RESUMO

A significant increase in plasma corticosterone levels and depletion of ascorbic acid content in adrenal glands occurred in rats between 1 and 5 h after a single dose (50% LD50) of fenitrothion, returning to normal levels within 12 h; brain AChE activity decreased to about 59-43% and significant inhibition of the enzyme was observed for up to 24 h after intoxication. Repeated dosing with fenitrothion (2.5% LD50) for 14 days caused insignificant elevation of plasma corticosterone levels, while brain AChE activity was inhibited to about 19% of control values and uptake of 4-14C-corticosterone in tissues was significantly diminished.

RESUMO

Plasma corticosteroids concentrations, in rats intoxicated with chlorfenvinphos (p.o. single dose 6.15 mg/kg), were investigated. A significant increase of corticosterone was observed at 1 and 3 h and aldosterone from 1 to 6 h after treatment. Brain and blood AChE activity was diminished to about 10-30% for up to 24 h, with maximal inhibition in brain at 2 h after treatment. Maximal increase in plasma corticosteroids levels occurred within 1 h, while the brain AChE was only slightly inhibited at that time. Results suggest that changes in plasma corticosteroids are not related to the decrease of AChE activity in brain.

Combined action of Thiuram with Carbaryl, Propoxur, Pirimor, Chlorfenvinphos, Malathion, Lindane, and Carbendazine was investigated in rats. The effects induced by Thiuram containing combinations were stronger in females than in males. The type of response was differentiated in each individual case. In rats receiving Chlorfenvinphos or Carbendazine with Thiuram the effects were on the borderline between more than additive and potentiated toxicity in males, while in females potentiation was observed. In the case of the combination with Propoxur the effects appeared to be less than additive in males and strictly additive in females. Other tested combinations exerted additive action in males, while in females effects ranged from additive to potentiating toxicity. It is evident, that combined action of Thiuram with some pesticides might have resulted in potentiation of toxicity. Hence, this fact ought to be considered in evaluation of Thiuram containing multicomponent pesticidal preparations that are to be introduced into practical use.

RESUMO

Male Wistar rats were acutely and chronically intoxicated with organophosphorus insecticides (OPI)-chlorfenvinphos, dichlorvos and fenitrothion. The effects on brain acetylcholine esterase activity (AChE) and norepinephrine (NE) level were determined. Moreover, the effects of chlorfenvinphos on the rate of disappearance and turnover of brain NE was measured. The whole studies revealed that the different degree of AChe inhibition was always accompanied by marked diminution of brain NE level. The rate of decline and turnover time of NE were affected by chlorfenvinphos.
