RESUMO

We investigated the differences between the serum proteomic spectral characteristics of acute myeloid leukemia (AML) patients and those of healthy people. We collected peripheral blood serum samples from 62 AML patients and 15 healthy controls. After removing high-abundance proteins, low-abundance serum proteins were separated using two-dimensional gel electrophoresis to identify differences between AML patients and healthy people. We investigated the different protein dots by mass fingerprint analysis, and evaluated the results using the Masort retrieval program provided by the MSDB protein bank. To further investigate the relationship between standard chemotherapy treatment efficacy and differences in protein patterns, we divided 21 patients into two groups (A and B) according to the efficacy of standard chemotherapy. Compared with the healthy cases, the AML patients demonstrated significant abnormal expression in 14 proteins (P < 0.05); α1-trypsin inhibitor (P < 0.01), prealbumin (P < 0.01), apolipoprotein E (P < 0.010), and apolipoprotein A-IV (P < 0.01) expression decreased, whereas haptoglobin HP2 (P < 0.05), serum exogenous lectin (P < 0.05), H factor homologue protein (P < 0.05), and serum amyloid A1 (P < 0.01) expression increased. Further stratified analysis revealed that patients with high serum lectin expression had poor outcomes. The study revealed various proteins with differential expression levels in the peripheral blood of AML patients, and the difference in serum lectin expression is related to the efficiency of standard chemotherapy. Therefore, these proteins are potential diagnosis markers or prognostic indicators of AML.

Adult respiratory distress syndrome (ARDS) is a special type of acute respiratory failure. 10 cases with ARDS were reported. Severe trauma, major abdominal operation and shock were the most common causative factors (9 of ten). All of six conscious patients felt chest dullness, shortness of breath, respiratory distress and accelerated respiratory rate. Rales in lungs were heard in 8 cases. Monitoring results: PaO2(FiO2 = 1) < 40kPa, P(A-a)O2 > 40kPa, PaO2/FiO2 < 300 in all cases; Qs/Qt > 30% in seven. In 5 of seven patients, chest X-ray showed abnormal shadows in the lung fields. PCWP 1.47-1.87kPa. 8 patients, whose failure organs were more than four, died, mortality rate was 80%. Clinical course of this group showed that ARDS and MOSF were two different kinds of syndromes, though they based on similar pathophysiologic mechanism. The insertion of a Swan-Ganz catheter and hemodynamic monitoring are helpful to the diagnosis of ARDS and the selection of the optimal PEEP.

RESUMO

The trimethylsilylation of testosterone and epitestosterone was discussed in detail in this report. Both derivative conditions under which testosterone and epi-testosterone were prepared into TMS-derivatives in the presence of mercaptoethanol as an antioxidizing agent and method for the analysis of the ratio of testosterone to epi-testosterone in urine, based on GC-MS, had been established. The conditions of detection were: carrier gas was helium, derivatives were separated with SE-54 fused silica capillary column, using temperature program and detected by using multiple ion detection mode in which the ion of m/z 432 was the monitoring ion. The method is rapid, sensitive and specific. For the ratio of testosterone to epi-testosterone (testosterone: 20 ng/microliters), there is a linearity between ratio 1:1 and 10:1 (r = 0.998), the limit of detection for testosterone and epi-testosterone is 1 ng, and the minimum concentration of detection in urine is 8 ng/ml.

A rapid, accurate and sensitive atomic absorption spectrophotometry was established for the determination of zinc in plasma after oral administration of licorzin to healthy volunteers. The plasma sample diluted with de-ionized water (1:1) was determined directly. A 5% glycerol solution was used as the solvent of zinc standard solution. The recovery of the proposed method was 97.3 +/- 4.3%. The precisions (CV%) of within-day and day-to-day were less than 5%. The pharmacokinetics of zinc in healthy volunteers after oral administration of 2.5 g licorzin was studied. The results showed that an one-compartment model (Ka = K) was found in 10 healthy volunteers. The pharmacokinetic parameters were as follows: Ka = K = 0.380h-1, Tmax = 2.7h, V = 33.03 L, T1/2 = 1.9h.
