RESUMO

This study aimed to investigate the nonclinical safety of lincomycin and spectinomycin hydrochloride (LC-SPH) intramuscular (i.m) doses on target animals (chickens) to provide guidelines for dose level design and side effect monitoring in clinical trials. A total of 80 healthy Arbor Acres plus broiler chicks were completely randomized and blindly divided into four treatment groups (control, one-time dose, three-time dose, and five-time dose) of 20 chicks each (20 chickens per group). At the age of day 15, all chickens (except the control group) were administered LC-SPH intramuscularly (chest muscles) at different doses of 20 mg/kg.bw, 60 mg/kg.bw, and 100 mg/kg.bw respectively for 9 consecutive days recommended by veterinary international cooperation on harmonization (VICH) guidelines. The chickens had ad libitum access to antibiotic-free feed and water. Feeding chickens were observed twice a day throughout the study. The drug safety was evaluated by complete blood count, biochemical parameters, histopathological, clinical signs, body weight gain, and feed conversion ratio (FCR). Hence, considering the minor toxicity of 60 mg/kg, our results reveal that intramuscular injection of at least 20 mg/kg body weight has no effects on growth performance, clinical blood parameters, organ coefficient, and histopathological parameters. Thus, a combination of LC-SPH 20 mg/kg body weight i.m injection investigated safe followed daily administration for nine consecutive days in healthy chickens. It is concluded that the experimental results support the safety of 20 mg/kg body weight in combination for the further clinical research study.

Objective: To provide the proper dose specification in pharmacodynamics and clinical application, the composition and dose of Xiaoyao Powder were clarified in Prescription of peaceful benevolent dispensary. Methods: The primary composition and dose of Xiaoyao Powder were verified according to the ancient medical documents, ancient weights, estimate, the results of modern research and so on, so as to confirm the specification conversion standard.Results: According to the weights and estimate in the Song dynasty, the dose of Xiaoyao Powder can be converted as follows: one Liang at that time is equal to 41.3 grams at present. Conclusion: The cognition of the composition and dose of Xiaoyao Powder was obscure and confused for a long time, which made the study of pharmacodynamics quite different.Only by using the primary composition and dose standard can make the results of researches on its medical effects reliable.

RESUMO

Objective To investigate the pharmacoepidemiologic features of drug-induced hypersensitivity syndrome (DHS) and improve the recognition of its particularity.Methods The clinical data of 28 patients with DHS were retrospectively analyzed and summarized.Results Of 28 DHS patients,the suspected drugs were allopurinol in 17 cases,anti-epileptic drugs in 4 cases,antiuberculotic drugs in 3 cases and non-steroidal anti-inflammatory drugs in 4 cases.The earliest symptoms of DHS were skin rash (89.3 %) and fever(85.7 %),secondly liver function damaged(75.0%),which may be accompanied by mucosa,eye,genital damage (53.6%),eosinophilia (32.1%),renal impairment (17.9%),even multiple organ failure and death.Conclusion DHS should be on the alert when rash,repeated fever and visceral lesion occurred in a patient,without good therapeutic efficacy through ordinary anti-anaphylaxis and anti-infective therapy,as well as with the above medication history.

OBJECTIVE:To provide reference for the application of antibiotics based on susceptibility test.METHODS:The problems appeared in the use of antibiotics based on susceptibility test in clinical pharmacy were analyzed and summarized.RESULTS & CONCLUSION:The antibiotics should be used rationally based on susceptibility test so as to improve the rational use of antibiotics.
