An outbreak of 92 abortions out of 1,700 pregnant cows (5.41%) in a period of 3 weeks (19 May to 05 June 2019) occurred in a Georgia Dairy, USA, in cattle that were between 3 and 7 months of gestation. Two sets of samples (aborted fetuses' organs, placental tissues, aborted cows blood) were submitted for laboratory investigations at the Tifton Veterinary Diagnostic and Investigational Laboratory, College of Veterinary Medicine, University of Georgia (TVDIL, Tifton, GA, USA). An abortion panel testing for the major abortion-causing agents [e.g. Bovine Viral Diarrhoea Virus (BVDV), Infectious Bovine Rhinotracheitis Virus/ Bovine Herpes Virus-I (IBR/ BHV-I), Brucella spp., Leptospira spp.] was conducted on several of the samples. On the first set of samples, microbial cultures, serology and PCR tests for the common abortifacient agents revealed the presence of Neospora caninum (N. caninum) DNA, which was positive by PCR on the placenta and fetal tissues. The second set of diagnostic investigations also identified two out of three submitted freshly aborted fetuses to be positive for N. caninum by PCR and immunohistochemistry. Moreover, all three dams were also sero-positive for N. caninum. The entire herd was being fed on grass silage harvested from a pasture where feral pigs were hunted previously and carcasses were left behind. As a consequence of this action a large population of wild coyotes were attracted to these carcasses, and likely contaminated the pasture with potential N. caninum-infected feces. After the abortion outbreak was resolved, it was recommended that the farmers should avoid disposal of cadavers of hunted animals in the wild, as it could attract carnivorous and omnivorous animals that may potentially spread the disease to the cattle and other wildlife.

Despite the likelihood that a horse's mass influences hoof morphology, empirical evidence is lacking. A clearer understanding of factors influencing hoof shape could enable prevention, or better treatment, of foot-based disorders; common causes of equine lameness. The study's aim was to investigate the relationship between horse body size, in terms of mass and height, and fore hoof dimensions. A further aim was to determine changes in the occurrence of hoof asymmetry as body size increases. Height, mass, and fore hoof dimensions; coronet band width (CBW); hoof base width (HBW); dorsal hoof wall angle (DHWA); and hoof spread (HS) of 63 riding school horses were measured within two weeks of routine shoeing. Regression analysis demonstrated positive relationships between body mass and both CBW and HBW in left and right hooves, indicating basic hoof dimensions increased as body mass increased. No relationship between horse height and hoof variables was found, suggesting mass is more influential on hoof morphology. Left and right DHWL were moderately correlated; however, paired t-test results identified a greater right than left DHWA. As left DHWA increased, left HS decreased, indicating development of a more upright hoof geometry. Both left and right HS increased as corresponding HBW increased. Both hooves tended toward a more upright conformation as horse height and body mass increased. However, asymmetries observed suggest a splayed left hoof compared to a "boxy" right hoof. Such morphological adjustments may indicate variation in horn tubule orientation in response to greater structural loading; an important consideration for hoof practitioners.

Johne's disease (JD) caused by Mycobacterium avium subspecies paratuberculosis (MAP) is a major threat to the dairy industry and possibly some cases of Crohn's disease in humans. A MAP vaccine that reduced of clinical disease and/or reduced fecal shedding would aid in the control of JD. The objectives of this study were (1) to evaluate the efficacy of 5 attenuated strains of MAP as vaccine candidates compared to a commercial control vaccine using the protocol proposed by the Johne's Disease Integrated Program (JDIP) Animal Model Standardization Committee (AMSC), and (2) to validate the AMSC Johne's disease goat challenge model. Eighty goat kids were vaccinated orally twice at 8 and 10 weeks of age with an experimental vaccine or once subcutaneously at 8 weeks with Silirum® (Zoetis), or a sham control oral vaccine at 8 and 10 weeks. Kids were challenged orally with a total of approximately 1.44 × 10(9) CFU divided in two consecutive daily doses using MAP ATCC-700535 (K10-like bovine isolate). All kids were necropsied at 13 months post-challenge. Results indicated that the AMSC goat challenge model is a highly efficient and valid model for JD challenge studies. None of the experimental or control vaccines evaluated prevented MAP infection or eliminated fecal shedding, although the 329 vaccine lowered the incidence of infection, fecal shedding, tissue colonization and reduced lesion scores, but less than the control vaccine. Based on our results the relative performance ranking of the experimental live-attenuated vaccines evaluated, the 329 vaccine was the best performer, followed by the 318 vaccine, then 316 vaccine, 315 vaccine and finally the 319 vaccine was the worst performer. The subcutaneously injected control vaccine outperformed the orally-delivered mutant vaccine candidates. Two vaccines (329 and 318) do reduce presence of JD gross and microscopic lesions, slow progression of disease, and one vaccine (329) reduced fecal shedding and tissue colonization.

OBJECTIVE: Mast cells are associated in angiogenesis in various human and animal neoplasms. However, association of mast cells with tumor microvessel density in canine lymphoma was not previously documented. The objective of the study is to determine if mast cells are increased in canine nodal lymphomas and to evaluate their correlation with tumor microvessel density and grading of lymphomas. METHODS: Nodal lymphomas from 33 dogs were studied and compared with nonneoplastic lymph nodes from 6 dogs as control. Mast cell count was made on Toluidine blue stained sections. Immunohistochemistry using antibody against Factor VIII was employed to visualize and determine microvessel density. RESULTS: The mast cell count in lymphoma (2.95 ± 2.4) was significantly higher (p < 0.05) than that in the control (0.83 ± 0.3) and was positively correlated with tumor microvessel density (r = 0.44, p = 0.009). Significant difference was not observed in mast cell count and tumor microvessel density among different gradings of lymphomas. CONCLUSIONS: Mast cells are associated with tumor microvessel density in canine nodal lymphoma with no significant difference among gradings of lymphomas. Mast cells may play an important role in development of canine nodal lymphomas. Further detailed investigation on the role of mast cells as important part of tumor microenvironment in canine nodal lymphomas is recommended.

The prevalence of Bovine viral diarrhea virus (BVDV) in free-ranging white-tailed deer (WTD, Odocoileus virginianus) in the state of Georgia was evaluated using ear notches collected from hunter-harvested deer during the hunting season of 2010-2011. From September to December 2010, 367 ear samples from WTD were collected from 37 counties in Georgia. The samples were from 178 (48.5%) female deer, 187 (51%) male deer, and 2 (0.5%) of unknown sex. The age of the animals varied from 6 months to 6.5 years. The age was not recorded in 34 animals (9.3%). Of the animals with known ages, 42% were under 2 years. Screening of 367 samples for BVDV using an antigen-capture enzyme-linked immunosorbent assay (AgELISA) resulted in 364 negative samples and 3 suspect samples. The 3 suspect samples were negative for BVDV reverse transcription polymerase chain reaction (RT-PCR), virus isolation, and immunohistochemistry. A subpopulation of samples (n = 89) selected from various geographical regions also tested negative for BVDV RT-PCR. In conclusion, although a few of the samples were suspect for the presence of BVDV by AgELISA, the presence of the virus within the deer population studied could not be confirmed further.

Four species (Dendrobates auratus, Phyllobates terribilis, Pyxicephalus adspersus, and Rhacophorus dennysi) of captive anurans with a clinical history of lethargy and inappetence were found dead and were submitted for necropsy. Gross lesions included irregular patches of sloughed skin and rare dermal ulcerations. Histologic findings included epidermal proliferation that was most pronounced on the digits and that included intracytoplasmic chytrid organisms. Bacteria were often associated with the epidermal lesions. Intracytoplasmic inclusion bodies were observed in hepatocytes. Real-time polymerase chain reaction yielded positive results for both Ranavirus and Batrachochytrium dendrobatidis (Bd). Bacterial culture of internal organs yielded Aeromonas hydrophila. This is the first report of concurrent infections in anurans by Ranavirus and Bd and A. hydrophila.

RESUMO

A Mycobacterium avium subspecies paratuberculosis (MAP) vaccine that reduced the incidence of clinical disease or reduced fecal shedding of MAP would aid control of Johne's disease (JD). The objective of the present study was to evaluate the efficacy of four MAP vaccine combinations, including cell-wall competent (CWC) alum adjuvant, CWC-QS21 adjuvant, cell-wall deficient (CWD) alum adjuvant and CWD-QS21 adjuvant vaccines. Eighty baby goats were vaccinated at 1 and 4 weeks of age with one of these vaccines or a sham control vaccine consisting of alum adjuvant. Kids were challenged orally with approximately 6.0x10(9) organisms in four divided doses of 1.5x10(9) organisms using a goat isolate of MAP. Vaccinated challenged and challenged control groups had 10 and 6 kids per group, respectively. Half of the kids within each group were necropsied at either 6 or 9 months post-challenge. Gross and microscopic lesions and relative number of acid-fast bacilli were evaluated and scored at necropsy. Results indicated all challenged kids had some lesions compatible with JD suggesting none of the vaccines prevented infection. Three vaccines (CWC-alum, CWC-QS21 and CWD-QS21) reduced lesion scores by 46-51% at 9 months. CWD-alum vaccine resulted in a more severe (+33.5%) lesion score than sham-vaccinated challenged control. Lesion scores were greater at 9 months than at 6 months post-challenge in the sham-vaccinated challenged group and CWD-alum vaccinated group, while lesion scores were generally stable with remaining vaccines. Mean fecal CFU/g were significantly different across time from challenge to 9-month necropsy (p=0.043) and the CWC-QS21 vaccine group had a marked reduction in fecal CFU/g at all time points post-challenge. A reduction in MAP CFU/g was also detected in necropsy tissues from kids given the CWC-alum, CWC-QS21 and CWD-QS21 vaccines, and increased CFU/g were detected in tissues from kids given the CWD-alum vaccine. Immunological tests evaluated included, humoral response evaluation by AGID, ELISA and Western blot, and cell mediated response by comparative PPD skin testing (M. avium, Old Johnin, M. bovis and Lot 2 Johnin PPD's), and production of MAP induced gamma-interferon. Vaccination also resulted in false-positive PPD skin test reactions for M. avium PPD, Old Johnin PPD and gamma-interferon tests. When a 2-mm cutoff above normal skin thickness was used to define positive skin test reactions, false-positive reactions for M. bovis were detected in only 2 of 32 kids given a vaccine with QS21 adjuvant.

Although Cryptosporidium spp. are found throughout the world and in multiple environmental conditions, few data are available that explore the possibility of an association between specific environmental parameters and the species or strain of Cryptosporidium. This study examines the potential association between a particular Cryptosporidium species/strain found in calves and soil provinces in Georgia, USA. Necropsy cases spanning the years 1996-2002 were tested. No significant differences (P=0.962, chi(2) test of homogeneity) between numbers of positive cases were noted among soil provinces. Phylogenetic analysis of the sequences for the PCR products revealed sequence similarity of the products with Cryptosporidium parvum strain C1. Although, clinical Cryptosporidiosis in calves was not found to be affected by soil province and may be caused by a single genotype, other genotypes may be responsible for subclinical infection and warrant further investigation.
