Objective@#To investigate the effect of silencing human leukocyte⁃associated antigen⁃G ( HLA⁃G) expression in the chorionic trophoblast cell line JEG⁃3 cells under hypoxic conditions on the biological function of JEG⁃3 cells and through the hypoxia response pathway of endothelial PAS1 region protein 1 (EPAS1)is involved in the molecular mechanism of preeclampsia under high altitude hypoxia. @*Methods@#The expression of HLA⁃G in JEG⁃3 cells was inhibited by transfection of small interfering RNA (siRNA) . The JEG⁃3 cells after transfection were divided into four groups : normoxic control group , hypoxic control group , normoxic inhibition group , and hypoxic inhibition group. CCK⁃8 test and Transwell test were used to detect the proliferation and invasion ability of the cells in four groups ; The effects of four groups of apoptosis and cell cycle were detected by flow cytometry; HLA⁃G and EPAS1 mRNA and protein expression levels were detected by real⁃time fluorescence quantitative PCR ( qPCR) and Western blot.@*Results @#① Compared with the normoxic control group , hypoxic control group , normoxic inhibition group , and hypoxic inhibition group could reduce the proliferation activity and invasion ability of JEG⁃3 cells , and significantly increase the apoptosis rate. The hypoxic control group and hypoxic inhibition group also produced anobvious necrotic cell population ; Under the condition of hypoxia , after reducing the expression of HLA⁃G , the cell necrosis rate was further aggravated ; Whether under normoxia or hypoxia , inhibition of HLA⁃G expression caused the cells to be blocked in the G1 phase. ② Compared with the normoxic control group , hypoxia control group , normoxic inhibition group , and hypoxia inhibition group decreased the expression of HLA⁃G protein , and hypoxia and inhibition of HLA⁃G had a synergistic effect; Hypoxia⁃inducible factors⁃2α ( HIF⁃2α ) , vascular endothelial growth factor(VEGF) and endothelin⁃1(ET⁃1) protein expression could be added , inhibition of HLA⁃G decreased the expression of inducible nitric oxide synthase( NOS2) . @*Conclusion@#In the hypoxic environment , silencing HLA⁃G may affect the biological behavior of trophoblast through the EPAS1 hypoxic response pathway and participate in the occurrence and development of preeclampsia.

To quantitatively evaluate the upper-limb spasticity of stroke patients in recovery stage, the relationship between surface electromyography (sEMG) characteristic indexes from biceps brachii and triceps brachii and the spasticity were explored, which provides the electrophysiological basis for clinical rehabilitation. Ten patients with spasticity after stroke were selected to be estimated by modified Ashworth (MAS) assessment and a passive elbow sinusoidal motion experiment was carried out. At the same time, the sEMG of biceps and triceps were recorded. The results shows that the reflex electromyographic threshold could reflect the physiological mechanism of spasticity and had significant correlation with MAS scale which showed that sEMG could be prosperous for the clinical quantitative evaluation of spasticity of stroke patients.

RESUMO

Objective To evaluate and analyze the characteristics of cognitive and memory disorders in patients suffering cerebral infarction. Methods The scores of overall cognitive function including orientation to time and place, immediate memory, calculation, delayed memory, naming, repeat, understanding, reading, writing, visuospatial function were evaluated by mini-mental state estimate (MMSE) and the scores of memory function including immediate learning and memory , short time delayed recall , long time delay memories were evaluated by WHO/UCLA Auditory Verbal Learning Test. Statistical analysis were done in patients with cerebral infarction and healthy controls. Results The scores of overall cognitive function, memory scores as well as immediate recall , short-delayed recall , long-delayed recall in cerebral infarction patients were much lower than those of healthy controls (P < 0.001). Conclusion The overall cognitive function and memory function declined significantly , also immediate memory and short term memory were remarkable impaired in patients with cerebral infarction, resulting their memory characteristics were similar to patients with Alzheimer′s disease.

Objective To investigate the effects of intensive training for mimetic muscle on idiopathic facial paralysis. Methods 105 pa-tients with idiopathic facial paralysis were divided into control group (n=53) and treatment group (n=52). All the patients received the rou-tine treatment, including drugs, physical therapy, acupuncture and psychological support, while the treatment group was treated with inten-sive training for mimetic muscle in addition. Results 1 month after treatment, the muscle strength improved more in the treatment group than in the control group (P<0.01). The incidence of cure was 65.38%in the treatment group, more than 39.62%in the control group (P<0.01), as well the the total improvement of 96.15%and 83.02%respectively (P<0.05). Conclusion The intensive training for mimetic mus-cle can improve recovery of facial paralysis.

RESUMO

OBJECTIVE: To establish an animal model of myocardial ischemia with blood stasis syndrome in mini-swines. METHODS: An animal model of myocardial ischemia was established in mini-swines by oppressing the coronary artery through the expansion of inner layer of Ameroid constrictor and the Ameroid constrictor was implanted into the distal end of the initial part of the first branch of interventricular septum of ramus descendens anterior arteriae coronariae sinistrae. Dynamic observation of behavior changes, general health status and changes of hemorheological parameters in the mini-swines were made after operation. RESULTS: The coronary angiography showed that the stenosis rate in ischemic group was more than 75% four weeks after operation. Compared with before operation and sham-operated group, there were great changes of behavior, general health status, tongue color and hemorheological parameters in ischemic group (P<0.05). CONCLUSION: The animal model of myocardial ischemia with blood stasis syndrome in mini-swines was established successfully 4 weeks after operation. The pathological process in the animal model is similar to that in the patients with chronic myocardial ischemia. So this model can be adopted in the research of myocardial ischemia with blood stasis syndrome.

RESUMO

OBJECTIVE:To evaluate the economic efficiency of three oral administration schemes in treatment of benign prostatic hypertrophy(BPH).METHODS:73patients with BPH were divided into three groups:group A,Pule’an,group B,Alfuxosin hydrochloride,group C,Finasteride.Data was evaluated using the pharmacoeconomic cost-effectiveness analysis. RESULTS:The effective rates in the group B and group C were higher than that in group A,the effective rate in group B was comparable to that in group C.The cost-effectiveness ratios of A,B,C were1.30,1.80,4.36,respectively.Using sensitivity analysis,the cost-effectiveness ratios were1.17,1.62,3.92,respectively.CONCLUSION:Among the three schemes,scheme B was the best one.
