explore the effects of andrographolide(AD) on proliferation inhibition ,cells cycle distribution and the expression of Bax and Bcl‐2 mRNA in human tongue squamous cell carcinoma Tca8113 cells .Methods MTT was used to measure the levels of the proliferation of Tca8113 cells cultured with different concentraions of AD .Cell cycles were analyzed by flow cytom‐etry .RT‐polymerase chain reaction(RT‐PCR)technique was used to evaluate Bcl‐2 mRNA and Bax mRNA expression .Results An‐drographolide inhibited Tca8113 cells growth in a time and dose‐dependent manner ,the IC50 was 74 .66 μg/mL .A feature of apop‐tosis was observed under microscope .With a increase in concentration and increase in time ,the cells in G0/G1 phase increased from (39 .45 ± 0 .65)% to (63 .70 ± 0 .65)% ,the cells in S phase decreased from (56 .55 ± 0 .64)% to (32 .28 ± 0 .54)% .Statistics showed tha the cells arrested in G0/G1 phase .It was statistially significant in comparison with the control group (P< 0 .01) .AD could up‐regulate the experssion of Bax mRNA ,and AD could down‐regulate the experssion of Bcl‐2 mRNA .Conclusion An‐drographolide inhibited the growth of Tca8113 cells and increased the cell apoptosis .It may be promising as a new drug for treat‐ment of tongue squamous cell carcinoma .

Objective To investigate clinicopathologic characteristics of malignant tumors initiated with cervical lymph nodes enlargement.Methods Clinical and pathological data of 887 cases with cervical malignant tumors kept in the college during January 1998 to December 2003 were analyzed for their histological patterns and primary site of metastatic carcinoma.Results Cervical cancer with lympb nodes metastasis was mostly seen at age of 51~60 years (182/837,21.74%), and much more common in men than in women. Major metastatic carcinomas in cervical lymph nodes originated from the nasopharynex, lung, thyroid, stomach and esophagus (591/671,88.08%), with 60.66%(407/671)of squamous cell carcinoma mostly from the nasopharynex, lung, or esophagus (380/407,93.37%). There was significant difference in the site, size, number, quality, appearance of lymph nodes, course of disease, and clinical manifestations between lymphoma and metastatic cancer.Conclusions Cervical metastatic carcinomas in Guangdong mainly originated from the nasopharynx. Some specific immunohistochemical markers, CT, MRI and ultrasonic technology played a very important role in diagnosis for lymphoma and determination of primary site of metastatic cancer.

<p><b>BACKGROUND</b>To find a rapid and sensitive method for early diagnosis of nasopharyngeal carcinoma by using EBV TK kinase.</p><p><b>METHODS</b>Prokaryotic expression plasmid pRSETTK was constructed. EBV TK kinase was highly expressed in E.coil BL21 (DE3). The authors identified specificity of TK kinase by Western blot, then used purified TK kinase in ELISA to detect the IgG antibody in the serum of NPC patients.</p><p><b>RESULTS</b>Specific IgG antibody against TK kinase was found in the serum of NPC patients. The specificity and sensitivity of TK kinase were both 100% in Western blot and were 98.0% and 93.4% respectively in ELISA.</p><p><b>CONCLUSIONS</b>The EBV TK kinase showed high specificity and sensitivity in ELISA, therefore it can be used for early diagnosis of NPC</p>

Objective To explore the relationship between expressions of VEGF-C, VEGF-D and metastasis in papillary carcinoma of the thyroid. Methods Immunohistochemical method was used to detect the microvessel density(MVD) and expressions of VEGF-C and VEGF-D protein in 20 samples of nodular goiter and 115 samples with papillary carcinoma of the thyroid. Results The positive rates of VEGF-C and VEGF-D protein in nodular goiter were 15.0% and 20.0% respectively, and those in papillary carcinoma of the thyroid were 87.0% and 72.2%, respectively. The intratumour MVD in nodular goiter and papillary carcinoma of the thyroid was 15.2?6.7 and 21.5?8.5 respectively. The expressions of VEGF-C, VEGF-D protein and MVD in papillary carcinomas of the thyroid were significantly higher than those of nodular goiter. Objective To evaluate the significance of p53,Cdk_4 and Survivin in gastric epithelial dysplasia with colonic and small intestinal metaplasia.Methods Immunohistochemical SP method was used to detect p53,Cdk_4,and Survivin in 44 cases of colonic intestinal metaplasia(27 low grade epithelial dysplasia,17 high grade epithelial dysplasia) and 51 cases of small intestinal metaplasia(42 low grade epithelial dysplasia,9 high grade epithelial dysplasia) by endoscope.Results The positive expression rates of p53 and Survivin had significant difference in two types of intestinal metaplasia and the difference was significant between low-grade and high-grade epithelial dysplasia.The expression of CDK_4 had significant difference in two types of intestinal metaplasia and had not difference between low-grade and high-grade epithelial dysplasia.Conclusion The expressions of p53,CdK_4 and Survivin are closely associated not only with the types of intestinal metaplasia but also with the different grades of epithelial dysplasia.The expression of p53,Cdk_4,and Survivin may be valuable markers in assessing the biological behavior of gastric epithelial dysplasia with intestinal metaplasia.

RESUMO

Sodium pyridinethione, in vitro, possesses a significant inhibitory effect on human tumor cell strains, such as Hela, CNE-2 and Raji cell strain. In vivo, it has not inhibitory effect on the transplanted tumors, such as S180, LLC and EC, but significantly enhance the inhibitory effects of 5-Fu on S180.Sodium pyridinethione markedly decreases the weight of spleen and thymus, and serum hemolysins reaction induced by SRBC in mice and skin delayed type hypersensitivity reaction induced by DNCB in guinea pigs It also inhibits lyphocyte transformation induced by PHA. All these effects are shown clear dose-response relationship. Our experiments showed that sodium pyridinethion is a new antitumor and immuno-inhibitory compound.
