RESUMO

Objective @#To study the regulatory effect of coiled-coil domain containing 134 (CCDC134) on the osteogenic differentiation of human dental pulp stem cells (hDPSCs).@*Methods @# HDPSCs were isolated and cultured from dental pulp tissue and transfected with NC-CCDC134, shCCDC134 and CCDC134 lentiviruses. They were divided into the control group, negative control group, CCDC134 downregulation (shCCDC134) group and CCDC134 overexpression (CCDC134) group. Surface markers of hDPSCs (Stro-1, CD105, CD34, CD45) were detected by flow cytometry; colony formation was analyzed by toluidine blue staining; ALP expression was estimated by ALP staining; mineralized nodule formation was evaluated by alizarin red staining; lipid droplet formation was examined by oil red staining; and gene and protein expression of CCDC134, Runt-related transcription factor 2 (RUNX2), osteocalcin (OCN), bone morphogenetic protein-2 (BMP-2), and mothers against decapentaplegic homolog 1 (SMAD1) was detected by qPCR and western blot, respectively. Further, a BMP-2 activator (BMP-2) and inhibitor (Dorsomorphin) were used to down-regulate and up-regulate CCDC134, respectively (shCCDC134, shCCDC134+BMP-2, CCDC134, CCDC134+Dorsomorphin), in hDPSCs. The hDPSC aggregates were subcutaneously transplanted into nude mice for 2 months, and new bone formation was detected by H&E staining. The BMP-2/SMAD1 signaling in each group was detected by qPCR.@*Results@#hDPSCs showed high expression of mesenchymal markers and low expression of hematopoietic markers. Compared with the control group, the expression of CCDC134 was increased in the osteogenic-induced hDPSCs (P < 0.05). Compared with the negative control group, the expression of CCDC134 was decreased in the shCCDC134 group, whereas it was increased in the CCDC134 group (P < 0.05). The mineralized nodules, osteogenic genes and proteins in the shCCDC134 group were decreased (P < 0.05), while they were increased in the CCDC134 group (P < 0.05). The expression of BMP-2/SMAD1 signaling decreased in the shCCDC134 group, while it increased in the CCDC134 group (P < 0.05). Compared to the shCCDC134 group, osteogenic genes and proteins increased in the shCCDC134+BMP-2 group, and subcutaneous new bone formation increased in nude mice (P < 0.05). The indexes of the CCDC134+Dorsomorphin group decreased compared with the CCDC134 group (P < 0.05).@*Conclusion@#CCDC134 promotes the osteogenic differentiation of hDPSCs by regulating the BMP-2/SMAD1 signaling pathway.

OBJECTIVE: This work aimed to compare the effect of retention and preventive caries of the pit and fissure sealant on primary teeth by using glass ionomer cements (GIC) with or without acid etching technique. METHODS: In this clinical trail, 100 children aged 3-4 years old with a split-mouth design were equally divided into two groups (the left second upper and lower mandibular primary molars were present in group A, and the right second upper and lower mandibular primary molars were present in group B; each group had 200 molars). All participants were placed in pit and fissure sealant with Ketac® Molar Easymix and seated with atraumatic restorative treatmen press-finger technique in the two groups. Group A was subjected to acid etching (40 s, 35% H3PO4), while group B were not subjected to acid etching. GIC preservation rate, caries rate, and decayed teeth (dt) index were observed after 12 months. RESULTS: After a 12-month follow-up period, the completely lost, partially losing, and losing rates of GIC in 86 children (172 second primary molars) were 83.14%, 9.88%, 6.98% in group A and 62.79%, 20.35%, and 16.86% in group B, respectively. The differences between the two groups were insignificant (P<0.05). The caries rates of groups A and B were 8.14% and 16.86%, and their dt indices were 0.08±0.31 and 1.17±0.46, respectively, thereby indicating significant difference between the two groups (P<0.05). CONCLUSIONS: Using acid etching technology, the retention rate of the pit and fissure sealant increased, while its losing rate decreased. The retention effect of the GIC was improved, and the dental caries prevention effect was enhanced. Sealant processing was a self-curing procedure that sets without the external energy. Hence, this procedure is suitable for the preventive caries of primary teeth in children.

OBJECTIVE: To evaluate the therapeutic effects of pulpotomy and pulpectomy on deciduous molars with deep caries. METHODS: A total of 124 children (192 molars) with deep caries treated from February 2014 to February 2015 were selected. They each had at least one molar with deep caries. MTA pulpotomy (101 molars) and Vitapex pulpectomy (91 molars) as well as prefabricated metal crown repair were conducted. The patients were followed up for 18 months after surgery, and the therapeutic effects were evaluated through clinical and X-ray examinations. RESULTS: The proportion of molars without lesions was 80.20% in pulpotomy group, which significantly exceeded that of pulpectomy group (72.53%). The pulpotomy group with good clinical manifestations underwent spontaneous pain in four molars during follow-up, and five molars gradually underwent pain and gingival redness and swelling. The pulpectomy group suffered from occlusion discomfort in nine molars and gingival fistula in seven molars during follow-up. The postoperative morbidity of pulpectomy group was significantly higher than that of pulpotomy group (χ2=4.50, P=0.04). The 18-month tooth survival rates of pulpotomy and pulpectomy groups were 90% and 79% respectively, which were significantly different (χ2=4.645, P=0.031). CONCLUSION: The postoperative outcomes of pulpotomy are superior to those of pulpectomy.
