RESUMO

The brown planthopper (BPH), Nilaparvata lugens, is an important pest that affects rice (Oryza sativa) production in Asia. The flavone tricin (5,7,4'-trihydroxy-3',5'-dimethoxy flavone) is a valuable secondary metabolite commonly found in rice plants that can defend rice plants against infestation by BPH. BPH damage can reduce the metabolic level of tricin in rice. Our preliminary transcriptome research results showed that BPH salivary protein 7 (NlSP7), is highly responsive to tricin stimuli. However, the function of NlSP7 in mediating the interaction between the rice plant and the BPH is unknown. In this study, we cloned the NlSP7 gene in N. lugens and found that its mRNA level was greater in the presence of high tricin content than low tricin content, regardless of whether the BPHs were fed a rice plant diet or an artificial diet containing 100 mg/L tricin. Knocking down NlSP7 resulted in BPH individuals spending more time in the non-penetration and pathway phase, and less time feeding on the phloem of rice plants. These changes decreased BPH food intake, feeding behavior, and fitness, as well as the tricin content of the rice plants. These findings demonstrate that the salivary protein 7 of BPH functions as an effector for tricin metabolism in rice.

Euplatypus parallelus (F.) (Coleoptera: Curculionidea) is the most destructive cosmopolitan insect pest of the Platypodinae. Pheromone-based luring agents are used currently in controlling bark beetle. Antennae are the primary insect organs sensing volatiles of host trees and pheromones of pioneer males. We studied the external morphology of antennae and the type, distribution, and the number of the beetle sensilla. Our results show E. parallelus have a geniculate antenna composed of 6 segments, namely the scape, 4-segmented funicle and club. Ninety-seven percent of the antennal sensors were distributed in the club, and 3% were distributed in the scape and funicle. 6 types of sensilla on the antennae were found, including sensilla trichodea (subtypes: STI, STII and STIII), sensilla basiconica (subtypes: SBI, SBII, SBIII and SBIV), sensilla chaetica (subtypes: SChI, SChII and SChIII), as well as sensilla coeloconica, sensilla campaniform and sensilla furcatea. There was no significant difference in the type, distribution and number of sensilla in males and females. No significant difference in the shape and distribution of antennae was found between sexes, but the length of antennae and the number of SChI, SChII, STI, SBI, SBIII and SBIV were significantly larger in females than males. We revealed the external cuticular structure of the antennae in E. parallelus, which can be used to guide future electrophysiological investigations to understand the ability of this beetle to detect semiochemicals.

OBJECTIVE: To investigate the prevalence of syphilis infection in voluntary blood donors in Foshan City, so as to provide reference for improving the infusion blood safety strategy of city. METHODS: A total of 657 382 voluntary blood donors in Foshan City from January 2008 to December 2017 were selected. The blood samples were tested for treponema pallidum. The prevalence trends, the detection rate of treponema pallidum infection and epidemiological data were statistically analyzed. Enzyme-linked immunosorbent assay (ELISA) was performed for antibody screening in blood samples. The two-hole retest was then performed for the samples which were positive in initial test, meanwhile, they were also confirmed by the treponema hemagglutination blood coagulation test (TPHA). RESULTS: First, the annual infection rate of treponema pallidum in Foshan City from January 2008 to December 2017 was 0.43%, 0.40%, 0.41%, 0.43%, 0.50%, 0.32%, 0.36%, 0.32%, 0.37% and 0.32%, respectively. A total of 2502 cases of treponema pallidum infection were detected, the overall infection rate was 0.38%, and the infection rate decreased year by year (x2=23.826, P<0.05). Second, among the 657 382 samples of voluntary blood donors, 2 502 were showed the positive treponema pallidum antibody (0.38%). In 2015-2017, 791 donors were showed positive treponema pallidum antibody, 676 case out of which were confirmed positive, 0.34% with the positive coincidence rate of 85.46%. Third, this group of 676 syphilis infection cases were male-dominated (0.35%, 577/164 484). The age distribution was 18-53 years old, and the average age was 32 years, among which 35-44 years old accounted for 0.33% (138/41 858). The education level of in donors with treponema pallidom positive mainly ranked at low or lower levels under janior college (0.32%, 571/180 917). The occupation is mainly outside work and company staff (0.39%, 452/115 101). The marital status is mainly unmarried (0.29%, 377/130 905). The place of origin is mainly non-local (0.29%,570/198 063). CONCLUSION: The positive rate of treponema pallidum shows a downward trend in 2008-2017. Among the individuals with comfirmed TP antibody positive in 2015-2017, the non-local, unmarried and low educated males are main population, moreover more than half of them are migrant worker or company employeers.

Cyrtorhinus lividipennis Reuter (Hemiptera: Miridae: Orthotylinae), including nymphs and adults, are one of the dominant predators and have a significant role in the biological control of leafhoppers and planthoppers in irrigated rice. In this study, we investigated the antennal morphology, structure and sensilla distribution of C. lividipennis in different instars using scanning electron microscopy. The antennae of both five different nymphal stages and adults were filiform in shape, which consisted of the scape, pedicel and flagellum with two flagellomeres. There were significant differences found in the types of antennal sensilla between nymphs and adults. The multiporous placodea sensilla (MPLA), basiconica sensilla II (BAS II), and sensory pits (SP) only occurred on the antennae of adult C. lividipennis of both sexes. Moreover, there was chaetica sensilla III (CHA III) only observed in males. Sixteen types of antennal sensilla were recorded altogether. They were microtrichia sensilla (MIC), three types of trichoidea sensilla (TRI I-III), three types of chaetica sensilla (CHA I-III), three types of basiconica sensilla (BAS I-III), two types of coeloconica sensilla (COE I and COE II), placodea sensilla (PLA), campaniform sensilla (CAM), MPLA, and SP. In the five different nymphal stages of C. lividipennis, the length of their antennae was significantly increased with the increase of the instar, as well as the number of the TRI II and TRI III. Moreover, sexual dimorphism usually occurred not only in the distribution (CHA III and SP) and the number of antennal sensilla (MIC, BAS II, TRI II, TRI III and MPLA), but also in the length of flagellum (F1 and F2). The possible functions of antennal sensilla are discussed. Those observations could contribute to a better understanding of the development of the olfactory system, and facilitate future studies on the antennal functions in C. lividipennis.

Trichlorfon (TF), an organophosphorus insecticide, has been widely used in seawater aquaculture; it is easily degraded to the highly toxic insecticide, dichlorvos (DDVP). In this study, the enantioseparation of TF enantiomers, as well as their degradation behavior and product (DDVP) formation in mariculture pond sediments, was investigated. The results show that both TF enantiomers degrade into DDVP, which is the main degradation product. Furthermore, S-(+)-TF is preferentially degraded under natural conditions, suggesting that TF enantiomers degrade enantioselectively. Nevertheless, the degradation behavior of TF enantiomers is not enantiospecific under sterile conditions. The formation of DDVP and the enantiospecific degradation of TF enantiomers are attributed to the activities of microbes present in the sediments.

In this study the analysis and confirmation of flumequine enantiomers in rat plasma by ultra-fast liquid chromatography coupled with electron spray ionization mass spectrometry (using propranolol as an internal standard [IS]) was developed and validated. Plasma samples were prepared by liquid-liquid extraction using methyl tert-butyl ether as the extraction solvent. Direct resolution of the R- and S-isomers was performed on a CHIRALCEL OJ-RH column (4.6 × 150 mm, 5 µm) using acetonitrile / 0.1% formic acid / 1 mM ammonium acetate as the mobile phase. Detection was operated by electron spray ionization in the selected ion monitoring and positive ion mode. The target ions at m/z 262.1 and m/z 260.1 were selected for the quantification of the enantiomers and IS, respectively. The linear range was 0.5-500 ng/mL. The precisions (coefficient of variation, CV%) and recoveries were 1.43-8.68 and 94.24-106.76%, respectively. The lowest quantitation limit for both enantiomers is 0.5 ng/mL, which is sensitive enough to be applied to sample analysis in other related studies.

In this work, flumequine (FLU) enantiomers were separated using a Chiralpak OD-H column, with n-hexane-ethanol (20:80, v/v) as the mobile phase at a flow rate of 0.6 mL/min. Solid phase extraction (SPE) was used for cleanup and enrichment. The limit of detection, limit of quantitation, linearity, precision, and intra/interday variation of the chiral high-performance liquid chromatography (HPLC) method were determined. The developed method was then applied to investigate the degradation behavior of FLU enantiomers in mariculture pond water samples. The results showed that the degradation of FLU enantiomers under natural, sterile, or dark conditions was not enantioselective. Chirality 28:649-655, 2016. © 2016 Wiley Periodicals, Inc.

A new method for the isolation and enrichment of ofloxacin enantiomers from fish samples was developed using magnetic molecularly imprinted polymers (MMIPs). These polymers can be easily collected and rapidly separated using an external magnetic field, and also exhibit a high specific recognition for ofloxacin enantiomers. The preparation of amino-functionalized MMIPs was carried out via suspension polymerization and a ring-opening reaction using rac-ofloxacin as a template, ethylenediamine as an active group, glycidyl methacrylate and methyl methacrylate as functional monomers, divinylbenzene as a cross-linker, and Fe3O4 nanoparticles as magnetic cores. The characteristics of the MMIPs were assessed using transmission electron microscopy (TEM), X-ray powder diffraction (XRD), Fourier-transform infrared spectroscopy (FT-IR), and vibrating sample magnetometer (VSM) measurements. Furthermore, the adsorption properties were determined using Langmuir and Freundlich isotherm models. The conditions for use of these MMIPs as magnetic solid-phase extraction (MSPE) sorbents, including pH, adsorption time, desorption time, and eluent, were investigated in detail. An extraction method using MMIPs coupled with high performance liquid chromatography (HPLC) was developed for the determination of ofloxacin enantiomers in fish samples. The limits of quantitation (LOQ) for the developed method were 0.059 and 0.067 µg∙mL(-1) for levofloxacin and dextrofloxacin, respectively. The recovery of ofloxacin enantiomers ranged from 79.2% ± 5.6% to 84.4% ± 4.6% and ofloxacin enantiomers had good linear relationships within the concentration range of 0.25-5.0 µg∙mL(-1) (R² > 0.999). The obtained results demonstrate that MSPE-HPLC is a promising approach for preconcentration, purification, and simultaneous separation of ofloxacin enantiomers in biomatrix samples.

(S)-(-)-Ofloxacin and (R)-(+)-ofloxacin concentrations in the plasma of Pagrosomus major after drug treatment were detected by chiral high-performance liquid chromatography, and various pharmacokinetic parameters were calculated from these data. The elimination half-life of (S)-(-)-ofloxacin was significantly shorter than that of the (R)-(+) enantiomer. (S)-(-)-Ofloxacin also had a significantly lower maximum plasma concentration, area under the concentration-time curve from zero to infinity, and mean residence time than (R)-(+)-ofloxacin. However, the apparent volume of distribution and total body clearance of (S)-(-)-ofloxacin were greater than those of (R)-(+)-ofloxacin. The ratio of the (S)-(-)- to (R)-(+)-ofloxacin plasma concentration was always <1.0. Together, these data suggest that (S)-(-)-ofloxacin was preferentially excreted and (R)-(+)-ofloxacin was preferentially absorbed. Although the difference in pharmacokinetic parameters was small, the metabolic behavior of the ofloxacin enantiomers in P. major was enantioselective.

To develop a more sensitive immunoassay for malachite green (MG) and leucomalachite green (LMG), we identified the immunocomplex binding phage-borne peptides for use in the noncompetitive phage anti-immunocomplex assay (PHAIA). An anti-LMG monoclonal antibody (mAb) was used to select immunocomplex binding peptides from a circular random eight-amino-acid phage-displayed library. After three rounds of panning-elution, five peptides that bound the LMG-mAb immunocomplex were obtained. One of the phage-borne peptide clones that resulted in an assay with the highest sensitivity was chosen for further research. The concentration of LMG producing 50% of the saturated signal and the limit of detection of the assay were 7.02 and 0.55 ng/mL, respectively, with a linear range of 1.35 to 21.56 ng/mL. The PHAIA based on the same antibody was 16 times more sensitive compared to the competitive immunoassay. PHAIA was used to analyze LMG, MG, and two mixtures of spiked fish samples, with validation by high-performance liquid chromatography (HPLC) with fluorescence detector. Results showed a good correlation (R(2)LMG = 0.9841; R(2)MG = 0.993; R(2)Mixture = 0.9903) between the data of PHAIA and HPLC, thus the assay was an efficient method for monitoring food safety.

A Pichia pastoris (P. pastoris) cell surface display system of Bombyx mori acetylcholinesterase (BmAChE) was constructed and its bioactivity was studied. The modified Bombyx mori acetylcholinesterase gene (bmace) was fused with the anchor protein (AGα1) from Saccharomyces cerevisiae and transformed into P. pastoris strain GS115. The recombinant strain harboring the fusion gene bmace-AGα1 was induced to display BmAChE on the P. pastoris cell surface. Fluorescence microscopy and flow cytometry assays revealed that the BmAChE was successfully displayed on the cell surface of P. pastoris GS115. The enzyme activity of the displayed BmAChE was detected by the Ellman method at 787.7 U/g (wet cell weight). In addition, bioactivity of the displayed BmAChE was verified by inhibition tests conducted with eserine, and with carbamate and organophosphorus pesticides. The displayed BmAChE had an IC50 of 4.17×10(-8) M and was highly sensitive to eserine and five carbamate pesticides, as well as seven organophosphorus pesticides. Results suggest that the displayed BmAChE had good bioactivity.

Encarsia guadeloupae Viggiani (Hymenoptera: Aphelinidae) is a minute, obligate endoparasitoid against the spiraling whitefly Aleurodicus dispersus nymph. The external morphology and distribution of the antennal sensilla of female E. guadeloupae were observed by scanning electron microscopy. Antennae of female E. guadeloupae were geniculate in shape, which consist of scape with a radicula, pedicel, and flagellum. Eight morphological sensilla types were recorded in the females: nonporous sensilla chaetica (CH-NP) and nonporous sensilla trichodea (ST-NP); uniporous sensilla chaetica (CH-UP) and uniporous sensilla trichodea (ST-UP) with a tip pore, basiconic capitate peg sensilla with numerous pores open at the bottom of the grooves; multiporous sensilla placoid (MSP) with the multiporous cuticular structure; uniporous rod-like sensilla (RO-UP) with robust grooved surfaces and the tremendous apical hole; nonporous finger-like sensilla (FI-NP) with abundant pimples at the bulgy, mortar-shaped short stalk. In order to further explore the host location mechanisms and courtship behavior of E. guadeloupae, the possible roles of the antennal sensilla of this species were discussed.

Apanteles cypris is an endoparasitoid of rice pest Cnaphalocrocis medinalis. To better understand its host location mechanism, we examined the external morphology of antennal sensilla of A. cypris via scanning electron microscopy. The antennae of male and female A. cypris were filiform in shape, which consist of scape, pedicel, and flagellum with 18 antennomeres. Eleven types of sensilla were recorded in both sexes, including two types of chaetica sensilla, trichodea sensilla without pore, basiconica sensilla I with a tip pore, basiconica sensilla II with wall pores, basiconica sensilla III with a mastoid at the tip, coeloconica sensilla with apparent fingerlike projections, campaniform sensilla with a sprout tip pore, clavatea sensilla, smell pore, and placodea sensilla (PLA) with the multiporous cuticular structure. We also compared the morphology, number, and distribution of the sensory receptors between the two sexes. The length and width of the antennae on the males were significantly greater than that of females. In addition, males of A. cypris showed a larger number and a higher density of PLA compared to females. The morphology and structure of these sensilla were compared to that found in other braconid wasps, and their possible functions in host location were discussed. These observations could facilitate future studies on the biology of olfaction in A. cypris.

This study was aimed to evaluate the impact of regular donating platelets on serum ferritin (SF) of donors. A total of 93 male blood donors including 24 initial plateletpheresis donors and 69 regular plateletpheresis donors were selected randomly. Their SF level was measured by ELISA. The results showed that the SF level of initial plateletpheresis donors and regular plateletpheresis donors were 91.08 ± 23.38 µg/L and 57.16 ± 35.48 µg/L respectively, and all were in normal levels, but there was significant difference between the 2 groups (p < 0.05). The SF level decreased when the donation frequency increased, there were no significant differences between the groups with different donation frequency. Correlation with lifetime donations of platelets was not found. It is concluded that regular plateletpheresis donors may have lower SF level.

By the methods of geostatistics, this paper studied the spatial pattern of Fruhstorferiola tonkinensis nymphs in mulberry fields of north Guangdong. The results showed that the semivariogram of F. tonkinensis nymphs at all development stages could be described by spherical model, implying an aggregative distribution, and the range of their spatial correlation was 1. 973, 1. 968, 2. 169, 2. 302 and 4. 307 m from 1st to 5th instars. Based on the information obtained from investigation site, the isoclines maps of the instars were created by the geostatistical software Surfer 8. 0 with Kriging interpolation, and the resulting maps gave a clear indication about the spatial patterns of the instars. Unlike other locusts, the younger instars of F. tonkinensis had an aggregative distribution, while the elder ones were dispersive in distribution.
