RESUMO

OBJECTIVES: To report clinical features, gross post mortem and histopathological findings from an investigation into sudden or unexpected death in rabbits that was undertaken during an outbreak of rabbit haemorrhagic disease. MATERIALS AND METHODS: Using a standard protocol, veterinarians were invited to submit case histories and results of their post mortem examination of pet rabbits that died unexpectedly. Histopathological examination of heart, lungs, liver, spleen and kidney samples was collated with macroscopic appearance and clinical details. RESULTS: Hepatocellular necrosis, characteristic of rabbit haemorrhagic disease, was observed in 185 of 300 (62%) submissions, often accompanied by glomerular thrombosis and changes in other organs. Evidence of rabbit haemorrhagic disease was not apparent on histopathology in 113 of 300 (38%) rabbits. Gross post mortem examination by veterinary practitioners did not always reflect reported histopathological changes. No macroscopic abnormalities were seen in 78/185 (42%) of rabbit haemorrhagic disease cases. Rapid death and death of other rabbits in the household were common features of rabbit haemorrhagic disease. Ante mortem clinical signs included anorexia, collapse, lethargy, seizures, icterus, bleeding from the mouth, dyspnoea, hypothermia, pyrexia, bradycardia or poor blood clotting. CLINICAL IMPORTANCE: Rabbit haemorrhagic disease can be suspected from a history of sudden death, especially if multiple rabbits are affected. There is not always macroscopic evidence of the disease but histopathology is useful to support or refute a diagnosis of rabbit haemorrhagic disease and provide information about other causes of death.

The histological and immunohistochemical features of 13 cases of suspected vaginal fibroepithelial polyps are reported. The characteristic microscopical features of these lesions were an abundant oedematous or fibrous stroma containing spindle-shaped and stellate cells and the presence of variable inflammation and haemorrhage. There was often a superficial layer of compressed tissue, but the stroma in the peripheral areas of the masses was generally more loosely arranged than in central areas. The connective tissue cells expressed vimentin and desmin, but did not express smooth muscle actin or calponin. Individual cases had additional changes including granulomatous inflammation, epithelial dysplasia suggestive of papillomavirus infection and a lesion resembling phyllodes tumour in women.

Histopathological examination was carried out on post mortem samples of liver from 12 donkeys (Equus asinus), aged 21-57 years (4 females, 1 stallion, 7 geldings). Variable amounts of haemosiderin were present in Kupffer cells, portal macrophages and hepatocytes in all cases. In all cases there was infiltration of connective tissue around portal tracts by variable numbers of inflammatory cells (lymphocytes, plasma cells and macrophages) but obvious portal fibrosis was present in only four animals. Subjective assessment of overall haemosiderin staining (including extent and intensity) generally reflected biochemical measurements of liver iron content (measured by an inductively-coupled plasma method) as well as quantitative histochemical measurements (using an image analysis package and sections stained with Perl's Prussian blue stain). Accumulation of hepatic iron in old donkeys was not directly related to other pathological changes and may be an incidental finding.

Post mortem liver samples from 12 donkeys (Equus asinus) aged 21-57 years (4 females, 1 stallion, 7 geldings), were assessed chemically for copper and iron content on a wet weight basis and histologically for stainable iron. Chemical liver copper content ranged from 2.7 to 4.8µg/g (mean 3.5±0.05µg/g). Chemical liver iron content ranged from 524 to 5010µg/g (mean 1723±1258µg/g). Histochemical iron was measured morphometrically using a computer-based image analysis system; percentage section area staining for iron ranged from 0.84% to 26.69% (mean 10.82±8.36%). There was no clear correlation, within the wide range of iron values, between histochemically demonstrable iron and chemically measured iron content. No clear age-related increase was apparent for either parameter in these aged donkeys. The accumulation of iron in the liver of donkeys may represent a physiological haemosiderosis rather than pathological haemochromatosis.

A histological, histochemical and morphometric study was performed on submandibular salivary glands from 13 dogs which had presented with a submandibular mass or swelling that proved to be a portion of non-inflammatory and non-neoplastic submandibular salivary gland. There were no consistent changes in lectin-binding histochemistry or immunohistochemical expression of various cell markers, and, in most cases, there was no measurable difference in acinar size in the affected gland. The possible explanation for the clinical salivary gland enlargement is therefore unclear.

RESUMO

Plexiform vascularisation of a retropharyngeal lymph node is described in an adult cat. The cat presented with a chronic history of inspiratory stridor and a slowly growing mass in the cranial cervical area. Clinical signs resolved after excision of the affected node. This appears to be the first clinical report of plexiform vascularisation of a retropharyngeal lymph node and its treatment in a cat.

RESUMO

Three cats with upper respiratory tract obstruction due to laryngeal inflammation are presented. Cervical radiography showed the presence of a soft tissue mass in the laryngeal region in all cases, and laryngoscopy allowed direct visualization of a mass associated with the larynx. Laryngeal samples were obtained by a combination of fine needle aspiration, cutting biopsy forceps, by ventral laryngotomy, and at post-mortem. Histopathology of the laryngeal samples showed the presence of a predominantly granulomatous inflammation, with macrophage and lymphocyte infiltration. One case was euthanased due to severe dyspnoea. The remaining two cases underwent combined medical (corticosteroid and antibiotic) and surgical (permanent tracheostomy or excision of laryngeal tissue by ventral laryngotomy) treatment. One case died of an undetermined cause 15 weeks after surgery while the other case remains clinically well 20 months after diagnosis. Recognition of the existence of granulomatous laryngitis is important as clinical signs and radiographic findings are indistinguishable from laryngeal neoplasia.

RESUMO

Benign connective tumours of the canine salivary glands are rare. This report describes lipomatous infiltration of parotid or submandibular salivary glands in seven dogs in which the glands were enlarged as a result of infiltration by fat cells; they appeared to have been successfully treated by local excision. The precise cause of the lipomatous infiltration in the dogs is unclear but different causes of similar lesions in humans are discussed.

RESUMO

A connective tissue disease resembling the human Ehlers-Danlos syndrome is reported in two sibling rabbits about four months old. The clinical signs included skin hyperextensibility and fragility and poor wound healing. There were ultrastructural abnormalities in the structure of the collagen fibrils and in their arrangement in bundles of fibres. Affected rabbits may be a useful laboratory animal model for collagen disorders in human beings.

A distinctive lymphadenopathy in nine cats, four of which were of the Longhair breed, was characterized by capillary vasoproliferation and lymphoid atrophy. It occurred in the cervical or inguinal regions. The animals were, apart from local manifestations, healthy at the time and remained so subsequently. In the absence of any clinical or morphological evidence for viral, neoplastic or developmental aetiologies, we postulate an ischaemic pathogenesis for the lesions which may be related to human "vascular transformation of lymph node sinuses". The lesions appear to be by no means a rare cause of non-neoplastic lymphadenopathy in domestic cats, since they constituted over 5 per cent of surgical biopsies of cat lymph nodes seen in the past 5 years. The lesions merit wider recognition and investigation.

A method for examining lymphocyte blastogenesis in whole blood cultures of neonatal calves is presented. Considerable variation in magnitude of thymidine incorporation was noted between animals but the general trend of response was uniform. Maximum responses occurred at different culture times for each mitogen. In a standard 0.2 ml micro culture system using 10,000 mononuclear cells per culture incubated for 96 hours in 5 per cent fetal bovine serum maximum mitogenic responses were obtained with 0.8 microgram phytohaemagglutinin per culture, 1 microgram concanavalin A per culture and 0.08 microgram pokeweed mitogen per culture.

RESUMO

Serum immunoglobulin concentrations in clinically healthy German shepherd dogs were compared with those in mongrel dogs and Irish setters. Serum IgA concentrations were significantly lower in the German shepherd dogs than in the other two groups, while there were no differences in the concentrations of IgM or IgG. These findings suggest that production of IgA by gut associated lymphoid tissue may be abnormally low in the German shepherd dog.
