OBJECTIVE: The study aims to propose an accurate labelling method of interscapular BAT (iBAT) in rats using dynamic MR fat fraction (FF) images with noradrenaline (NE) stimulation and then develop an automatic iBAT segmentation method using a U-Net model. MATERIALS AND METHODS: Thirty-four rats fed different diets or housed at different temperatures underwent successive MR scans before and after NE injection. The iBAT were labelled automatically by identifying the regions with obvious FF change in response to the NE stimulation. Further, these FF images along with the recognized iBAT mask images were used to develop a deep learning network to accomplish the robust segmentation of iBAT in various rat models, even without NE stimulation. The trained model was then validated in rats fed with high-fat diet (HFD) in comparison with normal diet (ND). RESULT: A total of 6510 FF images were collected using a clinical 3.0 T MR scanner. The dice similarity coefficient (DSC) between the automatic and manual labelled results was 0.895 ± 0.022. For the network training, the DSC, precision rate, and recall rate were found to be 0.897 ± 0.061, 0.901 ± 0.068 and 0.899 ± 0.086, respectively. The volumes and FF values of iBAT in HFD rats were higher than ND rats, while the FF decrease was larger in ND rats after NE injection. CONCLUSION: An automatic iBAT segmentation method for rats was successfully developed using the dynamic labelled FF images of activated BAT and deep learning network.

OBJECTIVE: To achieve efficient segmentation of human supraclavicular adipose tissue (sclavAT) using high-resolution T2-weighted magnetic resonance images. METHODS: High-resolution 1.0 mm isotropic 3D T2-weighted images covering human supraclavicular area were acquired in transverse or coronary plane from 29 volunteers using a 3.0 T MRI scanner. There were typically 144/288 slices for the transverse/coronary scans for each subject, which amounts to a total of 6816 images in 29 volunteers. A U-NET network was trained to segment the supraclavicular adipose tissue (sclavAT). The performance of the automatic segmentation method was evaluated by comparing the output results with the manual labels using the quantitative indices of dice similarity coefficient (DSC), precision rate (PR), and recall rate (RR). The auto-segmented images were used to calculate the sclavAT volumes and registered to the MR fat fraction (FF) images to quantify the fat component of the sclavAT area. The relationship between body mass index (BMI), the volume and FF of sclavAT area was evaluated for all subjects. RESULTS: The DSC, PR and RR of the automatic sclavAT segmentation method on the testing datasets were 0.920 ± 0.048, 0.915 ± 0.070 and 0.930 ± 0.058. The volume and the mean FF of sclavAT were both found to be strongly correlated to BMI, with the correlation coefficient of 0.703 and 0.625 (p < 0.05), respectively. The averaged computation time of the automatic segmentation method was approximately 0.06 s per slice, compared to more than 5 min for manual labeling. CONCLUSION: The present study demonstrates that the proposed automatic segmentation method using U-Net network is able to identify human sclavAT efficiently and accurately.

OBJECTIVE: To investigate the effects of calcium-sensitive receptors (CaSR) on the expression of 11ß-hydroxysteroid dehydrogenase type 2 (11ß-HSD2) and cortisol concentration in a neonatal mouse model of persistent pulmonary hypertension (PPH). METHODS: Fifty-six newborn C57BL/6 mice were randomly divided into a control group (n=14), a PPH group (n=14), an agonist group (n=14), and an inhibitor group (n=14). The mice in the PPH, agonist, and inhibitor groups were exposed to a 12% oxygen concentration, and the agonist group and inhibitor group were given CaSR agonist (GdCl3, 16 mg/kg) and CaSR antagonist (NPS2390, 1 mg/kg) intraperitoneally once a day, respectively. The mice in control group were exposed to air, and then injected with an equal volume of normal saline as those in the PPH group every day. All mice were treated for 14 days. Morphological examination of heart and lung tissues was performed using hematoxylin-eosin staining. The expression levels of 11ß-HSD2 mRNA and 11ß-HSD2 protein in lung tissues were measured by qRT-PCR and Western blot respectively. Brain natriuretic peptide (BNP) and cortisol levels in lung tissues were determined using ELISA. RESULTS: Compared with the control group, the PPH group had significantly increased pulmonary artery wall thickness (WT%), ratio of right to left ventricular thickness (RV/LV), alveolar mean linear intercept, and BNP concentration and a significantly reduced radial alveolar count (P<0.05); compared with the PPH group, the agonist group showed significant increases in WT% and BNP concentration, while the inhibitor group showed significant reductions in the two indicators (P<0.05). Compared with the control group, the PPH group showed significant reductions in the expression levels of 11ß-HSD2 mRNA and 11ß-HSD2 protein, but a significant increase in cortisol concentration (P<0.05); compared with the PPH group, the agonist group had significantly lower expression levels of 11ß-HSD2 mRNA and 11ß-HSD2 protein, but a significant higher cortisol concentration, while the inhibitor group showed opposite changes in these indicators (P<0.05). CONCLUSIONS: CaSR may control the development and progression of PPH in newborn mice by regulating the expression of 11ß-HSD2 and cortisol concentration.

OBJECTIVE: To study the effect of calcium-sensitive receptors (CaSR) on the expression of endothelial nitric oxide synthase (eNOS) and the concentration of nitric oxide (NO) in a neonatal mouse model of persistent pulmonary hypertension (PPH). METHODS: Eighty neonatal C57BL/6 mice were randomly divided into control, PPH, agonist and antagonist groups. The control group was exposed to air, and the other three groups were exposed to 12% oxygen. The agonist and antagonist groups were intraperitoneally injected with a CaSR agonist (GdCl3 16 mg/kg) and a CaSR antagonist (NPS2390, 1 mg/kg), respectively, while the PPH and control groups were intraperitoneally injected with normal saline instead. All mice were treated for 14 days. Alveolar development and pulmonary vessels were assessed by hematoxylin-eosin staining. The protein and mRNA expression of eNOS and its localization in lung tissues were determined by Western blot, qRT-PCR and immunohistochemistry. The levels of brain natriuretic peptide (BNP) and NO in lung homogenate were determined using ELISA. RESULTS: Compared with the control group, the PPH and agonist groups showed significant increases in alveolar mean linear intercept, the percent wall thickness of pulmonary arterioles, right to left ventricular wall thickness ratio (RV/LV) and BNP concentration, but a significant reduction in radial alveolar count (P<0.05). The antagonist group had significant improvements in all the above indices except RV/LV compared with the PPH and agonist groups (P<0.05). Compared with those in the control group, the protein and mRNA expression of eNOS and NO concentration were significantly increased in the PPH group and increased more significantly in the agonist group, but were significantly reduced in the antagonist group (P<0.05). CONCLUSIONS: CaSR plays an important role in the development of PPH in neonatal mice, possibly by increasing eNOS expression and NO concentration.

Nitrogen-doped and nitrogen and oxygen codoped carbon nanobelts (CNBs) (denoted as N-CNBs and N-O-CNBs, respectively) are respectively obtained by pyrolyzing the self-aligned polypyrrole (PPy) NBs and Se@poly(2-methoxy-5-nitroaniline) core@shell nanowires. Particularly, the uniform size, unique nanostructure, and well-defined edges of the PPy NBs result in the uniform size of the doped CNBs with an extraordinarily high N doping level (≈16 at%), especially the very large concentrations of the redox active pyridinic (9 at%) and pyrrolic N (3.5 at%) species. Furthermore, the precursors in highly self-aligned, dense arrays give rise to a very high packing density for the N-CNBs and N-O-CNBs. These incomparable features provide not only appropriate pathways for the introduction of pseudocapacitance via rapid Faradaic reactions and enhancement of volumetric capacitance but also structural design and synthesis approach to new types of nanostructured carbon. Notably, the N-CNBs obtained at the pyrolysis temperature of 800 °C (N-CNB8) in symmetric electrochemical cells deliver a specific capacitance of 458 F g-1 and ultrahigh volumetric capacitance of 645 F cm-3 in aqueous solution, which are among the best performance ever reported for carbon-based supercapacitive materials.

By using trenching method and infrared gas analyzer, this paper studied the seasonal variation of soil respiration (SR), including root respiration (RR) and heterotrophic respiration (HR), in tropical seasonal rain forest (RF) and rubber (Hevea brasiliensis) plantation (RP) in Xishuangbanna of Yunnan, China. The results showed that the SR and HR rates were significantly higher in RF than in RP (P < 0.01), while the RR rate had less difference between the two forests. Soil temperature and moisture were the key factors affecting the SR, RR and HR. The SR and HR rates in the two forests were rainy season > dry-hot season > foggy season, but the RR rate was rainy season > foggy season > dry-hot season in RF, and foggy season > rainy season > dry-hot season in RP. The contribution of RR to SR in RF (29%) was much lower than that in RP (42%, P < 0.01), while the contribution of HR to SR was 71% in RF and 58% in RP. When the soil temperature at 5 cm depth varied from 12 degrees C to 32 degrees C, the Q10 values for SR, HR, and RR rates were higher in RF than in RP. HR had the highest Q10 value, while RR had the lowest one.
