RESUMO

To enhance the bioavailability and antihypertensive effect of the anti-depressant drug citalopram hydrobromide (CTH) we developed a sustained-release transdermal delivery system containing CTH. A transdermal diffusion meter was first used to determine the optimal formulation of the CTH transdermal drug delivery system (TDDS). Then, based on the determined formulation, a sustained-release patch was prepared; its physical characteristics, including quality, stickiness, and appearance, were evaluated, and its pharmacokinetics and irritation to the skin were evaluated by applying it to rabbits and rats. The optimal formulation of the CTH TDDS was 49.2% hydroxypropyl methyl cellulose K100M, 32.8% polyvinylpyrrolidone K30, 16% oleic acid-azone, and 2% polyacrylic acid resin II. The system continuously released an effective dose of CTH for 24 h and significantly enhanced its bioavailability, with a higher area under the curve, good stability, and no skin irritation. The developed CTH TDDS possessed a sustained-release effect and good characteristics and pharmacokinetics; therefore, it has the potential for clinical application as an antidepressant.

Zeolitic imidazole framework (ZIF)-derived iron-nitrogen-carbon (FeNC) materials are expected to be high-efficiency catalysts for oxygen reduction reaction (ORR). However, increasing the density of active sites while avoiding metal accumulation still faces significant challenges. Herein, solvent environment engineering is used to synthesize the FeNC containing dense Fe-Nx moieties by adjusting the solvent during the ZIF precursor synthesis process. Compared with methanol and water/methanol, the aqueous media can provide a more moderate Fe content for the ZIF precursor, which facilitates the construction of high-density Fe-Nx sites and prevent the appearance of iron-based nanoparticles during pyrolysis. Therefore, the FeNC(C) nanocubes synthesized in an aqueous media have the highest single atom Fe loading (0.6 at%) among the prepared samples, which presents excellent oxygen reduction properties and durability under alkaline and acidic conditions. The advantage of FeNC(C) is proven in Zn-air batteries, with outstanding performance and long-term stability.

To achieve high efficiency of water electrolysis to produce hydrogen (H2), developing non-noble metal-based catalysts with considerable performance have been considered as a crucial strategy, which is correlated with both the interphase properties and multi-metal synergistic effects. Herein, as a proof of concept, a delicate NiCo(OH)x-CoyW catalyst with a bush-like heterostructure was realized via gas-template-assisted electrodeposition, followed by an electrochemical etching-growth process, which ensured a high active area and fast gas release kinetics for a superior hydrogen evolution reaction, with an overpotential of 21 and 139 mV at 10 and 500 mA cm-2, respectively. Physical and electrochemical analyses demonstrated that the synergistic effect of the NiCo(OH)x/CoyW heterogeneous interface resulted in favorable electron redistribution and faster electron transfer efficiency. The amorphous NiCo(OH)x strengthened the water dissociation step, and metal phase of CoW provided sufficient sites for moderate H immediate adsorption/H2 desorption. In addition, NiCo(OH)x-CoyW exhibited desirable urea oxidation reaction activity for matching H2 generation with a low voltage of 1.51 V at 50 mA cm-2. More importantly, the synthesis and testing of the NiCo(OH)x-CoyW catalyst in this study were all solar-powered, suggesting a promising environmentally friendly process for practical applications.

Carbon-based, non-noble metal catalysts for the oxygen reduction reaction (ORR) are crucial for the large-scale application of metal-air batteries and fuel cells. Density functional theory calculations were performed to explore the potential of atomically dispersed MN4/C (M = Fe or Mn) as an ORR catalyst in an acidic electrolyte and the ORR mechanism on MN4/C was systematically studied. The results indicated MN4 as the active site of MN4/C and a four-electron OOH transformation pathway as the preferred ORR mechanism on the MN4/C surface. The Gibbs free energy diagram showed that the rate-determining step of the FeN4/C and MnN4/C catalysts is the formation of the second H2O molecule and OOH*, respectively. FeN4/C exhibited higher thermodynamic limiting potential (0.79 V) and, thus, higher ORR activity than MnN4/C (0.52 V) in an acidic environment; its excellent catalytic performance is due to the nice electron structure and adsorption properties of the FeN4 site. Therefore, this work demonstrates that atomically dispersed MN4/C is a promising catalyst for the ORR.

The search for metal-free catalysts for oxygen reduction reactions (ORRs) in energy storage and conversion devices, such as fuel cells and metal-air batteries, is highly desirable but challenging. Here, we have designed and synthesized controllable 3D nitrogen and phosphorous co-doped holey graphene foams (N,P-HGFs) as a high-efficiency ORR catalyst through structural regulation and electronic engineering. The obtained catalyst shows a half-wave potential of 0.865 V in alkaline electrolytes. It is found that Zn-air batteries with the N,P-HGFs-1000 air electrode exhibit excellent discharge performance and durability. Our study suggests that the remarkable ORR performance of N,P co-doped graphene is mainly due to the graphite N-C-P structure, where an enhanced charge density and increased HOMO energy level are confirmed by both experimental results and theoretical density-functional theory calculations.

OBJECTIVE: Attention bias modification (ABM) is a promising treatment for depression, but trial data remain restricted to adults. The present trial examined effects of ABM on adolescent depression. METHOD: A total of 45 adolescents with major depressive disorder (MDD), selected from a school population (n = 2,731) using a 2-stage case-finding procedure, were randomized to an active ABM intervention (n = 23) or placebo ABM training (n = 22). In the active condition, participants completed a neutral ABM over 2 weeks for 8 sessions (320 trials each) to shift attention away from sad words to neutral words. At a 9-week follow-up, they received a positive ABM for 2 weeks with 4 more sessions (480 trials each), shifting attention to positive words. The placebo training used the same tasks but shifted attention toward neutral and sad words equally often. Attentional biases and clinical status determined by semi-structured interviews and questionnaires were obtained before and after each training. Depressive symptoms were reassessed at 8- and 12-month follow-ups. RESULTS: Greater reductions in attentional bias score and clinician-rated depressive symptoms were found for active ABM compared with placebo after initial neutral ABM. More participants no longer met diagnostic criteria for MDD in active ABM than in placebo. Greater reductions in self-reported depressive and anxious symptoms at the 12-month follow-up were also found in active ABM compared with placebo. CONCLUSION: ABM may be a potential treatment tool for mild to moderate adolescent major depression. The identification of biomarkers helps to perform early diagnosis, thus benefits the outcome of patients with systemic lupus erythematosus (SLE), in which delayed treatment has been proposed as an independent adverse prognostic factor. In this study, we assessed the values of expression levels of five type I interferon (IFN)-inducible genes (LY6E, OAS1, OASL, MX1, and ISG15) and total IFN score for the diagnosis of SLE. Quantitative real-time PCR was applied to determine gene expressions at transcription level in peripheral blood from 69 SLE patients, 42 patients with other connective tissue diseases, and 26 normal controls. Expressions of five genes and IFN score, calculated according to the expressions of IFN-inducible genes, were all significantly increased in SLE patients compared to those in normal subjects and disease controls. IFN score was not related to age, gender, and the dose of steroids, but weakly correlated with SLE disease activity index. None of the gene expression was associated with concomitant infection status or elevated antibodies against Epstein-Barr (EB) virus in SLE. Both modified IFN score (calculated by the expression of three major IFN-inducible genes) and LY6E level showed good diagnostic accuracy in discriminating between SLE patients and disease controls as well as normal subjects (area under the receiver operating characteristic curve was 0.812 and 0.815, respectively), with 70-80 % specificity and 70-80 % sensitivity at the cutoff of 2.37 and 3.23. In conclusion, high IFN-inducible gene expression is constitutional for SLE patients. The modified IFN score or the LY6E level alone may serve as good biomarkers for SLE diagnosis.

The present study investigated if the emotional valence of words modulates the subliminal repetition priming effect in the brain, in particular, the occipitotemporal visual cortex, by adopting a rapid presentation event-related fMRI design. A masked repetition priming paradigm was adopted, in which, before the presentation of the target (either positive or negative or neutral in meaning), a masked prime word that was either a repetition or an unrelated word of the target was presented. The subject made a perceptual judgment on the target. The results revealed that the left mid-fusiform gyrus was sensitive to the emotional manipulation of the repetition priming effect and that the priming effect in the region was greater in the positive than in the negative word condition. The priming effect in the fusiform gyrus in neutral words was not significant, which might be a result of suppression caused by the emotional context. No effect of valence or repetition was found in the amygdala.
